Molekulární diagnostika deficience lipoproteinové lipásy (LPLD) jako výběr pacientů vhodných pro genovou terapii (AAV1-LPL S447X )

Lipoprotein lipase deficiency is a malfunkction of a key enzyme for lipoprotein metabolism. Lipoprotein lipase deficiency can cause serious episodes of pancreatitis. The deficiency is caused by pathology in the LPL gene. There are many variants in the LPL gene and quite a lot of them are pathogenic.The first gene therapy in Europe was licensed for the treatment of lipoprotein lipase deficienciency using a gain-of-function variant of the LPL gene. A pattern of examination was established to including MLPA, Light SNiP, quantitative real time PCR and sequencing in order to find pacients for the treatment. Three pacients suitable for the gene therapy Alipogene tiparvovec (AAV1-LPL S447X) were found amongst a group of patients with history of pancreatitis.
Research of the ability of retention information from verbal education in nutrition mode and the effect of this education by overweight middle-aged people with type 2 diabetes mellitus

This diploma thesis focuses on the disease diabetes mellitus. The theoretical part describes the characteristics of this disease. It describes typical symptoms, classification and diagnostics. It also outlines and explains the complications that may occur as a result of the disease and that may be avoided or whose inception and development may at least be delayed. Depending on their type and extent, these complications may significantly reduce quality of life as well as overall life expectancy. It is therefore essential to ensure that patients are properly educated in the right therapy. Education itself is covered in the section of the chapter devoted to treating diabetes mellitus. The practical part of the diploma thesis is devoted to research, focusing on the ability to retain information following verbal nutritional education of patients with diabetes. The aim of the research is to determine the percentage of information that patients gain from education on diet therapy, thus confirming or rebutting the established hypotheses. Abraham in the New Testament

The thesis Abraham in the New Testament commences with the narrative of Abraham in the Book of Genesis, evaluates responses to that story in the Old Testament as well as in the extra-biblical literature, and then examines its use in the New Testament. Abraham is introduced as God's friend, a model of faith, and a forefather. The questions are raised as to who are entitled to think of themselves as heirs to Abraham's promises and what are the consequences for the Curch. Appendices provide a list of Bible verses wherein Abraham is mentioned or which make allusions to his story. Other attachments present key texts from the Old and New Testaments and their literal translations as source materials supporting the conclusions of the main text.
Life Stories and Professional Careers of Women Headmasters of Vocational High Schools

Thesis with the title "Life stories and professional careers of women headmasters of vocational high schools" deals with professional life of women headmasters of vocational high schools and analyzes this professional life. Per this analysis is detected, what is the benefit of these headmasters for school and if they promote a development of the school. Narrative analysis was used on this research.Beside the research there are answered other questions, which are related to professional life and are associated with the primary research question. There are described the phases of professional life and there are stated the effects, which influence the decision making of the headmasters. In the conclusion of research the headmasters alone evaluate their actions in position of headmaster.The primary research question is answered, how it results from research, all headmasters are benefit for their school. Schools, which were guided by them, had evolved and it is them, who are the reason of its growth. Recommendations for futher research is analysis of professional men headmasters.
Refeeding syndrome

Refeeding syndrome (RFS) is a serious metabolic disorder caused by improper procedure of realimentation in patients, who are starved or severely malnourished. All clinical features of the syndrome are result of many metabolic changes, which are mainly caused by depletion of phosphate, potassium, magnesium and thiamine, as well as retention of sodium and disturbed glucose metabolism. The theoretical part of this thesis summarizes the current knowledge about the prevention of the RFS. The research part of the thesis explores the influence of intense realimentation in high-risk patients, particularly the values of various biochemical parameters before or during the early beginning of the realimentation. We measured values of indirect calorimetry, BMI, serum concentrations of phosphate, potassium, magnesium, prealbumin, albumin, zinc, iron and vitamin D. We also monitored concentrations of phosphorus, potassium, magnesium and cortisol in urine. We reported reduced values of potassium in urine for all patients (5/5) and reduced values of phosphate in urine for four patients (4/5). In four patients (4/5) we also monitored decreased levels of serum prealbumin, including two patients even below the value of 0.1 g/l, which is considered to be a significant risk factor for the development of the RFS. Quality of life on home parenteral nutrition

This bachelor thesis pursues the topic of quality of life in patients on home parenteral nutrition. In the theoretical part is processed the problem of patient with malnutrition and the subsequent initiation of artificial nutrition support with a focus on parenteral nutrition. Further, the home based parenteral nutrition, its organization and patient care in the Czech Republic is described. The last part of the study is focused on assessment and evaluation of patients on home based parenteral nutrition and outcomes on the quality of their lives. The practical part evaluates quality of life in patients on home parenteral nutrition by SF- 36 standardized questionnaire. The questionnaire targets on the subjective assessment of physical health, emotional aspects of perception, patient socialization and the overall view of the patient health. The aim of the study is to compare hypotheses based on the assumption of the negative impact of home parenteral nutrition to quality of life in specific domains. The analysis of digital archive Ask the Library

This bachelor's thesis seeks to analyze records of answered queries, saved in their online digital archive of the Czech national digital reference service called Ask your library. Two first chapters are based on studying sources, found within a bibliographical exploration of the subject. The first chapter briefly presents the service (development, current state, function, organization and administration, receiving and answering processes, etc.). The second chapter describes in detail the creation of online archive with answered queries and researching information from it. In the third chapter is detailed analysis of answered queries. The presentation of the results forms a major part of the work. The analysis includes the presentation and interpretation of basic statistics published on the Web site (eg. the proportion of questions within years, the share of stakeholders on answering queries, etc..), and analysis of the content of the answered queries within selected thematic categories (content and nature of the response, used and others recommended resources). Finally, the author evaluates the results of the analysis of the online archive of the answered queries and perspectives of next development of this service. Enteral nutrition in oncology patients

This bachelor thesis considers possibilities of enteral nutrition and its influence on life quality of oncological patients. The thesis consists of a theoretical and an experimental part. The theoretical part compiles the issue of malnutrition and nutritional support with emphasis on enteral nutrition. It deals with malnutrition in oncology and its solution. In the experimental part, a study of influence of enteral nutrition on life quality of oncological patients was conducted. A question was, how health condition, life quality and energy intake of patients are changed after one month of enteral nutrition use. The survey includes a physical examination, measuring patients with a machine Bodystat Quadscan 4000, filling up a questionnaire, evaluation of laboratory tests, and analysis of the patient's diet. The results of the study prove positive influence of enteral nutrition on health condition of patients with respect to laboratory tests. According to the questionnaire, the life quality of patients increased and the diet report shows increased energy intake. The physical examination results and Bodystat measurements did not prove positive influence of enteral nutrition.
Deputy headmaster-shadow or second headmaster?

Final thesis deals with a research of the role of a deputy headmaster of secondary vocational schools in the Central Region, determines what is his role and how it affects the management and function of the school. Futhermore examines, if the deputy headmaster was a shadow of the headmaster, the one standing in the background, or he is the person, who participates in the decisions and takes over responsibility for the headmaster in the school management. In research was found that the position of deputy headmaster, in the surveyed schools, is in accordance with the definition of his powers, which are designated by the headmaster. He is always by his side and codecisions in those cases, which are given to him by his competences. The deputy headmaster also cooperates and is a good counselor to the headmaster. He stands in the background, but not because he would have been overlooked, but because he is given the role that the headmaster had intended to him. The headmaster r takes the deputy headmaster as his closest associate, who is indispensable when organizing, controlling and problem solving. He can also work independently, interact and represent the school. There is always the headmaster, who leads the school from the inside as well as from outside.
