Constant Current Chronopotentiometry as a Method for Detection of Singlet Oxygen Protein Damage
Ostatná, Veronika ; Vargová, Veronika ; Černocká, Hana ; Paleček, Emil
Proteins are one of the major targets for oxidative damage in the cell.Indirect non-radical oxidation of the protein via formation and subsequent reaction with single oxygen (O-1(2))is one of the major processes. In this work we studied the single oxygen(O-1(2))-mediated oxidation of bovine serum albumin (BSA) by constant current chronopotentiometry in combination with mercury electrode. Our chronopotentiometric data show that photo-oxidized BSA was more susceptible to the electric field-induced denaturation than non-oxidized native BSA. Our method utilizing intrinsic electrochemical signal of proteins provides a sensitive detection methods for minor damages in various proteins.
Voltammetric Detection of Purine Nucleobases on Screen-Printed Graphite Electrodes Modified by Nanocarbon Materials
Hasoň, Stanislav ; Ostatná, Veronika ; Fojta, Miroslav
Recent studies have shown that the electrochemical oxidation of purine nucleobases (adenine and geaoine) at graphite electrodes is strongly dependent on the orientation of graphene sheets. their size and mutual distribution on the electrode surface. Edges and defects of graphene sheets affect the kinetics of oxidation processes, basal planes support the adsorption processes The aim of this study is to determine how model carbon nanomaterials including graphene, single-walled or multi-wailed carbon nanotubes. carbon nanofibres and mesoporous graphene affect the electrochemical oxidation or purine nuclebases on screen-printed carbon electrodes modified by these nanostructures in solution without and in the presence of copper.
Affinity microfluidic chip based on packed microbeads
Novotný, Jakub ; Ostatná, Veronika ; Foret, František
In this project, a plastic device was fabricated using CNC milling in polycarbonate. Chip contains integrated pneumatic valves for flow control and allows for injection and stacking of beaded supports. Intended use of these supports is formation of packed affinity columns suitable for glycoprotein assays.
