Title: Dynamic light scattering in the world of biomolecules Author: Katarína Križanová Department: Institute of Physics of Charles University Supervisor: RNDr. Kateřina Hofbauerová, Ph.D., Institute of Physics of Charles University Abstract: Dynamic light scattering (DLS) represents a non-destructive method which can determine the hydrodynamic radius of particles in a sample by laser scattering. Consequently, their real sizes can be calculated or, in the case of proteins, molecular masses can be estimated. DLS is nowadays widely used for the study of biomolecules and their reactions, as is documented in the thesis in a brief review. Our experimental work was focused on the measurement of proteins by DLS - lysozyme was used as a model protein. The laboratory protocol for sample preparation and DLS measurement was successfully optimized. In the framework of methodology optimization, the ability of correct detection of small particles was tested using sucrose. We came to the lowest limit of particle dimensions measurable by DLS using sucrose. We were able to observe the linear dependence of the measured hydrodynamic radius on sucrose concentration in the solution and we were also able to determine the hydrodynamic radius of sucrose correctly, in agreement with literature. Title: Spectroscopic study of key biomolecular interactions for therapeutic applications of modified oligonucleotides Author: Martin Kříž Department: Institute of Physics of Charles University Supervisor: Prof. RNDr. Josef Štěpánek, CSc., Institute of Physics of Charles University Abstract: This dissertation thesis is focused on study of biomolecular interactions that are crucial for therapeutic applications of modified oligonucleotides. Interactions of nucleic acids, in particular hybrid DNA: RNA duplexes with fully complementary sequences, as well as DNA: RNA duplexes with a central mismatch were studied. The molecular model was created. With this model the influence of five different chemical modifications in the DNA strand to its binding ability was studied. Structural changes were characterized by Raman spectroscopy using a precise differential and double differential spectra. These data were supplemented by thermodynamic parameters computed from melting curves obtained by UV absorption spectroscopy. Furthermore, the Raman spectroscopy was applied on study of conformation changes in the RNase L protein. These changes in the conformation occur after ligand binding, which in this case, either natural or modified short oligonucleotide. Characterization of proteins of 2'-5' oligoadenylate pathway by means of vibrational spectroscopy

The work concerns to structural characterization of two important proteins of 2'-5' oligoadenylate pathway participating in an immune response of organism to a viral infection. Studied proteins were ankyrin domain of mouse RNase L, the C-terminal part of human phosphodiesterase 12 and the complete human phosphodiesterase 12. The proteins were characterized by Raman spectroscopy, infrared spectroscopy, electronic circular dichroism, dynamic light scattering and in addition by two non-spectroscopic methods- differential calorimetry and electrophoresis. For each protein the secondary structures, thermal stability, weight of oligomers and generally a basic characterization by above mentioned methods were provided.
Atomic force microscopy in the region of biomacromolecules

Atomic force microscopy (AFM) enables sample imaging at the micro and nanoscale. Recently, the method is applied to investigate biomacromolecules. Here, we describe the basic principles of AFM with a special emphasis for bioapplications. We tested experimental abilities of Alpha 300 - the Raman microscope with AFM/SNOM accessory from WITec company. The ability of AFM to study objects of cellular dimensions was demonstrated on erythrocytes and green algae Desmodesmus quadricauda. We were able to observe growing of lysozyme protein fibrils on day scale - from dimensions of seeds (~3 nm height) up to fibrils itself (3-10 nm height and 100 nm up to micrometers length). Subsequently, we observed separate protein molecules of thyroglobulin (~6 nm) and also γ-globulin (~3 nm). It seems plausible to image objects up to 2 nm dimensions by the given device with respect to the signal/noise ratio.
British Foreign Policy: the Iraq-Kuwait Matter in the UN Security Council from 1997 to 2003

This diploma thesis "British Foreign Policy: the Iraq-Kuwait Matter in the United Nations Security Council from 1997 to 2003" deals with the policy of the British Government of the Prime Minister Tony Blair in the UN Security Council charged with maintaining international peace and security against the Saddam Hussein's regime in Iraq. The research starts with the victory of the Labour Party in the general election in Great Britain in May 1997 and ends with the use of military power of the coaliton led by the USA against the Iraqi regime in March 2003. The main objective of this diploma thesis is to examine the relationship between the foreign policy of the British Government formulated in basic documents (The Labour Party's 1997 and 2001 General Election Manifestos) and foreign minister Robin Cook's The FCO Mission Statement of May 1997 and its practical fullfilment in a concrete crisis situation. The thesis also aims to define the main strategic goal of the British policy in the Iraq-Kuwait matter in researching time, and to decide if the policy was succesful.
Analysis of the media image of Milos Zeman in the ČTK news for the first year in presidency

This thesis titled Analysis of Media Image of Milos Zeman as Portrayed by CTK during the First Year of Presidency focuses on news releases made by Czech News Agency CTK that within the time frame of 8. 3. 2013 - 8. 3. 2014 inform about the President of the Czech Republic Milos Zeman. The goal of this thesis was to aquire a complex media impression of President as presented by CTK's news production. Primarily this thesis inquires into the total number of news releases within particular time periods, their topics and tonality. Secondarily this thesis observes the trends of these phenomenons within a one-year time horizon and the connection and coherence among the aforementioned phenomenons. The basis of this thesis is theoretical context that deals with news releases and pursues media construction of reality in the news. For this research there was used quantitative content analysis with qualitative overlap in form of emotional impression of news releases. As a data resource there was used an aggregate of texts from CTK's infobank. Research proved certain hypothesis, as e.g. a long-term dropping tendency of media interest in President Zeman or a higher number of negatively rather than positively tinted news. Translation of obsolete expressions in the works of Victor Hugo and Émile Zola

The subject of this thesis is the translation of diachronically marked expressions in the works of Victor Hugo and Émile Zola and their use in Czech literary translation in general. This paper gives a definition of "obsolete words" and it follows their usage in Czech and French literature at different times. It summarizes Czech and French Romanticism and Naturalism and it describes the individual styles of Hugo and Zola. It presents several translation theories concerning the translation of old literature, obsolete words and the ageing of translation. It analyses the most recent Czech translations of the two authors, and based on our findings, it formulates a hypothesis about the current approach to translation of archaisms and "historisms" in the works of these authors. Bachelor thesis has analyzed the accidents on the secondary schools, which are common part of studies. I recapitulate the types of injures in my work, which are divided to the groups.These dates are extraced from database of Emergency of South Bohemia. Concre-tely are this intervention in the secondery school in České Budějovice, during the years 2008 to 2018. My work deal about the most frequent diagnosis,where is emergency interferes, and analyzed most vulnerability group students from the aspect age and genders.
