The role of PKN family kinases in cancer

This bachelor thesis is focused on the PKN family of Ser/Thr kinases. This family includes three isoforms PKN1, PKN2 and PKN3. Especially it deals with the kinase PKN3 in more detail. These are kinases related to protein kinase C, belonging to the AGC superfamily. PKN kinases are activated via small G proteins of the Rho GTPase family or unsaturated fatty acids. PKN kinases are involved in many cellular processes, such as the regulation of cytoskeletal rearrangements, affect cell adhesion, cell movement, embryonic development and the cell cycle. Expression of PKN3 is particularly increased in cancer cells but is only present in small amounts in normal body cells. Therefore, PKN3 appears to be a very interesting therapeutic target for the treatment of cancer. Studies have shown that PKN3 has a significant effect on the motility of cancer cells, thus contributing to their migration and ability to form metastases.
Extraction and Classification of Atrial Activity using Multi-Site Intracardiac Electrograms

The aim of this thesis is to acquaint the reader with the origin of supraventricular, mainly their manifestations in intracardiac electrograms. There are described basic methods of analysis of electrocardiographic records. Practical part contains extraction of atrial activity and classification of atrial rate in MATLAB program. Atrial activity is extracted from preprocessed data. The extraction of atrial activity is followed by the classification of atrial rhythm using the K–means method.
Premature atrial contraction detection in ECG

This thesis discuses detection of premature atrial contraction from ECG. In the first part, thesis describes electrocardiogram, cardiac conduction system and extrasystoles. Extrasystoles are premature contraction caused by ectopic heartbeats. Next part is devoted to signal preprocessing, the feature description methods, reduction of feature vector and methods of classification. Realized method and results of classifier are discused in the last part.
Advanced scoring of sleep data

The master´s thesis is focused on advanced scoring of sleep data, which was performed using deep neural network. Heart rate data and the movement information were used for scoring measured using an Apple Watch smartwatch. After appropriate pre-processing, this data serves as input parameters to the designed networks. The goal of the LSTM network was to classify data into either two groups for sleep and wake or into three groups for wake, Non-REM and REM. The best results were achieved by network doing classification of sleep vs. wake using the accelerometer. The statistical evaluation of this best-designed network reached the values of sensitivity 71,06 %, specificity 57,05 %, accuracy 70,01 % and F1 score 81,42 %.
Návrh pilařského provozu rodiny Jirečků ve Svatoslavi

This thesis describes the design of a new sawmill operation of Jireček families in Svatoslav near Třebíč. This is a proposal for operation of a particular forest property. The main requirement is to design appropriate technological arrangements, machinery and automated equipment that will meet the maximum sawing coniferous 3000 m3 per year range. The paper presents the layout of the premises Still hunting lodge where a sawmill operation to be designed, requirements analysis and calculations required values for the design of the operation, according to which the proposed machine-technological equipment and traffic arrangements. The work described processing streams with a specific variant of the machinery. Of these proposals were selected, one option that has been recommended as the most suitable. The thesis included drawings of floor plans proposed sawmills, lateral cuts recommended variants sawmills and drawings situation with the location of the building sawmills deployment and storage of logs and lumber.

Novotná, P. Influence of ISO 9001 certification on the company performance. Thesis. Brno: Mendel University in Brno, 2018. Thesis deals with the influence evaluation of the quality system implementation on increasing company efficiency. It focuses on the analysis of its current state, management of the quality system and its administration. Aim of the thesis is to demonstrate the impact of ISO 9001 quality certification on the company financial and non-financial performance and identification of areas and indicators in which certification impacts are most expressed. For the solution was used an analysis of internal environment according to the 7S method, company self-assessment according to ISO 9004 and the methodology based on the principles of the EFQM model. Main finding is the underestimated impact of quality system implementation on company performance due to non-compliance with all requirements and principles of interconnected standard. Companies meet requirements of the standard primarily for purpose of meeting conditions for recertification audits and for entering into a business agreement with customers.

The work deals with problems of detection of the atrial activity using intracardiac recordings. In the introductory part, individual supraventricular tachycardias with examples of their manifestations in ECG recordings are described. In the practical part, the designed detector is implemented in Matlab and recordings are classified according to the heart rhytm.
Detection of premature ventricular contractions in ECG

This thesis focusses on the detection methods of extrasystoles from ECG and description of electrocardiogram, cardiac conduction system, extrasystoles and ventricular tachycardia. Extrasystoles are premature ventricular contraction caused by ectopic heartbeats. Classification is based on signal preprocessing, detection of R peak, the heartbeat segmentation, the feature description methods, normalization of features and the learning algorithms used. Selected and realized methods achieved classification accuracy ACC = 98 %, sensitivity SE = 100 % and specificity SP = 96,1 %. Gained features are also used for detection bundle branch block.
Body Surface Potential Mapping In Animal Experiments - Pilot Study

The aim of this article is to present a pilot case study in the area of body surface potential mapping. This technology was used to record data from animal experiments with the goal to find out, what information could be derived from recorded ECGs in the protocol including induced myocardial ischemia. Data recorded before and after the ischemia were compared. Differences in ECG beat morphology were observed, also local changes in analysed intervals (QT, ST, QS, PT) were visible. It was hypothesiced that QT interval will be prolonged after myocardial ischemia. Nevertheless, according to out pilot study this hypothesis was declined.
