Bártová, Hana ; Obšil, Tomáš (advisor) ; Gryčová, Lenka (referee)
The goal of this diploma thesis was to find optimal conditions for expression of ASK1 kinase in prokaryotic expression system and to optimize purification protocol which enables preparing of milligram amounts of stable and soluble protein. Different conditions of expression were tested in E. coli cells including temperature of expression, cultivation medium or the length of induction. Different methods of purification were tested during the development of the purification protocol. The final protocol is based on chelate chromatography followed by gel permeation chromatography. The result of the diploma thesis is a protocol that allows preparing 1 mg of pure ASK1 kinase from 1 liter of medium.

Dušková, Markéta ; Bártová, Hana (advisor) ; Jablonský, Petr (referee)
This bachelor thesis deals with the analysis of bank charges associated with the current account. The aim of the thesis is to find the best current account for students by using two model clients for selected accounts. Another objective is to analyze the possibilities of covering the costs incurred with the use of a current account by means of financial rewards provided by some banks in the current account and by other bank products. The partial objective of the thesis is to verify the hypothesis whether small low-cost banks offer a current account associated with lower bank charges than large banks.
Effect of selected women's religious orders in the social and health
BÁRTOVÁ, Hana
The aim of this thesis is to monitor the work of selected female orders in social service and healthcare and to classify the target groups that nuns work with. At the same time the thesis studies the interdigitation of spiritual life and the values of conventual order with practical nuns' work and it also monitors the experience of particular nuns relating to their work in ordinary life outside their community. An important aspect is also their concept of social work and healthcare with respect to their order and their concept with respect to secular concept. To meet the target of this thesis I have chosen a qualitative research method, particularly a half-structured dialogue method, which provided both free narration of individual nuns and possible further questions of the inquirer, which helped explain the details. In chapter "Results" there are portraits of each nun, created on the basis of half-structured dialogues. The portraits keep their authenticity, although redundant words were omitted and dialectal and colloquial expressions were translated into standard Czech. The main value of this thesis lies in the compact overview of induvidual institutions, where nuns work. These organizations are either own by particular congregations, which concern Sisters of Mercy of St. Borromeo and Congregation of the Mercy Sisters of the Third Order of St. Francis under the Protection of the Holy Family in Brno, to whom these organizations were returned in the 90s of the 20th century or the nuns work in organizations that are owned by someone else, which is the case of Daughters of Charity of st. Vincent de Paul. The significant value of this thesis lies in the complete number of nuns in particular congregations, the data are clearly written in the table, created on the basis of the study of all parts of the Catalogue of the male and female congregations in the Czech Republic. Table 12 shows the number of nuns in particular congregations from the end of the 90s of the 20th century till the year 2015. Table 13 shows the number of nuns in selected congregations who work in healthcare and social service at present.
Selma Brandl: Die Moderne Reitschule, Der richtige Weg zur Harmonie im Sattel (chap. Dressur) - commented translation
Bártová, Hana ; Svoboda, Tomáš (advisor) ; Kloudová, Věra (referee)
The object of my bachelor thesis - which is a translation of a text about horse riding and its commentary - is to make a translation that could be published without many improvements in the magazine Jezdectví or on the website Equichannel. While translating, I followed the translation process of Jirí Levý which he describes in his book Umení prekladu. First, I analysed the original text according to Christiane Nord, then I looked for text parts difficult to translate. I analysed them in their broader context and I chose suitable Czech equivalents for them. During terminology translation, I followed the rules of Czech Equestrian Federation and other literature. Because there is not enough Czech written or into Czech translated literature about horse riding - especially about dressage - I would like to extend the literary basis of this sport. Dressage and horse riding in general are not as popular as e.g. in Germany that is why I try to address the recipient more strongly to get him interested in the text. The first part of my work is dedicated to the translation, in which I do not leave out translation of the picture legend; the second part deals with the analysis of the original text and in the last part I discuss translation problems that I had to solve and I confront the original text with the translation...
