Motivation and psychosocial needs of workers of the social and legal protection of children

This diploma thesis deals with social workers who work in the department of social and legal protection of children. The aim of the work was to characterize the motivation factors and psychosocial needs for the performance of the work of OSPOD social workers in Strakonice. My diploma thesis was divided into a theoretical part and a practical part. In the theoretical part of my diploma thesis, I described the basic concepts related to the concept of social-legal protection of children, including historical development, legislative framework, and I described the functioning system of social-legal protection of children in the Czech Republic. Furthermore, in this section I described terms such as the Code of Ethics for social workers, motivation, psychosocial needs, psychohygiene and burnout syndrome. In the practical part, the research strategy of qualitative research was used and the method of questioning and the technique of data collection in the form of a structured interview were chosen. Biotechnological production of PHA by selected thermophilic bacteria

This thesis is mainly focused on the production of PHA copolymers and study of metabolism of the thermophilic genus Aneurinibacillus. Theoretical part mainly deals with general characterization and production of PHA and with PHA producing bacteria, especially the extremophiles. The last chapter talks about the bacterial genus Aneurinibacillus and previous knowledge about this PHA producer. The experimental part is focused on testing the PHA synthase from Aneurinibacillus, which can synthesize large scale of different monomers. The two bacterial isolates, AFN2 and AH30, were used to produce copolymers with unsusual monomers in their structure, such as 3HB, 3HV, 4HB, 4HV, 5HV and even 3HHx and 4HHx. This means that the PHA synthase has low substrate specifity. The production of copolymers was supported by adding precursors which encompass lactones, diols and organic acides. There were used two different types of medium. The mineral medium, which generate the environment with limited content of nitrogen, and complex medium, which provide to bacteria all required elements. It was observed that the genus Aneurinibacillus don´t require the nitrogen limitation to produce PHA. One of the most successful experiments was the cultivation of AFN2 on complex medium with added glucose and -valerolactone as precursor. The concentration of biomass was 4,2 g/l and the syntesized PHA reached 2,6 g/l. This copolymer was composed nearly of 60 mol. % monomer 4HB. In the last section of experimental part the two pair lacton-diol were selected to produce more PHA copolymer wich was later isolated and characterized. When the lactone was used as precursor, the resulting PHA copolymer reported higher molecular weight. This can be explained by alcoholytical activity of PHA synthase from bacterial genus Aneurinibacillus.
Foster Care of Romani Children

This bachelor thesis deals with foster care providers and their motivation to perform foster care. The aim of the work was to find out what are the factors of motivation for foster care of a Roma child. In the theoretical part, I described the basic concepts related to the concept of alternative family care, including the legislative framework. Subsequently, I also described the topic of the Roma community, the Roma family and the formation of the identity of a Roma child in a non-Roma surrogate family. In the practical part, qualitative research was used, more precisely biographical interviews, which I supplemented by subsequent questioning. The communication partners were families who provide foster care, have at least one child of the Roma ethnic group and are themselves of the non-Roma ethnic group. Another condition for the selection of communication partners was that they have a permanent residence in the South Bohemian Region. All obtained data are analyzed in a way based on grounded theory. While processing this bachelor's thesis, I found that motivation can be different and different from each individual separately. The main factors of initial motivation can be external and internal influences, in each case the greatest impact on such decisions has the environment, family, friends or even professional life. Cooperation with the relevant authorities and social workers is especially important for every applicant for alternative family care. If the applicant is sure that his interest is also supported by the authorities, this decision is often full and fulfilled.
Interaction of Borrelia sp. with HL-60 cells and monocytes and cultivation of Anaplasma phagocytophilum in HL-60 cell culture

Borrelia burgdorferi sensu lato and Anaplasma phagocytophilum are causative agents of Lyme disease and human granulocytic anaplasmosis. Their common vector in Europe are the ticks from the genus Ixodes. In our work, we focused on interaction of innate immune cells with the causative agent of Lyme diseases, that are insubstitutable in their function in the early phase of the disease. Anaplasma phagocytophilum is hard to cultivate, the only possibility is to cultivate it in cell cultures. Successful cultivation of Anaplasma phagocytophilum acquired from patients in our geographic area is crucial for following experiments and for diagnostics too. In our experiments, we used validated cell cultures of HL-60 cells, canine monocytes DH82 and murine monocytes P388D1. During our studies of interaction of the causative agent of Lyme diseases with cells, we used two strains of different species Borrelia. Borrelia garinii M192 and Borrelia burgdorferi sensu stricto B31. These strains vary in virulence. The strain M192 is virulent, but the strain B31 lost its virulence by passages. Prague City Hall's attitude to prostitution

The diploma thesis The City Hall of Prague attitude towards prostitution is based on the constructivist paradigm and uses feminist theories. It examines, by means of a critical reading, the attitudes of the City Hall of Prague towards prostitution and its development in course of time. The period of investigation was determined by the beginning of municipal intention to introduce a regulatory bill. For this purpose all the relevant press releases from the period 2003 and 2015 were gathered. These documents are concerned with prostitution and they are accessible on press service section of www.praha.eu portal used by the City Hall as a medium for press releases. A theory part introduces basic concepts and trends in feminist debates concerning prostitution. The core of the thesis consists of four categories derived from the source data.These are: definition of prostitution, prostitution as a problem, needs and goals, and support and repression. The categories are treated critically from the viewpoint of feminist theories. The author follows not only the topics concerning prostitution, but also the way they are presented. Pilsen music scene in 1968-1983

The aim of essay is to broaden knowledge of Pilsen alternative music scene in 1968- 1983. The study is focused on the conditions, under which was the rock subculture in Pilsen formed, by what has been this subculture affected and how to deal with various restrictions by the communist regime in Czechoslovakia. The main basis for this research, are interviews with members of rock band in West Bohemian metropolis in a given period. These interviews were realized using oral history. Beside this, the study is supported also by knowledge of literature, period newspaper and archive materials. The study presents new facts in the areas of retraining examinations, Festival of political songs, whose national phase was held in Sokolov and public performances. Furthermore, authoress presents musician response to article Nová vlna se starým obsahem, published in magazine Tribuna in 1983. The research is focused on musician view on said areas, which could enrich the already know facts and help to fill space in history of pilsen music scene in the past regime. Práce uvádí historické a právní souvislosti, které vedly k současnému problematickému stavu v oblasti zpřístupňování tohoto informačního pramene. Hlavní část diplomové práce tvoří analýza tradičních i elektronických zdrojů judikatury a pokus o zmapování současné nabídky těchto zdrojů. Mezi nimi jde v první řadě o systémy aSPI a LexData, pak také rozhodnutí zpřístupněná na webových prezentacích nejvyšších soudů, např. Nejvyššího soudu Čr. Use of analytical methods and chemometry to verify the geographical authenticity of honey

The teoretical part of this bachelor thesis is focused on the process of producing the honey by bees, its composition and means of its adulteration or ways to detect substandard products. The experimental part describes methodes, which were used to observe the samples. Sacharides, organic acids, mineral content, the amount of dry matter, titration acidity and conductivity were meassured. Obtained data were used for differentiation the Czech and foreign honey. The result of the statistical analysis shows the calcium, copper and glucose content are suitable paramteres to differentiate the Czech honey from that externals. Theese parameters could be used to distinguish the two different geographical origin of honey. Nevertheless, the total evaluation seems to be insufficient for differentiation these two groups. The efficiency could be icreased by the including more samples for analysis, or meassuring more parameters. Finally, some of the parameters were used for quality control of honey.
Optimization of Tax Burden of a Concern

The diploma thesis deals with a proposal of recommendations that will lead to tax reduction in corporate income tax for a selected concern. The theoretical part is focused on the legal framework of corporate taxation and the definition of basic areas in which it is possible to perform tax optimization. Thesis also defines the concern, its forms and methods of organization and describes its connection in the international context. In the practical part, the acquired knowledge is applied to the data about the selected company and an analysis is performed in the given areas of optimization, while recommendations are formulated leading to a reduction of the company’s tax liability.
