Mutation Screening in Familial Cardiovascular Diseases

Introduction: Hypertrophic cardiomyopathy is a congenital cardiac disease with autosomal dominant pattern of inheritance and incomplete penetrance. With the knowledge of the responsible genes, the ability to detect the underlying genetic change and with the study of functional analysis of defected protein, we might be able to determine whether specific genotypes lead to different phenotypes. Aims of Study: To comprehensively analyze the mechanism of genesis of hypertrophic cardiomyopathy in Czech patients afflicted with this disorder from molecular genetic point of view (MYH7, TNNT2 gene) to functional analysis of the 3D molecular model of defected β-myosin heavy chain protein in silico. Beside these aims of the study, the reduction of production of inflammatory aggregates in the cardiovascular system was studied in patients with type 2 diabetes mellitus. The reason of this study was to look into possibilities of therapeutical effect on selected cardiovascular risks in patients with hypertrophic cardiomyopathy simultaneously suffering from type 2 diabetes mellitus. Both of these groups of patients have substantially increased risk of cardiovascular diseases due to development of premature atherosclerosis. Polymorphism of STR loci on the X chromosome and its usage in forensic genetics.

Personal or kinship identification by STR (short tandem repeat) loci is currently a routinely used method, not only in forensic practise. Autosomal STRs are usually analyzed, Y chromosome also being popular because of his great importance (especially in genealogy) due to his recombination incapability. The X chromosome has been used least frequently in these respect, even though in specific cases, it might be more descriptive than autosomal loci. The purpose of this work is to provide a basic overview of the STR sequences and methods that are commonly used to analyze microsatellites, with emphasis on their specific behavior (if located on the X chromosome) and the subsequent possibilities of using these specifities especially in forensic genetics.
Multifunctional Building in Dobříš

The master´s thesis is focused on the design and preparation of project documentation of the polyfunctional house in Dobříš city. Polyfunctional house is a rectangular shape with dimensions of 16,12 x 26,12 m. The building has one underground and three floors, the last floor is the attic. Horizontal and vertical structural system is BS Klatovy, the roof is gabled with a skylights.
Comparison of algorithm animation tools

The diploma thesis focuses on software tools which enable algorithm animation. In theoretical section of the work are introduced different ways how to present algorithms. Then the field of algorithm animation is described; it's history, development and current state. In the last part of theoretical section are shown possibilities how to use algorithm animation in teaching. Practical section of the thesis focuses on comparison of selected software tools. Selected tools are evaluated based on several criteria. The applications are then compared by multi-criteria decision making methods. Main goal of this thesis is to compare the selected software tools. Partial goals are to introduce advantages of using such applications compared to writing the algorithm in text form.
