Methylation of the regulatory sequences of integrated and extrachromosomal form of HPV 16 in tonsillar tumors

Human papillomavirus is connected with induction of cervical carcinoma as well as for some other anogenital carcinomas and subset of carcinomas of head and neck. Presence of viral E6 and E7 oncoproteins may induce cell transformation, higher load of oncoproteins is caused by the regulatory E2 protein inactivation. Aims of recent study are mechanisms of E2 protein inactivation. One option is integration of viral DNA into the host genome, which is located into the E2 gene region. Some carcinomas, where virus with extrachromosomal form was presented, were found. It appears that epigenetic changes can play the role in the development of this type of tumors, especially DNA methylation or mutation in the regulatory region of the virus. The methylation degree analysis on samples of tonsillar carcinomas with extrachromosomal and integrated form of the virus was conduced, as well as viral load of both groups was compared and the expression of E6 and E7 gene was confirmed. The results of methylation analysis showed increased methylation of the virus with integrated DNA. Mutations in the E2 protein binding sites are not revealed. Detection of human respiratory DNA viruses in the respiratory tract samples of immunocompromissed patients.

Respiratory tract diseases are of the most common infectious diseases among both children and adult population all over the world. Viruses are the most frequent cause of respiratory diseases. In healthy immunocompetent individuals respiratory infection proceeds mostly without major complications. Immunocompromissed hosts, for example patients after transplatation, are more susceptible to infection and even common infection may be life threatening for them. Human polyomaviruses KI (KIPyV) and WU (WUPyV) and human bocavirus (HBoV) are most frequently detected in the respiratory tract of patients with acute respiratory tract infection primarily in children and in immunosuppressed patients. However, clear causative link between presence of these viruses and the respiratory disease has not been established. In this retrospective study were tested by quantitative real-time PCR 822 (745 from adults and 77 from children) respiratory samples from 380 immunocompromissed patients included 326 adults and 54 children. Viruses were also detected in the 84 peripheral blood samples. The most frequently detected virus was HBoV (6,32 % positive pacients), followed by KIPyV (5,79 % positive pacients) and WUPyV (0, 53 % positive patients). Only HBoV was detected in blood samples. The objective of this diploma thesis is to create a financial plan for a selected company. The first part deals with theoretical knowledge which is necessary for the formation of the financial plan. The second part focuses on the presentation of the selected company, and creating a strategical and financial analysis. Based on all the gathered information, two options of a financial plan will be created. The last part presents an overall evaluation of the plan.
Human parvovirus B19 genotype study among the patients of Motol Univeristy Hospital

Parvovirus B19 is a common human pathogen that typically infects erythroid progenitors and causes hematological problems such as anemia and aplastic crises. The clinical presentation depends mainly on the immunological status of the patient. PVB19 can cause serious clinical disorders in immunocompromised patients after transplantation. More than 1500 samples from 90 patients who passed the HSCT in 2015 were tested for the presence of PVB19 in this work. This work describes the incidence of the virus and two typical periods of onset of infection in patients after the transplantation. Although several sources report the negative effect of PVB19 infection on the survival of allogeneic graft patients, this work did not confirm this assertion. Also, the results of this work suggest that allogenic grafts are not the main source for transmission, but that it is likely to be reactivated after long-term persistent or latent PVB19 infections. PVB19 is divided into 3 genotypes. Genotype 1 is the most widespread, genotype 2 is very rare in Europe for the last 10 years, and genotype 3 occurs mainly in tropical localities. Detection and pathological impact of human Parvovirus B19 in the general population and in the immunocompromissed host

Parvovirus B19 is a non-enveloped single-stranded DNA virus, belonging to the Parvoviridae family. Despite parvoviruses are one of the smallest DNA viruses, some of them are able to infect humans. In general, most of humans experienced parvovirus B19 infection. In case of infection in healthy individual there are usually no complications. Most frequently infected are children and their symptomatic infection is known as fifth disease - erythema infectiosum (EI). However, serious complications may occur if infected person suffers from erythropoiesis disorders, weakened immunity or is pregnant. This thesis aimes the structure of the virus, the course of the infection, infectious diseases caused by parvovirus B19 and particular genotypes especially to pathogenesis of specific disorders. Genotypic analysis of human cytomegalovirus in the patients after allogeneic haematopoietic stem cell transplatation.

In patients after allogeneic haematopoietic stem cell transplantation (HSCT) is a human cytomegalovirus (CMV) one of the most important viral pathogens. Its detailed characteristic could provide information about the impact of each CMV genotype on overall survival of the patient, and some serious complications, such as graft versus host disease (GvHD). This thesis deals with retrospective genetic analysis of samples from 1877 patients transplanted at the Clinic of Pediatric Hematology and Oncology, University Hospital Motol and the Institute of Hematology and Blood Transfusion since 2002. DNA from biological samples (especially whole blood) was isolated kit Qiagen DNA Blood Mini or Qiagen DNA Mini and samples were prospectively detected presence of CMV DNA. Samples were subsequently stored at -20 řC. Genotyping was performed using real-time PCR technologies to the genes of 2 structural proteins glycoprotein B, glycoprotein H and using sequence specific primers and probes. In 1343 samples (71.6%) from 390 patients there was only one strain of CMV; in 256 (13.6%) samples from 113 patients have detected mixed infection caused by two or more strains of CMV. Prevalence and biological consequence of chromosomally integrated human herpesvirus 6 (HHV-6) in Czech population

8 Abstract Human Herpes Virus 6 (HHV-6) consists of two closely related DNA viruses: HHV-6A and HHV-6B. The primoinfection proceeds at an early childhood usually as sixth exanthem disease or without any clinical symptoms. Both HHV-6 viruses are able to integrate to human genome using recombinant mechanisms which is unique compared to other human herpesviruses. The aim of the thesis is to study Ci-HHV-6 in a group of patients with malign disease and in the healthy population. We analysed 812 patients with malign disease and 420 healthy subjects from general population. The Ci-HHV-6 was assessed by real-time PCR, the specific localization of Ci-HHV-6 was determined using fluorescent in situ hybridization (FISH). Using comparative study, we did not identify significant difference between frequency of Ci-HHV-6 in patients with malign disease (1.11%) and healthy subjects (0.95%) (P-value 0.8). Consequently, we proved the heritability of Ci-HHV-6 in affected families. We determined the localization of Ci-HHV-6 to telomeric regions of chromosomes 2 and 18. We studied the production of viral proteins in the subjects with Ci-HHV-6. In this work, we conducted the first epidemiological study of Ci-HHV-6 in the Czech Republic. We also introduced novel methods which contribute to better characterization of this phenomenom.
