Anežka Adamcová Supervisor: Prof. RNDr. Dalibor Šatínský, Ph.D. Title of the Dissertation Thesis: The use of HPLC technique in the analysis of phenolic compounds in plant material of fruit trees Although fruits are well studied and known for their high content of antioxidants and positive health benefits, the main aim of the presented dissertation thesis consists in the characterization of biological active compounds in individual parts of fruit tree material (leaves, bark, buds, flowers, and wood chips). The theoretical part is focused on fruit trees and the characterization of phenolic compounds. Their properties and the effects based on current scientific studies are shown. A large part of thesis is dealing with the chromatographic methods for the separation of phenolic substances including the current trends. The extraction process is also included. The practical part deals with the validation and development of screening HPLC-DAD method for the analysis of monitored phenolic compounds (phloridzin, phloretin, chlorogenic acid, quercitrin, rutin, arbutin, 1,5-dicafeoylquinic acid, and 3,5-dicafeoylquinic acid) in individual parts of fruit trees. Bachelor thesis Charles University in Prague, Faculty of pharmacy in Hradec Králové Laboratory technician This bachelor thesis deals with diarrheal disease caused by Campylobacter sp. and Salmonella sp. The thesis is divided to four sections. The first section is about diarrhoea and its characteristics and causes. In the second section the characteristics of Campylobacter sp. is described and the major part of this section describes campylobacter enteritis or campylobacteriosis. Campylobacter jejuni is the most common agent. The third section deals with Salmonella sp. It again describes characteristics of this genus and focuses on salmonellosis enteritis or salmonellosis. Salmonella Enteritidis is the most common agent. Both genera are gram-negative bacteria and they affect intestinal tract. They cause bacterial enteritis, which is often connected with diarrhoea. Incidence of campylobacteriosis exceeded the incidence of salmonellosis for the first time in 2007. Infections have similar course. Besides diarrhoea, fever and stomach ache can also occur. RNDr. Dalibor Šatínský, Ph.D. Title of Diploma Thesis: Determination of floridzin and other phenolic compounds in apple leafs by HPLC This diploma thesis is divided to theoretical and experimental part. The theoretical part is focused on phloridzin and its characterization. In the experimental part the phloridzin, phloretin, chlorogenic acid, rutin and quercitrin were analyzed with a high performance liquid chromatography (HPLC) method from leaves of genus Malus. These compounds were identified in barks and buds of apple trees. The experimental part deals with optimization and validation of method too. Column YMC-Triart C18 ExRS 150 x 4.6 mm x 5um, 8 nm and gradient elution consisting of organic part of ACN and water part with phosphoric acid (pH= 2.2) was used for analysis. The detection was performed by DAD detector at wavelengths 280 nm, 327 nm a 354 nm. The temperature of column space was 30 řC, injection volume was 1 μl and flow rate 1 ml/min.
