Canonical and non-canonical signalling triggered by activated TRAIL receptors in human cells

TRAIL ligand can trigger apoptosis of permissive human cells via engagement of its two pro- apoptotic receptors TRAIL-R1 (DR4) and TRAIL-R2 (DR5). Its ability to induce apoptosis independently on p53 status and to selectively kill cancer cells in vitro and in vivo made this ligand an attractive target in cancer research. However, acquired resistance of primary cancer cells, unsatisfactory outcome of clinical trials and recent studies arguing that TRAIL might under specific conditions promote cancer progression, opened new plethora of questions, which need to be addressed. Though both receptors DR4 and DR5 are ubiquitously expressed, different types of tumours show preference for either of the receptors. The relative participation of DR4 and DR5 in TRAIL- induced signalling is still largely unknown. To analyse TRAIL receptor-specific signalling, I prepared Strep-tagged, trimerised variants of recombinant human TRAIL ligands with high affinity for either DR4 or DR5 receptor. Using these receptor-specific ligands, I examined a contribution of individual pro-apoptotic receptors to TRAIL-induced signalling pathways. This bachelor thesis deals with strategic analysis of the company Termo Děčín a.s. The thesis is divided into two parts (theoretical and practical). The author defines the theoretical part by using the basic concepts from the literature listed. The practical part parallels the theoretical part where the author applies theoretical knowledge in practical part. It is an analysis of the company both internally and externally. These findings are from a financial analysis which provides the basis for a strategic analysis of the company.

In the thesis is discussed problem with soil compaction in the agriculture in the Czech Republic, causing of aspects of this phenomenon as well as a method suppressing this phenomenon and prevent its. In this work are also mentioned machinery and equipment and their possibilities editing to reduce the compression of the soil. There are also the results of field measurements, which were carried out during the season on land that has been processed by different technology in several of its parts and other measurements carried out occasionally in the Czech Republic to observe the state of soil compaction.

PharmDr. Petr Pávek, Ph.D. Title of Doctoral Thesis Gene expression regulation of biotransformation enzymes and transporters in placental barrier Placenta is a unique organ facilitating the communication between mother and fetus. It serves as a respiratory, excretory, endocrine and metabolic organ during intrauterine development of the fetus and is necessary for maintenance of pregnancy and for the fetal protection. Syncytiotrophoblast is a single layer that represents the critical morphological and metabolic component of the placental metabolic and exchange barrier. The placental trophoblast contains multiple drug transporters and metabolizing enzymes that form placental metabolic barrier. The levels of the enzymes are not stable and fluctuate throughout pregnancy. Most of the enzymes are expressed at very low mRNA levels, however, in some cases there is absence of any relevant detectable protein or catalytic activity. Cytochrome CYP1A1 is the only placental xenobiotic-metabolizing enzyme of the cytochrome P450 superfamily for which significant expression and catalytic activity have been conclusively demonstrated in placental trophoblasts...
Legal and economic aspects of termination

This thesis focuses on the institute termination of employment from the legal and economic point of view. From a legal perspective, it is mainly about the presentation of the most common ways of termination of employment, including acquainting the reader with a steady legal interpretation. From a microeconomic point of view, the termination of employment relates to the consequences for employers and employees and from a macroeconomic perspective, with consequences for the whole society. This is followed by a practical part which connects the foregoing and the current example of OKD Inc., which employs nearly 10 thousand employees and currently finds itself in serious financial difficulties, the state estimates the costs associated with the mass redundancies at different alternatives. These variants are bankruptcy or reorganization provided financial assistance to the state. At the conclusion of the two calculations are compared and outlines a list of partial measures to mitigate the effects of similar events in the future.
Implementation of managing and controlling system in public governance

The current legislation that regulates internal managing and controlling system in administrative bodies of the public governance lags behind in the implementation of international experiences and standards for excersing the internal audits. This fact causes many reactions in the general public and also in the professional society. Alarming news about corruption and wasteful use of the public sources are just the token of the problem. The goal of this thesis is to define and analyze the process of implementing the managing and controlling system and subsequently to create a plan for a successful implementation of the effective managing and controlling system in the public governance. In the theoretical part are characteristics and goals of the managing and controlling system, legislative framework and description of components for the managing and controlling system, that are based on international experiences and practice. In the analytical part is the analyze of the particular elements from the managing and controlling system and finally in the practical part is the proposal of implementation managing and controlling system and also the proposal of directive for the internal control, which is supposed to simplify the practical use in the public governance. There is a draft of the assessment of Internal Control System Effectiveness in the practical part of this thesis.
