Optimization of p53 mutant protein isolation and its DNA binding properties

P53 protein is one of the most important molecules in the human body. It is responsible for several cellular processes such as DNA repair, cell cycle or induction of apoptosis. P53 is also known as “guardian of the genome”. DNA binding ability of p53 is important for the normal cell development and growth. Different mutation of TP53 gene, may cause lost of p53 binding ability andits tumor suppressor function that may lead to the cancer development. The theoretical part of this diploma thesis, is focused on the characterization of main properties, function and mechanism of p53 protein activation and description oflocal secondary DNA structures. The main goal of the experimental part was production of four mutant forms of p53 protein and wild-type p53 protein and studying its binding properties with different local secondary DNA structures. Using Gateway cloning system, four expression vectors were prepared and used for protein production in the bacterial expression system. In total, four mutant and one wild-type p53 protein were prepared and their DNA binding properties were evaluated by Electrophoretic Mobility-Shift Assay The results suggest different DNA-binding properties of wild-type p53 and mutant forms of this protein. All studied mutant proteins lost ability to bind DNA sequence specifically, whereas non- specific interaction with DNA were sustained for three out of four mutant forms. One of the studied mutant proteins showed preference for superhelical form of DNA.
Příprava konstruktů pro izolaci proteinů a jejich testování

Tato práce je zaměřená na popis produkce rekombinantních proteinu. Pro studium dané problematiky byl zvolen protein p53, který je jedním z hlavních nádorových supresorových proteinu. Protein p53 je zodpovědný za regulaci mnoha genů, které mohou kontrolovat buněčný cyklus a replikaci DNA, a je nejčastěji mutovaným genem u nádorových onemocněni člověka. Pro danou prací byly zvolený různé varianty bodových mutaci proteinů p53, které mohou ovlivnit jeho vazebné vlastnosti a funkce. Znalosti mechanizmu působení mutovaných p53 proteinů v buňce je důležité pro efektivní léčbu rakoviny. V teoretické části jsou popsaný vlastnosti proteinu a jednotlivé expresní systémy, metody Gateway klonováni, a metody použité při purifikaci a izolaci proteinů (afinitní chromatografie, SDS-PAGE analýza a Western blot analýza). V experimentální časti jsou popsané postupy přípravy expresních vektoru pomoci Gateway technologie, transformace do buněk a izolace plazmidové DNA. Klonováním byly připraveny tři expresních klony a po jejich transformace do kompetentních buněk byla provedena izolace DNA.
