Changes in selected postural stability tests in hemiparetic patients after botulinum toxin application
ZEMANOVÁ, Lucie
The subject of this bachelor thesis is to compare changes in selected clinical and posturographic tests of postural stability, before and after application of botulinum toxin A to selected lower limb muscles in patients with spastic hemiparesis. The theoretical part deals with the introduction of general information related to posture, such as clinical tests of postural stability, their significance and use, as well as the issue of falls. The next part concerns spastic paresis, its origin, causes, consequences and also clinical scales that evaluate spasticity. The last theoretical part summarize the treatment options of spastic paresis. The practical part describes in detail the kinesiological analyzes of patients and the results of clinical and posturographic tests. Functional Reach Test, Timed Up and Go and Mini BESTest were selected from the clinical trials. Within the posturography, the Weight Bearing Squat test, Modified CTSIB and Limits of Stability were selected. The overall results of this research were not entirely clear, as the individual tests focus on different aspects of postural stability. Some tests were better after botulinum toxin application and some of them before botulinum toxin application. However, impairment of postural stability was in relation to the hemiparetic half of the body. Research has also shown that vision is a key aspect of postural stability in hemiparetic patients. This research could serve as inspiration in the treatment of postural stability not only in hemiparetics, but also in stability disorders in general.

To má následně vliv na provoz, charakteristiky a celkovou účinnost, zvlášť v případě čerpadel s nízkými specifickými otáčkami, protože stav proudění určuje vznikající diskové ztráty. Rovněž ovlivňuje rozložení tlaku a tím výslednou axiální sílu.
Early reading literacy development of early readers at the beginning of the school attendance
Zemanová, Lenka
Early reading literacy development of early readers at the beginning of the school attendance Abstract The aim of the dissertation is to monitor the development of the initial reading literacy of early readers, i.e., children who have learned to read with the comprehension before beginning of their school attendance. The aim of the research was to find answers to the questions how and when these children learned to read and how their initial reading literacy and motivation to read developed during the first year of primary school. Subsequently, the subject of the research was the verification of the connection between early reading and intellectual giftedness and family background. The theoretical part is devoted to current trends in primary school, the basic principles of primary education are described with emphasis on personal and constructivist conception of teaching, focusing on the topics of pupil assessment and inclusive education, including the education of gifted and exceptionally gifted students. The term reading literacy is defined, the stages of its development are distinguished and questions concerning reading are discussed, including the results of research on early reading in the Czech and foreign contexts. The chapter devoted to giftedness defines the term of giftedness and its...

Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences University of Gothenburg Faculty of Science Department of Chemistry and Molecular Biology Candidate: Veronika Nešverová Supervisor: Vladimír Wsól Title of diploma thesis: Studies of the interaction between human AQP5 and PIP Aquaporin 5 is a membrane canal which maintains water balance mainly in lacrimal and salivary glands. A defect in this protein's trafficking from cytoplasm to cytoplasmic membrane is thought to play a role in development of Sjögren's syndrome - a chronic autoimmune disease. Prolactin-inducible protein is a cytoplasmic protein which was found to be contributing to this disease's pathogenesis. This project was aimed to study the proposed interaction between aquaporin 5 and prolactin-inducible protein. Both recombinant proteins were overexpressed in the yeast Pichia pastoris and purified using low-pressure column chromatography on an ÄKTA purification system. Glycosylation of prolactin-inducible protein was confirmed using specific glycoprotein staining. Two coelution tries were run on a HisTrap column. The fractions eluted from the second try were analysed by mass spectrometry. However results did not confirm any interaction. Further investigation is needed for the complete...

Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences University of Seville, Faculty of Pharmacy Department of Biochemistry and Molecular Biology Candidate: Karolína Veselá Supervisors: Prof. Dr. José Luis Venero Recio, Prof. Ing. Vladimír Wsól, Ph.D Title of diploma thesis: Role of Systemic Inflammation in Parkinson's Disease. Parkinson's disease (PD) is the second most common ageing-related neurodegenerative disorder after Alzheimer's disease and the prevalence in population is increasing. The characteristic movement disorder is caused by selective dopaminergic neurons loss, while the mechanism of this neurodegeneration is not well understood. Increasing evidence points out the key role of vicious cycle of microglial overactivation and oxidative stress, while the questions "where it begins" and "how to stop it" remain without clear answers. This thesis investigates implication of peripheral inflammation as a deteriorating circumstance and possible inductor of brain inflammation and progressive dopaminergic neurodegeneration. We use mice model of Parkinson's disease employing single intraperitoneal injection of toxin N-methyl-4-phenyl-1,2,3,6- tetrahydropyridine (MPTP) that induces specific degeneration of DA neurons and in contrast to other animal...
Oxidative damage to proteins
Bártová, Jana ; Boušová, Iva (advisor) ; Zemanová, Lucie (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Jana Bártová Supervisor: PharmDr. Iva Boušová, Ph.D. Title of diploma thesis: Oxidative damage to proteins Hypochlorous acid is a strong oxidant, which is used by phagocytic cells to kill ingested bacteria. Surrounding structures including proteins may be damaged in case the content of a phagocyte is spilled to the vicinity. Catechins belong to a group of flavonoids whose antioxidant properties were demonstrated in many studies. My diploma thesis was focused on a potential effect of eight naturally occurring derivatives of catechin towards oxidative damage of human serum albumin (HSA) by hypochlorous acid. I used two spectrophotometric (determination of carbonyl groups and chloramines) and three electrophoretic methods (native PAGE, SDS-PAGE, immunoblotting). Dependence of an antioxidant effect on its concentration (0-3 mM) was observed during determination of chloramines and obtained data were used for calculation of EC50 values of individual catechins using program GraphPad Prism. The most effective compound was catechin whose EC50 was 4.8 µM. Results suggest that antioxidant ability decreases with increasing number of hydroxyl groups in the molecule of flavonoid. Carbonyl groups are a...

Javorská, Lenka ; Zemanová, Lucie (advisor) ; Novotná, Eva (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Biochemical Sciences Candidate: Bc. Lenka Javorská Supervisor: RNDr. Lucie Škarydová, Ph.D Title of diploma thesis: Detection of His-tag and FLAG-tag recombinant proteins Recombinant proteins are proteins produced in genetically modified organism by recombinant DNA technology that enable insert DNA sequence of target protein in expression cell. This technology allows preparation of actually any protein in any quantity without difficulties associated with demanding purification of native protein from tissues. Recombinant proteins are produced often in form of fusion proteins with suitable peptide tag (e.g. His-tag, FLAG- tag) that can help with their detection and enable simple purification. The aim of this diploma thesis was to introduce and optimize universal method for the detection of recombinant proteins labelled with His-tag (6x histidine) and FLAG-tag. The model recombinant protein with His-tag was enzyme carbonylreductase 1 (CBR1) produced by expression system Escherichia coli. For FLAG-tag labelled protein were chosen enzymes DHRS12, HSD11β1, RODH4, DHRS3, DHRS7, DHRS8 produced by insect cells (strain Sf9). SDS polyacrylamide electrophoresis for protein separation and immunochemical detection after Western...

Popovská, Lenka ; Solich, Petr (advisor) ; Zemanová, Lucie (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of Analytical Chemistry Candidate: Mgr. Lenka Popovská Supervisor: Prof. RNDr. Petr Solich, CSc. Consultant Supervisor: Mgr. Barbora Šalovská, Ph.D. Title of the rigorous thesis: Optimization of procedure for isolation of phosphorylated peptides from a peptide mixture by metal oxide affinity chromatography for mass spectrometry analysis. Protein phosphorylation is the key regulatory mechanism in cellular signalling. It is involved in most cellular processes that are regulated by a complex interplay of protein kinases and protein phosphatases and include proliferation, differentiation, and apoptosis. Defective or altered signalling pathways can lead to various diseases, and therefore it is very important to study protein phosphorylation in detail and try to understand it as much as possible. Technological development of mass spectrometry (MS) in recent years has enabled a significant increase in the number of phosphorylation sites that may be identified in a sample. Increasing scan speed together with commercial availability of high resolution MS have improved quantity and quality of the acquired data, which can be obtained in a relatively short time. However, even high-end devices would not allow to comprehensively identify the...
