Ecotoxicological Assessment of Point Pollution in the Ecosystem

The main subject of this dissertation is the ecotoxicological evaluation of significant point sources of pollution. These include wildfires and wide fires of factories and also industrial and urban wastewater treatment plants. The local application of pyrolytic product obtained from sewage sludge as the point source of pollution was also included in the thesis. Tests in aquatic and contact arrangement with using both water and soil organisms for ecotoxicity evaluation of selected matrices were chosen. Crustaceans Thamnocephalus platyurus, Daphnia magna Straus and sea bacteria Vibrio fischeri were selected as representative organisms of water animals. Duckweed Lemna minor was chosen as water plant. Further collembola Folsomia candida and earthworm Eisenia fetida were chosen as representatives of soil animals. Finally onion Allium cepa L. and mustard Sinapis alba are representatives of terrestrial plants. Submitted thesis deals about analytes of interest, their properties and ways of distribution to the environments. The output of the thesis is a comprehensive assessment of the effects of selected point sources of pollution on aquatic and terrestrial ecosystems organisms and ecotoxicological risks assessment. The fire extinguishing agents have been proven to be the most stressful substances for all selecteded organisms.

The bachelor thesis focuses on evaluation of financial situation of the company TM Stav, spol. s.r.o. and proposals to its improvement for period 2016-2020. This work is divided into three parts – theoretical, analytic and practical. In the theoretical part are implied theoretical basis about financial analysis. Analytic part contains analysis present condition of the company. Practical part is focusing on proposals to improve current situation of the company TM Stav, spol. s.r.o.
Evolutionary and genetic engineering of bacterial producers of polyhydroxyalkanoates

This doctoral thesis deals with the topic of evolutionary and genetic engineering of polyhydroxyalkanoates (PHA) producing bacteria. Apart from these topics, the issue of biotechnological production of PHA on model hydrolysates of lignocellulosic biomass with the use of extremophilic microorganisms is also studied, as well as the development of an alternative method of PHA isolation. The themes were freely linked to previous experiments and reflected the currently solved projects in a working group. Doctoral thesis is prepared in the form of a commented discussion of published works, which are part of it in the form of appendices. Evolutionary engineering was mainly applied to the model PHA producing bacterial strain Cupriavidus necator H16. By adaptation to levulinic acid, isolates producing copolymer P(3HB-co-3HV) with a higher content of the 3HV fraction were obtained, which leads to improved properties of the polymer for further processing. As well as culture growth also the amount of total PHA in the biomass was higher. By long-term adaptation of the same strain to osmotic stress and the presence of copper ions, the isolates which are characterized in the second publication, were obtained. Based on obtained data, it was possible to observe differences in the adaptation process, where the adaptation to osmotic stress was gradual, while a significant step in the increase of biomass and PHA signaling faster adaptation was observed for copper. Based on the analyses, the significant role of PHA in the adaptation of the C. necator H16 strain to the tested stressors was discussed, it did not consist only in the increase in the amount of polymer in the biomass, but also in enhancement of whole PHA cycle, which also leads to an increase of the pool of monomeric units showing protective functions. By adaptation to -captolactone, a unique precursor of 4HB, the copolymer P(3HB-co-4HB) was obtained. The properties of this copolymer are again more favorable than of the homopolymer P(3HB), even with a low content of 4HB, which we also achieved in a laboratory bioreactor. A further increase in the 4HB fraction could be achieved using deletion mutants with the absence of relevant genes, which is discussed more in the text. The production of PHA on models of lignocellulosic biomass hydrolysates originating from, for example, the food industry was tested in combination with the use of extremophile producers, when the preference of the contained monosaccharides (hexoses, pentoses) for individual producers was discussed. For the purpose to get closer to real hydrolysates, the resistance of the strains to relevant potential microbial inhibitors was also tested. The susceptibility of halophilic and thermophilic PHA producers to osmotic stress was used in the development of an alternative isolation approach that would reduce the economic and ecological burden of the process compared to standard extraction using chlorinated solvents. Application of SDS detergent at low concentrations while simultaneously exposing the cells to higher temperatures led to the gain of high purity polymer without loss of yield. The recycling process of used SDS is also a possibility.
Study on degradation processes of bioplastics

Biodegradable bioplastics are currently developed as a more environmentally friendly replacement for conventional plastics to be used in agriculture or packaging materials. As the production of these products increases, so will the likelihood of introducing them in the environment. Biodegradable materials are expected to biodegrade rapidly, thereby they should reduce the number of microplastics in the environment. Biodegradability is currently determined by biodegradation tests, the parameters of which are defined in very general terms and often do not reflect the actual conditions under which the bioplastic will degrade in the environment. There is a lack of knowledge about the fate and impact of these particles in different ecosystems (especially in soil, where contamination is expected to be highest). Therefore, in this dissertation, we critically evaluate the ISO soil biodegradation test and extend it with a subsequent determination of bioplastic residues in soil. For this reason, an analytical method for the determination of polyhydroxybutyrate and polylactic acid microplastics based on methods used for the determination of conventional microplastics. The effect of microbioplastics on soil is also discussed, focusing on the effects that may pose problems in agriculture. The influence of microbioplastics on abiotic (soil water properties, soil structure) and biotic (soil diversity, nutrient cycling) soil properties immediately after their entry into the soil are described. The effect of biodegradation and long-term presence of microbioplastics in soil on soil organic matter, nutrient content, and soil microbial diversity (described by determining the activity of 5 key enzymes involved in carbon, nitrogen, and phosphorus cycling) are also studied. The last chapter demonstrates the potential problems that can be encountered when bioplastic products are applied in agriculture, where the growth of fungi and molds naturally occurring in the soil can be accelerated. As a result of this dissertation, a comprehensive view of bioplastics in the environment is presented, which can serve to assess the suitability of the application of current generation bioplastic products in agriculture and the possible management of bioplastic waste.
