Preservative systems for natural cosmetics

The aim of this bachelor thesis is to evaluate the antimicrobial activity of naturally derived preservatives against selected microorganisms. Theoretical part is focused on description of the most used natural, naturally derived and synthetic preservatives, on the safety of preservatives and finally, the issue of contamination of cosmetic products by microorganisms. In experimental part, antimicrobial activity of 4 naturally derived preservatives was evaluated against selected microorganisms, which were bacteria Micrococcus luteus, Kocuria rosea, Bacillus cereus, Bacillus subtilis, Serratia marcescens and one yeast Candida vini. The tested preservatives were from MinaSolve company - E-LEEN Green A, which contains pentylene glycol and phenylpropanol, E-LEEN Green OR, which is composed of citrus extracts, citric acid, ascorbic acid and pentylene glycol, Green B, which contains benzoic acid, sodium benzoate and pentylene glycol and Hexam+, which is a mixture of pentylene glycol and hexamidin diisethionate. Antimicrobial activity was evaluated by agar diffusion method. The highest antimicrobial activity showed Hexam+, the lowest E-LEEN Green OR. E-LEEN Green A and Green B did not show any antimicrobial activity against any microorganism in the 1% tested concentration. The measured values of the inhibition zones show that the most resistant microorganisms from all the tested in this thesis are Bacillus cereus and Bacillus subtilis and the least resistant are Micrococcus luteus and Kocuria rosea.
Preparation of nanoparticles and nanofibers for application in anti-acne products

The diploma thesis was focused on the preparation and characterization of nanoparticles and nanofibres with active substances from chosen herbs with the aim to apply this materials to the products against acne. Various types of extracts were tested on the content of polyphenols, flavonoids and antioxidation activity. These extracts were encapsulated to the liposomes and fibres of polyhydroxybutyrate. Prepared liposomes and fibres were tested mainly on antioxidation activity and antimicrobial activity against the strain Propionibacterium acnes. Then, liposomes were applied to cosmetic emulsions. These creams reported high antioxidation activity and excellent stability determined by analytical centrifugation. Prepared nanofibres also reported high antioxidation activity and antimicrobial effect as well. Finally, particles and fibres were tested in contact with human cells. In appropriate concentration, there was no cytotoxic effect and tested materials can be used in applications on problems with acne.
Study of electrophotocatalytic water disinfenction

This bachelor thesis was focused on the photocatalytic disinfection of water with titanium dioxide applied to the working electrode. The experiment was conducted under normal conditions, with the application of UV-radiation with intensity of 40 W/m2 and with UV radiation and of 1 V electrical voltage at the same time. Each part of the experiment was run for 60 and 120 minutes. The microbicidal effect was monitored on Escherichia coli bacteria.
Infection of salmonelosis, their source and diagnosis.

This bachelor‘s thesis deals with infection of salmonelosis, it’s source, prevention and diagnosis. Theoretical part includes general information about Salmonella species and its morphology. It describes pathogenesis, source of infection, clinical symptoms, treatment and diet recommendations. It analyzes statistics of number of infection occurrences in Czech Republic. It also deals with prevention of infection origin on schools, population foreknowledge and climatic effects. Experimental part of this thesis contains description of optimization procedure of identification Enterobacteriaceae by adjusting of pH of prepared medium MIU.
Primary Architecture / Teaching Architecture Elements

The thesis addresses the connection of the Faculty of Architecture and the Faculty of Fine Arts in the newly designed building. Both faculties are defined primarily by practical creation and corresponding elementary teaching spaces - studios. From these basic elements literally grows the final form of the school. The design process itself is accompanied by constant changes and the search for adequate spatial expression for individual functions of the school. The final solution is the result of gradual rationalisation, finding certainties and sticking points in the proposal. In the end, the barest, most elemental, is crystallized from a multitude of variations and reflections on space.
Jaroslavice – place in the landscape

The work is aimed at the center of the village, which is a square that, in size comparable to that of the Square of Freedom in Brno, is an exhibition of problems: considerable deterioration of the building fund, underground use of the public space, which is moreover unreadable and unreachable. The proposed solution divides the area of the square into three smaller public spaces that have a different character and function. Mariánské námětí is the torso of the existing one, which has been earmarked for the building of a new municipal office and library, is characterized by town houses and the location of services and public amenities. The second area is a semi-detached cottage. It is dominated by walnuts and water. The proposed buildings are oriented towards the library and the wine house. The third such defined area is the Wine Market, which is the crossroads, the scattered area of the surrounding houses, like a school, shops, or wine house. A large part of the COOP building has a sports complex with a halo and a swimming pool. There are solved especially the buildings of the town hall with a library and a wine house.
Characterization of edible films with the potential usage in food packaging

In the last years, the edible packaging has been gaining popularity due to the potential usage as the packaging for food instead of synthetic materials. The edible packaging can positively affect the packaged food by the active properties or due to its intelligent properties can monitor the changes in food. In theory, the legislation about the packaging of food and edible packaging was described. The substances for edible films preparation and additives, which can improve the properties of the edible films, were described too. In this part, the undesirable microorganisms occur in food and the chapters about active and intelligent properties of the edible films were summarized. The main aim of the thesis was the practical preparation of edible packaging/films made from furcellaran, gelatin hydrolysate with the addition of water dried or fresh rosemary extracts in concentrations 5 %, 10 %, and 20 %. In the experimental part, the physical analysis was done – textural parameters such as tensile strength and elongation at break, water content, and thickness. In the chemical analysis the total polyphenols content, determination of polyphenols by HPLC, antioxidant activity, optical properties and antimicrobial activity of the prepared edible films were done. There were found out, that the total polyphenols content and antioxidant activity were the highest in the 20%DR (TPC: 6,79 ± 0,06 mg gallic acid/g; DPPH: 87,84 ± 0,07 %). Also, the 20%DR and 20%FR changed color in different pH (3 and 12). The antimicrobial activity was negative. The thesis showed that the addition of natural extracts to edible films can increase the polyphenols content and they can also work as the UV protector.
Microbial profile of model cheese

This Diploma thesis deals with the identification of microorganisms in samples of experimentally produced cheese. Model samples of cheese were produced in association with a private manufacturer. Raw organic milk was used to make the cheese. The theoretical part provides an overview of the issues such as the characteristics of the cheese, its properties and compounds. A special chapter is dedicated to molecular diagnostic methods, which serve for identification of microorganisms. In the experimental part of the thesis were identified some microorganisms in the samples. A polymerase chain reaction was used. The DNA was isolated from coarse lysates using phenol extraction; it was amplified using specific primers and demonstrated by gel electrophoresis. DNA of lactic acid bacteria (Lactobacillus spp., Lactococcus spp.) and yeast was ascertained in the cheese. The samples do not contain DNA of pathogenic genera Bacillus and Salmonella.
Analysis of Socio-Economic Effects of Transport Infrastructure Construction

Bachelor’s thesis is focused on analysis of socio-economic effects of transport infrastructure construction. The aim of the thesis is to analyse which of the variables of input quantity has the greatest effect on the economic efficiency of the public benefit project in the transport sector. The analysis is performed by using cost-benefit analysis. Analysis is calculated for the decision-making process of choosing a project for possible implementation. The work discusses in detail financial and socio-economic cash flows in the calculation of criteria economic efficiency indicators.
