Development of methodology for detection of circulating nucleic acids in cancer patients

Cancer diagnosis represents one of the most common causes of death in the world, also linked to an ever-increasing incidence of even some more difficult-to-treat variants. One of the most common types of cancer in the world is colorectal cancer, which ranks second in terms of mortality. For these reasons, various methods have been developed for an early diagnosis of the disease. One of the modern and non-invasive methods is the so-called liquid biopsy, based on the detection of circulating DNA and RNA from the blood of patients. This method is also suitable for choosing a therapy, monitoring its effectiveness, and observing the relapse of the disease. In this work, a methodical workflow was created for the monitoring of circulating nucleic acids for its further use in a cohort of patients with colorectal cancer. Various commercial kits were used for the isolation of circulating nucleic acids from plasma of healthy controls, among which the best ones, in terms of quality and quantity, was chosen. The threshold for hemolysis detection by qPCR was also determined using samples from healthy donors. ZINOVEVA, P. Strategic financial planning and forecasting of economic development of enterprises in the agrarian sector. Diploma thesis. Brno: Mendel University in Brno, 2023. The thesis focuses on the identification of key aspects for improving strategic planning and forecasting of financial development of enterprises in the agricultural sector. The literature search provides an overview of the current available knowledge on the topic. The methodological basis of the thesis was selected methods of scientific research; analytical, tabular, graphical outputs were used, as well as a summary of methods of economic and statistical analysis. The application part of the thesis focuses on the current situation of the enterprise in the agrarian sector in the selected Belgorod region by means of strategic analysis. According to the results of the study, in order to develop strategic planning and forecasting, strategic directions were proposed to increase the efficiency of the use of resource potential of the regional agro-industrial complex.

The second part of the thesis offers a prognosis of the development of this game species on the basis of data obtained from a mathematical model, assuming that there will be no major changes in the way of management of Japanese Sika deer in the hunting grounds of the Bouzov breeding area.
Prediction of therapeutical response to neoadjuvant therapy in rectal cancer

The treatment of locally advanced rectal cancer is multimodal. It includes neoadjuvant radiochemotherapy (NCHRT), which reduces the risk of local recurrence. However, this treatment is also accompanied by side effects. Accordingly, there is an unmet need to identify predictive markers allowing to identify non-responders to avoid its adverse effects. We monitored circulating tumor DNA (ctDNA) as a potential liquid biopsy-based biomarker. We have investigated ctDNA changes plasma during the early days of NCHRT and its relationship to the immediate tumor response as well as overall patients survival. In all patients, ctDNA was strongly reduced or completely eliminated from plasma by the end of the first week of NCHRT, with no correlation to any of the parameters analyzed. As ctDNA was reduced indiscriminately from the circulation of all patients, therefore the dynamics during the first week of NCHRT is not suitable for predicting the immediate therapeutic response in rectal cancer. The baseline ctDNA presence represented a statistically significant negative prognostic biomarker for the overall patient survival. However, the general effect of rapid ctDNA disappearance apparently occurring during the initial days of NCHRT is noteworthy and should further be studied. Keywords: rectal cancer, neoadjuvant...
Analysis and forecast of prize money development in selected sports taking into account unexpected external influences

Title: Analysis and forecast of the development of prize money in selected sports, taking into account unexpected external influences. Objectives: The main objective of this thesis is to analyze the development of prize money at the largest sporting events - tennis, golf and biathlon, and then create a forecast until 2033. A partial objective is to consider the effects of unexpected external influences (covid-19 and the war between Russia and Ukraine) on the prize money in recent years, which significantly affected the prize money. Methods used: Quantitative prognostic methods in the Excel program were used to compile the forecast of selected sports events. Using a linear and exponential trend line, the future course of the monitored development was recorded. In most cases, the exponential trend line, which showed a higher R2 reliability value, had a higher predictive value. Results: The result is a forecast of the development of prize money at the largest sporting events of tennis, golf and biathlon and an assessment of external influences (covid and the war between Russia and Ukraine) on their development. Dynamics of the volume-volatility relationship in the currency markets

This work investigates the volume-volatility relationship dynamics in the currency markets using data of five currency pairs in the period between 2010 and 2022. By employing multiple specifications of the HAR model with volume- related regressors and also with time-varying parameters (TVP), we examine the relationships' changing dynamics over time with a focus on improving volatility forecasting performance. Our main findings suggest a strong correlation between volume and volatility. The TVP-HARV model shows significantly changing dy- namics of the volume-volatility relationship, especially during periods affected by politics, changing monetary policies or global crises. The proposed models, however, do not improve out-of-sample volatility forecasting performance com- pared to the benchmark HAR model. The causal effect in the volume-volatility relationship in the currency markets is slightly more substantial in the direction of volatility towards volume, where we find slight forecasting improvements. Our findings conclude that volume and volatility in the currency markets are mainly moving simultaneously with a very strong correlation and much weaker and often insignificant causal effects on both sides, which supports the mixture of distributions hypothesis.

Teoretická část se věnuje finančním ukazatelům. z nichž některé jsou vybrány pro statistickou analýzu, ukazatelů analýzy, časovým řadám, regresní a korelační analýze. V analytické části je provedena finanční a statistická analýza, která určuje současnou situaci a očekávaný vývoj ukazatelů v následujících letech. Poslední část zahrnuje návrhy na zlepšení současné situace na základě zjištěných problematických oblastí.

The bachelor thesis focuses on the analysis of economic data of WYZ, s. r. o. The first part of the thesis is devoted to basic theoretical knowledge in the field of financial analysis and statistics. In the second part, the actual analysis of the current state of the company is carried out on the basis of the company’s accounting data and using the methods described in the previous part. The last part of the bachelor thesis summarizes and evatuates the results of the analysis and also proposes some measures that could help the company to improve the current economic situation.

It specifies individual indicators, the results of which are analyzed on the basis of the financial statements provided. In the statistical part, the time series and the associated forecast for the coming years are specified. Finally, it contains suggestions for improving the financial situation, which will help the accounting unit to manage better.
Prognosis in Ischemic Stroke Patients and Related Prognostic Faktors

Stroke is a significant cause of morbidity and mortality in the developed world. In the theoretical part of the thesis, we try to summarize the overall knowledge of stroke, the definitions and the general approach to the diagnosis and treatment, with a focus on ischemic stroke. Furthermore, the issue of acute phase treatment and secondary preventive treatment of ischemic stroke is reviewed and the modalities of the subsequent neurological deficit evaluation, the occurrence of mood disorders and the assessment of the quality of life of individuals after stroke are discussed. Our research aimed to assess the perspective of ischemic stroke patients admitted to the hospital. The first part is focusing on the mortality of patients with ischemic stroke in the period 2003-2019 in the Westbohemian region and its relation to secondary prevention. The second part aims to identify the main clinical and psychosocial factors affecting the quality of life of chronic stable post-stroke patients after their first in-life episode of stroke. The relation of poor quality of life indicators, mortality and conventional cardiovascular risk factors is prospectively assessed.
