Michaela Šadibolová Title of diploma thesis: Effect of xanthohumol and 6-prenylnaringenin in the in vitro model of non-alcoholic fatty liver disease Non-alcoholic fatty liver disease (NAFLD) is a chronic disease characterized by the accumulation of triglycerides (TAG) in hepatocytes encompassing a wide spectrum of pathological states ranging from simple steatosis to non-alcoholic steatohepatitis (NASH) and eventually progressing to cirrhosis and hepatocellular carcinoma. Xanthohumol (XH), a prenylflavonoid found in common hop (Humulus lupulus), is currently being studied as a potential drug for NAFLD therapy due to its described anti- steatotic, anti-inflammatory, anti-fibrotic, hypolipidemic and antioxidant effects. The aim of this thesis was to study the potential hepatoprotective effect of XH and 6-prenylnaringenin (6PN), a minor prenylflavonoid also found in common hop. For this purpose, a murine in vitro NAFLD model was used in which steatosis was induced by a lipogenic medium containing palmitic and oleic acid, fructose, citrate, and insulin in precision-cut liver slices (PCLS). Barbora Szotáková, Ph.D. Title of diploma thesis: Effect of sesquiterpenes on biotransformation enzymes in tissue slices Sesquiterpenes are secondary metabolites, produced mainly by higher plants. They are structurally diverse compounds with a large spectrum of biological activity. They possess antiphlogistic, antibacterial, antimalarial and antitumour activity. The aim of this diploma thesis was to find out how selected sesquiterpenes - α-humulene (HUM) and β-caryophyllene (BC) influence the activity of chosen biotransformation enzymes - NAD(P)H-chinon oxidoreductase 1 (NQO1), glutathione S-transferase (GST) and sulfotransferase (SULT). Precision-cut tissue slices from the rat liver (Rattus norvegicus, tribe Wistar) were chosen as model system. Thickness of the slices was approximately 200-250 µm and diameter 8 mm. Liver slices were preincubated for 30 minutes and then incubated for 24 hours (in presence of HUM and BC) in a 12-well plate in the atmosphere of pneumoxide (95% O2/5% CO2). Petra Matoušková, Ph.D. Title of Doctoral Thesis: Potential toxicity of terpenes and their effects in liver cells The public interest in herbal products, supplements, extracts or isolated active compounds has been increasing during last decades. The aim of this doctoral thesis was to study selected compounds from the group of monoterpenes and sesquiterpenes and their interactions with liver cells: interactions with drug metabolising enzymes, their potential liver toxicity and looking for novel mechanisms of action. The literature about hepatotoxic properties of several monoterpenes and sesquiterpenes, that showed liver toxicity, was summarized first. Then, the study was aimed toward a known hepatotoxicant (R)-pulegone and its presumed metabolite (R)- menthofuran. The toxicity results in precision-cut human liver slices have shown that (R)- menthofuran was less toxic to human liver and that the reason may be inter-species differences between human and mice.
