Title of Doctoral Thesis: Nanofibrous membranes as drug delivery systems Nanofibres are material structures of unique properties that are well-useable in many exciting areas, including medical products. In the field of air filtration, different types of nanofibre filters are produced industrially. On the other hand, in the area of medicated nanofibres, there is still a major barrier for production, and hence the commercial use of nanofibres, especially due to the low weight and required content uniformity of the final products. Electrospinning technology is the most promising in this context, however, no results on the evaluation of nanofibrous products intended for the systemic administration of drugs obtained by certified manufacturing process have been published. In this doctoral thesis the individual steps of the nanofibrous products production are described, experimentally evaluated in compliance with good manufacturing practice and documented with an emphasis on the validation procedures and thus on the desired repeatability of all steps leading to the production of the final product. Polymeric nanofibrous scaffolds reinforced with diamond and ceramic nanoparticles for bone tissue engineering

Three types of nanofibrous scaffolds were prepared by electrospining: (1) poly(lactide-co-glycoside) (PLGA) scaffolds reinforced with 23 wt.% of diamond nanoparticles (DNPs), (2) poly(L-lactide) (PLLA) scaffolds with DNPs in concentration ranging from from 0.4 wt.% to 12.3 wt.%, and (3) PLLA scaffolds with 5 wt.% or 15 wt.% of hydroxyapatite (HAp) nanoparticles. The diameter of the nanofibers ranged between 160 and 729 nm. The nanofibers with nanoparticles were thicker and the void spaces among them were smaller. Mechanical properties of the nanoparticle-loaded scaffolds were better, as demonstrated by a rupture test in scaffolds with DNPs and by a creep behavior test in scaffolds with HAp. On PLGA scaffolds with DNPs, the human osteoblast-like MG-63 cells adhered in similar numbers and grew with similar kinetics as on pure PLGA scaffolds. Human bone marrow mesenchymal stem cells grew faster and reached higher population densities on PLGA-DNP scaffolds. However, on PLLA-based scaffolds, the activity of mitochondrial enzymes and concentration of osteocalcin in MG-63 cells decreased with increasing DNP concentration. On the other hand, the metabolic activity of MG-63 cells and content of osteocalcin in these cells were positively correlated with the HAp concentration in PLLA scaffolds. Thus, PLGA nanofibers with 23 wt% of DNPs and PLLA nanofibers with 5 and particularly 15 wt.% of HAp seem to be promising for bone tissue engineering.
The butoom on the times transformation
My diploma work objective describe history of the development of claps and butooms from primeval age to the present times. I desribe the development of the claps with brief development from the primeval age to 19th. century of the fashion. One part of my diploma work is obout manufacture of the butooms in Czech. In 18th. and 19th. century is typical homework´s manufacture of the butooms in Czech. The another chapters describe these manufacture of the butooms. The chapters are separete in an accordance with materials of the butooms. I describe the manufacture from materials to the butoom. The mentiond obout regions manufacture is at all kind of the butooms. You can find the list about museums of the butooms on Czech in my diploma work. I desribe something about interes of the buttoms at the end my diploma work.
