Marking of rainbow trout with Alizarin

Method of using alizarin red (ARS) for mass-marking of rainbow trout was verified and optimized. Based on the results, it was possible to create methodological recommendations for marking individuals of rainbow trout using ARS: 1. Marking can be used shortly after hatching (around 14 days) since the fin rays are sufficiently developed or calcified. The standard application time and solution concentration (1 hour, 150 mg/l) can be used if the expected detection time of marker is around 4-5 weeks. It is necessary to increase the application time on 3 hours in order to distinguish individuals 2-3 months after marking. There was no evidence of a negative effect of the dye on rainbow trout individuals (mortality) or a change in chemistry (pH, conductivity, oxygen saturation of water) within standard concentrations and application time and economic parameters. The use of Alizarin red can significantly reduce financial (around 1 CZK/individual) and time requirements for fish marking. When compared with other types of marking, costs can be reduced to 10 percent (or less) and time-consuming (meaning labor costs) can be reduced to several percent. Another advantage that is difficult to quantify is benefit from information obtained by marking on the farms.
Methodology for measuring the character of the mucus layer in fish

The aim of the methodology is to enable easy quantification of selected characteristics of the fish integument, namely the number of secretory cells in the surface layer of the skin, the amount of epidermal melanophores and the ability of the mucus layer to absorb UV radiation. The target group of the methodology are mainly persons from veterinary practice and ichthyologists dealing with the issue of fish integument, in relation to their health status or developmental-morphological studies. It can also be used in fish farming or protection. Representatives of different taxonomic groups were preferred in the selection of fish species for verification.

Based on the knowledge and evaluation, to enable the most efficient collection of material for subsequent research. Influence of sampling and fixation on measuring of genome size

Assessment of genome size is an important aspect of many modern studies on fish. Blood is the most common DNA source in fish studies due to its relative ease of sampling and processing. Fish blood, however, precipitates rapidly, leading to clotting. Consequently, anticoagulants (primarily heparin) are added at the time of sampling. Using of increasing heparin concentrations resulted in notable increases in fish erythrocyte genome size readings compared to a non-heparinised control. The differences observed were highly significant, suggesting that the results of previous comparative studies or those determining individual polyploidy may need reinterpretation.

Management plan of small water supply reservoirs of Landštejn type

The aim of the methodology is to provide managers and users of small water reservoirs Landštejn type with methodological tools for optimization of the water quality monitoring based on the current level of knowledge and research. Furthermore, provides a design of a methodology for fish community monitoring and optimization of treatments to reduce unwanted fish species and strengthen the status of suitable fish species with the aim of achieving the topdown effect and maintaining or improving water quality in these water reservoirs. The presented methodology summarizes the findings obtained during the monitoring and evaluation of selected parameters of water quality, planktonic and fish communities in water supply reservoirs Landštejn in recent years. It also contains an evaluation of applied treatments, focused mainly on fish stock. Unwanted fish species were caught and the predatory fish were stocked with the aim of optimizing the fish community composition and positively influencing the quality of the water in the reservoirs. The methodology contains recommendations for monitoring of the water quality in the reservoir, including monitoring of planktonic communities, monitoring of fish and recommended manipulation with fish stock. The methodology provides guidance and information for optimization of monitoring and treatments for small water supply reservoirs of the Landštejn type.

Cílem studie bylo sledování vybraných morfologických znaků sledovaných druhů či skupin lososovitých ryb : pstruh duhový (Oncorhynchus mykiss) původem z Dánska, Itálie, Jihoafrické republiky, siven americký (Salvelinus fontinalis), siven arktický (S. alpinus), siven obrovský (S. namaycush), a jejich hybridi (S. fontinalis x S. alpinus). Sledovány byly vybrané morfologické parametry - stavba těla, hmotnost vnitřních orgánů, struktura pokožky a žáber.
