Biofluid biomarkers of Alzheimer's disease and Lewy body disease and their relationship with cognitive and structural markers

The diagnosis of Alzheimer's disease is based on the determination of specific proteins in cerebrospinal fluid (CSF) or their imaging using positron emission tomography. Both methods are invasive and can expose the patient to risk and discomfort. Blood biomarkers, therefore, represent hope for early diagnosis and monitoring of individuals at increased risk of developing Alzheimer's disease. The diploma thesis aimed to determine biomarkers in blood using the ultrasensitive Simoa TM (Single molecular assay) technology and to verify their relationship to the values in CSF. The second aim was to track the relationship between blood biomarkers and cognitive markers of Alzheimer's disease. We demonstrated a positive relationship between serum, plasma, and CSF p-tau 181 concentrations. A negative correlation was observed between the concentrations of p-tau 181 in blood and the results of the MMSE test, which is considered a standard cognitive assessment tool. These findings suggest that plasma and serum p-tau 181 may greatly help the clinical identification of neurodegenerative diseases in primary care. This diploma thesis deals with national assessment of language abilities of hearing impaired pupils and closer focuses on pre-lingual deaf pupils at the ends of the primary and lower secondary schools. Based on the definition of the characteristics of the national assessment, the diploma thesis also focuses on an overview of currently used language tests, both at international level as well as it gives information about the practice in selected foreign education systems. Special attention is devoted to information that deal with the assessment of language abilities in the Czech Republic, and the form of participation of deaf pupils in such testing . The practical part is done by a research which was done to find out the views of those involved in the education of pupils with hearing disorders just the current assessment practice language skills of the pupils. In the final part of my diploma thesis is a possible outline for innovations for testing related to information from the preceding parts of my diploma thesis.

Tests and testing of sign language abilities of deaf children

This bachelors thesis deals with the assessment of skills in sign language. It includes a deep insight into the general issue of evaluation methods (especially of creating tests) as well as description of selected tests of sign language. Part of the work is devoted to modifications of existing tests to which process linguists and other experts now often tend. Also is introduced a specific modification procedure of selected sign language test into Czech sign language. This should serve in the future to a complete version of such test.
