Laboratory diagnosis of pheochromocytoma

1 Phaeochromocytomas are tumors predominantly rare to chromaffin tissue in adrenal glands' medulla. Tumors in the extra-adrenal chromaffin tissue are called paragangliomas. Phaeochromocytomas cause secretion of high amounts of catecholamines, which can lead to fatal consequences if not medically treated. The aim of this thesis is to summarize the so far known facts about this severe disease in a written review. The first part of the study focuses on issues of phaeochromocytoma research and lists general information about these tumors and their clinical manifestations. The importance of genetic influence in connection with treatment strategies for patients with phaeochromocytoma is also discussed. The second part of the thesis summarizes procedures and methods used in laboratory diagnostics of phaeochromocytomas. The conclusion briefly describes the possibilities of tumor localization using imaging technologies and procedures of subsequent treatment of patients with phaeochromocytoma.

The aim of this thesis is to contribute to understanding the mechanisms and regulatory pathways by which hormones influence prostate tissue. Includes a total of 4 experimental work dealing with hormonal environment in men with prostate cancer and benign prostatic hyperplasia.
Genetic backround and new biochemical markers in pathological pregnancy

Receptor for advanced glycation end products (RAGE), its soluble form (sRAGE) and glyoxalase 1 (GLO 1) are important part of pathogenesis of many chronic diseases. The aim of this thesis was to elucidate role of sRAGE, four chosen RAGE polymorphisms and one GLO 1 polymorphism in physiologic pregnancy and in pregnancy with complications. Serum sRAGE levels were determined in healthy pregnant women (N=120) and in pregnancies complicated with threatening preterm labor (N=99), preeclampsia (N=35), intrauterine growth restriction (IUGR) (N=22) and intrahepatic cholestasis of pregnancy (ICP) (N=14). Four RAGE polymorphisms (RAGE -429T/C, -374A/T, RAGE Gly82Ser (557G/A), RAGE 2184A/G) and one glyoxalase 1 polymorphism GLO 1 Glu111Ala (419A/C) were studied in the same population of healthy pregnant women and women with pathological pregnancy. Serum sRAGE levels are low in comparison to non-pregnant controls, but they vary during the physiologic pregnancy. Serum sRAGE levels are low in the 1st trimester, increased in the 2nd trimester and again decreased in the 3rd trimester. Women with premature labor have significantly decreased serum sRAGE levels in comparison to women with threatening premature labor and in comparison to healthy pregnant women. sRAGE correlates negatively with leukocyte count in preterm...

Postprandial changes of gastrointestinal hormones in patients with anorexia nervosa and bulimia nervosa.

Background: The present work was aimed to contribute to current research of gut-brain axis peptides ghrelin, obestatin, neuropeptide Y (NPY) and peptide YY (PYY) in women patients with anorexia nervosa (AN) and bulimia nervosa (BN). These hormones play an important role in regulation of food intake and energy homeostasis and their secretion is disturbed under conditions of eating disorders. Various types of consumed macronutrients may induce different plasma hormone responses, therefore we studied plasma levels of ghrelin, obestatin, NPY and PYY after consumption of a high-carbohydrate (HC) and high- protein (HP) breakfast within the meal tests performed under medical supervision. Methods: Plasma hormone responses to high-carbohydrate and high-protein breakfast were examined in patients with AN (total n = 14; age: 24,6 ± 1.8 years, BMI: 15,3 ± 0,7), BN (total n = 15; age: 23,2 ± 1,7 years, BMI: 20,5 ± 0,9) and healthy controls (total n = 14; age: 24,9 ± 1,4 years, BMI: 21,1 ± 0,8). Blood samples were drawn from the cubital vein using an intravenous cannula, the first blood drawn was collected before meal, and then 30, 60, 90, 120 and 150 min after breakfast consumption.
