Influence of sigle nucleotide mutations on influenza pathogenicity

Influenza virus caused several times the global concerns about the emergence of pandemics, in recent years. The most serious problem, in the fight against this virus, is its ability to quickly change its unusually antigenic specificity by reassortment and point mutations, especially in surface glycoproteins - haemagglutinin and neuraminidase. In this thesis I focused on point mutations in the hemagglutinin of influenza virus causing change in binding preference, which enables both influenza virus transmission between different hosts and unlimited proliferation in their tissues. The permanent change in amino acid sequence and thus the surface of hemagglutinin also largely prevents the host's immune system to destroy the virus efficiently and quickly enough to recognize when another infection. Each of the haemagglutinin subtypes has a different amino acid positions affecting its binding preference. Their summary is included in this thesis. Selected localities of Prague for teaching biology and geology

This bachelor thesis is focused on the most important Prague locations that are suitable for biological and geological excursion. Very important is practical teaching biology and geology. It is desirable that students understand the processes of ancient geological history, that they have seen with their own eyes preserved fossils and that they know how plants and animals which are integral part of teaching biology look like. In this point of view the theoretical teaching in the classrooms is insufficient. In the work is included biological and geological characteristics of selected sites aimed at their geologicla formation, rock, fossils, plants and animals that are found there. Itinerary for each excursion should be designed. There are links to worksheets made by methodical portal RVP which are the part of my work. Short introduction of teaching biology and geology in foreign countries and their access to excursion is presented as well. This thesis was designed for teachers of biology and geology to make preparations of their own excursions easier.

Interaction of polyomaviruses with proteasomal system of host cells Abstract: Viral family Polyomaviridae includes besides model organisms - mouse polyomavirus and SV40 virus, also human pathogens, for example, BK virus. Polyomaviruses are small non- enveloped viruses with double-stranded DNA. Understanding of their life cycle is important for their use in gene therapy and immunotherapy as well as for prevention and treatment of complications caused by these viruses. This thesis is focused on early phases of MPyV and SV40 infection studying, mainly on delivery of viral genome to nucleus and role of proteasomal system in this stage of infection. It was found out that inhibition of proteasomes by specific inhibitor leads to increase of early non-structural protein LT expression, which was chosen as marker for viral entry to the nucleus and successful viral expression. Relative localization of proteasomes and VP1 protein of MPyV and SV40 was monitored and it showed 10% colocalization of mentioned structures. Further, it was found out that proteasomal inhibitor MG-132 negatively influences the replication of both viral and cellular DNA. Interaction of influenza virus with cell defence mechanisms

Influenza virus infection is one of the most current problems nowadays. Its unique ability to strongly inhibit cell immune response on many different levels and its pandemic potential make it a subject of interest of many research groups. The Influenza virus uses mainly NS1 protein to inhibit the immune response. NS1 protein is able, on one hand, to bind RNA and mask it against recognition by cellular sensors and other proteins. On the other hand, NS1 protein possesses a catalytic domain. Using this domain it interacts with many cellular proteins, interferes with signal transduction and guarantees successful infection. NS1 protein is one of principal pathogenicity instruments of the Influenza virus and it deserves appropriate attention.
New approaches in vaccination against HIV

A B S T R A C T Most current vaccines are based on using whole-inactivated viruses. After creating the immune response and immune memory is organism able to cope with infection create by patogens. In the case of HIV, however, fail to produce the vaccine, which would have been able vaccinated individual from subsequent infections protect. Virus HIV attacks CD4+ cells and destroys the immune system. Rate of his replication is high and virus HIV is resistant to existed antivirotics. And he is resistant before cells, which conveying the immune response. Moreover, the virus persists in cell in the form proviral DNA. For a successful vaccine against HIV is developed a lot of new vaccines and vaccination procedures. One way is the using recombinat viral glycoproteins, which are incorporated into the membrane of virus HIV, which should produce in the vaccinated organism production of neutralizing antibodies. Some modern models of vaccines strategies don't target the virus itself, but they target the restriction of HIV infection by destroying infected cells via apoptosis, or cytokine secretion. Antiviral vaccination - hepatitis A virus

Hepatitis A is an ifectious disease caused by Hepatitis A virus (HAV) from Picornaviridae family. HAV is an uneveloped virus composed of positive single-stranded RNA and a capsid with icosahedral symmetry. This infection is wide-spread and endemic in many regions of the world. The main way of transmission is throught the fecal-oral route. The possible symptoms, that can appear during the disease are fever, fatigue, muscle-pain, nausea, vomiting and afterward yellowish discoloration of the skin and mucous membranes. On the other hand, the disease can proceed without any symptoms, that is common especially in the childhood. The people can shelter from the disease by aplication of vaccine. At this time there are four inactivated hepatitis A vaccines at the world market. All of the vaccines are highly effective and provide a long-term protection. This work is intent on the infection of hepatitis A virus, the possibilities of prevention with vaccines and the inclusion of curriculum about viral diseases and vaccination in the School Educational Programmes.

Cancer treatment with polymer-bound cytostatic drugs and its potentiation through immunomodulation Poly[N-2-(hydroxypropyl)-methacrylamide] (PHPMA) is a synthetic water soluble and biocompatible polymer which can be used as a carrier of a cytostatic drug and an antibody as a targeting moiety. The antibody ensures the site-specific delivery of the conjugate. Nevertheless, even polymeric conjugates without any tumor-specific targeting moiety are passively accumulated within solid tumors via so called Enhanced Permeability and Retention (EPR) effect, in case that their molecular wight is at least 40 kDa. Antibody-targeted polymeric drugs have been shown previously to have a cytostatic activity in vitro and an antitumor activity in vivo. Since treatment of cancer diseases in practice is far from such ideal conditions and many tumors have no strictly specific marker suitable for targeted therapy, upgrading of the treatment efficacy represents the major challenge. One of the possible ways how to improve insufficient chemotherapy outcome can be using of a combination of polymer-bound cytostatic drug and potent immunomodulation able to induce a robust anti-cancer immune response. Utilization of Biology Olympiad in schools and implementation of selected tasks into highschool teaching.

This thesis deals with the Biology Olympiad and remaking of selected tasks of region round. The remaked tasks can be used in highschool education. The thesis includes theoretical and practical part. The theoretical part provides general information about the Biology Olympiad (BiO), charts the achievements of Czech students in International Biology Olympiad (IBO), includes literary review of selected topics. The practical part encompass four remaked tasks of different topics: Eucaryotic cell as a mosaic, Water in life of animals, Animals' areas and their distribution, History of the flora in Czech Republic. The tasks are processed in the form of worksheets. The worksheets were tested at schools. Knowledge of students was tested by using of created pretests and posttests. The practical part also presents the result of research based on a questionnaire survey focused on the involvement of school in BiO and teachers' opinions on these topics.
