T cells labelling by bimodal contrast agent for in vivo studies of stroke

Stroke is a serious brain injury, which causes sudden death or terminates in permanent neurological disability. Nowadays, tissue plasminogen activator (tPA) is used as the only effective treatment of stroke. One of the potential targets for novel therapy are T cells. Even though the explicit role of T cells in the pathogenesis of brain injury, amounts and timing of all T cell subtypes infiltrating into brain during the stroke still needs further investigation. The research in this field is complicated by the lack of efficient methods for in vivo cell tracking. Therefore the aim of this thesis was to develop a method of T cells labelling by MRI contrast agent in order to investigate T cells distribution in ischemic mice model using in vivo MR imaging. T cells were isolated from C57/BL6 mice in two step isolation protocol using gradient centrifugation and magnetic separation with the efficiency of 97 %. The isolated cells were labelled with 100 μg Fe/mL of Molday ION Rhodamine B contrast agent. The labelling efficiency after 17 hours of cells incubation was higher than 99 %. The labelled cells were cultured with CD3 and CD28 antibodies resulting into the 74 % viability of labelled T cells compared to 83 % viability of non labelled T cells. On curvature decomposition in circular space-times

Calculation of scalars obtained from the Riemann curvature tensor in co- ordinate components is not always efficient, this is true even in very simple spacetimes. Firstly, the calculation is not intuitive and secondly, many terms involved in such calculations tend to strongly diverge for example on black hole horizons, even though they should precisely "cancel out". Motivated by [1], main focus of this thesis are circular space times and introduction of 2+1+1 decomposition. The latter allows for rewriting curvature scalar using sectional curvatures, exterior curvatures and geometrically significant timelike congru- ences. Crucial part of this thesis is software implementation and verification of this approach in Wolfram Mathematica using xAct package. 1
Improvement of quality, function and transplantation outcomes of pancreatic islets using RNA interference

Transplantation of insulin producing tissue is the only therapeutic method so far allowing type 1 diabetic patients to achieve long-term independence of insulin administration. Transplantation of isolated pancreatic islets (PI) into liver represents a safer but less effective alternative to whole-organ transplantation. This is caused by a significant loss of transplanted islets within a short time after transplantation due to the instant blood-mediated inflammatory reaction (IBMIR). This reaction is triggered by molecules of tissue factor, abundantly expressed on the surface of PI cells. Tissue factor activates directly the coagulation pathway leading to platelet aggregation, complement activation, and infiltration of islet graft by leukocytes which results in a destruction of up to 60 % of transplanted tissue, and a formation of ischemic areas of downstream lying liver tissue. Tissue factor also stimulates angiogenesis which makes it necessary for revascularization of PI after transplantation. Inhibition of tissue factor gene in isolated PI using RNA interference provides an opportunity for short-term suppression of tissue factor expression, leading to protection of PI in the early post-transplantation period without hindering the connection of capillaries to the recipient vascular system later...
Starting points for a new physics curriculum

This thesis presents starting and supporting points for a new secondary school physics curricu- lum and the methodology which was used. The research design was inspired mainly by the objectivist grounded theory. 29 in-depth semistructured interviews were conducted with elite Czech physicists and there were identified 79 ideas about a secondary school physics curricu- lum. After approximately two years, 56 ideas were used in the questionnaire survey where re- spondents expressed on a Likert scale their degree of agreement with the ideas. Respondents were natural scientists (non-physicists), experts on didactics of physics, secondary school phys- ics teachers and 26 of 29 physicists who were also participants of the interviews. In the ques- tionnaire survey, there were identified ideas on which all groups of respondents agreed and which are stable in time in the group of physicists.
Effects of stable analogs of anorexigenic neuropeptides in models of metabolic syndrome

Obesity is a worldwide health problem and an effective treatment is still scarce. Anorexigenic neuropeptides, such as prolactin-releasing peptide (PrRP), have a potential for the treatment of obesity and its complications, but in their natural form they have several limitations such as poor bioavailability, low stability and inability to cross the blood-brain barrier after peripheral administration. Recently we have designed lipidized analogs of PrRP. Lipidization makes this peptide more stable and able to act centrally after peripheral administration. The aim of this study was to investigate the chronic effect of PrRP palmitoylated at position 11 (palm11 -PrRP31) on obesity and obesity-related metabolic parameters and to clarify mechanisms of its action. We used three rodent models of obesity: Wistar Kyoto (WKY) rats with high-fat diet-induced obesity (DIO) having intact leptin and leptin receptor as well as rodents with disrupted leptin function: leptin deficient ob/ob mice and fa/fa rats with a disturbed leptin signaling. Consumption of a high-fat diet in DIO WKY rats increased their body weight, caused strong glucose intolerance and increased liver mRNA expression of enzymes of de novo lipogenesis. The thesis deals with the disease diabetes mellitus type 2, especially compliance regime measures and education of patients with diabetes mellitus type 2 visiting diabetes clinics in Jeseník and in Prague. The work is devided into theoretical part and practical part. The theoretical part focuses on the characteristics of the diabetes mellitus, diagnosis and options of treatment. The practical part identifies and compares the degree of compliance with regime measures and education level in patients with diabetes mellitus type 2 in diabetes clinics in Jeseník and Prague. The thesis may outline the deficiencies in the education of patients with diabetes mellitus type 2 and their possible solution.
