Metodika péče o umělecky ztvárněnou kulturní krajinu

Cílem metodiky je vytvořeni souborného návodu využitelného při komplexní péči o umělecky ztvárněnou kulturní krajinu za využití aktuálně platné národní legislativy a mezinárodních dokumentů. Metodika je strukturovaná do sedmi kapitol, z nichž některé jsou dále členěny do podkapitol. Na vstupní údaje navazuje obecný úvod vymezující řešenou problematiku. Jádro dokumentu představuje kapitola „Doporučené postupy“. Jednotlivé kapitoly pojednávají o identifikaci kompozice umělecky ztvárněné kulturní krajiny včetně stanovení aktuálních hodnot těchto krajin. Proces péče o toto kulturní dědictví je zasazen do kontextu územního plánování. Zmíněn je fyzický proces obnovy a údržby umělecky ztvárněné kulturní krajiny ve vazbě na již vydané metodické materiály. Následuje důvodová zprava k metodice, závěr a závěrečné kapitoly věnované literárním zdrojům.

Possibilities of Revitalization of the Buďánka Heritage Zone

The bachelor thesis describes the aspects of the heritage conservation in the Czech Republic using a specific example. The aim of the thesis is to clarify the causes of devastation of the settlement Buďánka, a heritage zone. The purpose of the thesis is to find out whether the dismal conditions of the settlement buildings are caused by the strict conditions set by the authorities responsible for monument preservation. The thesis aims at outlining possible procedures in regeneration of similar areas using the example of revitalization of the conservation area for the purposes of citizens´ initiatives. In particular, the thesis makes use of researches of primary information sources, mainly documents, as well as observations and personal interviews with the parties involved.
Analysis of the structure of alkaloids using multidimensional NMR spectroscopy

This Thesis was focused on the structural analysis employing nuclear magnetic resonantion (NMR) of unknown natural substances, which were isolated at the Department of Pharmaceutical Botany and Ecology from the plants Berberis vulgaris L., Fumaria officinalis L. and Nerine bowdenii W. Watson. For this purpose, 1D (1 H NMR, 13 C NMR) and 2D (gCOSY, gHSQC, gHMBC, NOESY, …) NMR spectra were measured and interpreted. Isotopic induced effect was optimized for the determination of the constitutional isomers. To distinguish the possible stereoisomers Mosher method, chiral shift reagent, nuclear Overhauser effect, indirect spin-spin coupling and circular dichroism were used. The structures of forty- five alkaloids were determined including their absolute configuration. Ten of them have not been yet mentioned in the literature before.

Brain Tumor Cells Genome Instability. Correlation of clinial, morphological and molecular-cytogenetic data

Gliomas are brain tumors arising from neuroglia. In most cases astrocytic or oligodendroglial component is the main element of the tumor. Non-random chromosomal abberations are found in tumor cells as was revealed previously. The aim of this study was a fluorescence in-situ hybridisation analysis (FISH) of tissue samples obtained during neurosurgical procedures, determine the frequence of selected chromosomal abberations, further correlation with morphological and clinical data and statistical analysis of the results. During six years 264 tissue samples were gained in which FISH with defined probes was performed. The acquired results were compared with histological analysis and selected clinical data (age, Karnofsky score, extent of resection, overall survival). The whole series was divided into 7 groups by tumor type for further statistical analysis. In every group median and mean survival time was calculated, Kaplan-Meier analysis was focused on influence of selected parameters to overall survival. In some categories Cox regression model was created to achieve a hazard ratio of selected parameters. The significance of the S100B protein in patients with an injury to the central nervous system

Inroduction: The S100B protein is a small dimeric protein which belongs to a group of calcium binding proteins. It is found in astroglial cells in the central nervous system (CNS). The S100B protein is detected, at very low concentrations, in fat tissue, muscle and bone marrow. We can measure this marker in the cerebrospinal fluid, in serum and in urine. The commercially available assay measures the sum of two different dimers (the S100A1B and S100BB) which collectively are called the S100B protein.The level of this marker increases significantly in patiens with CNS damage. Studies of patients with isolated head trauma have demostrated a correlation between serum marker level, neurological findings and outcome. In patients with polytrauma there is a problem to evaluating the initial S100B protein level with regards to CNS damage, because the S100B protein level can be affected by extracerebral sources of the S100B protein. Aim of study: The aim of our study was to evaluate the significance of the S100B protein in patiens with either isolated head trauma or polytrauma and to evaluate the significance of the initial level and the significance of dynamic monitoring of the S100B protein levels. Use of NMR in structural analysis

This Diploma work is consecrated to structure determination of unknown substances. The Department of Pharmaceutical Botany and Ecology, Faculty of Pharmacy in Hradec Králové isolated number of alkaloids from three plants: Zanthoxylum nitidum (Roxb.) DC (Rutaceae), Corydalis cava (L.) Schw. et Koerte (Fumariaceae) and Zephyranthes robusta Baker (Amaryllidaceae). Some of thath alkaloids were analyzed in Laboratory of structure and interactions. The goal of this project was to determinate the structure of three unknown product, using NMR. NMR experiments (1 H NMR, 13 C NMR, ghmqfcops, ghsqc, ghmbc) were necessary to reveal the structure of unknown compounds.
The correlation of intracranial pressure and next investigative parameters at the cerebral contusion

Patients with a brain injury must overcome a central neurological insult and a systematic metabolic response. The systematic response includes hypermetabolism, hypercatabolism, altered vascular permeability, increased hormone and cytokine release, altered gastric emptying, altered mineral metabolism and altered immune status. This response may initiate mechanisms which lead to secondary brain injury and may adversely affect the function of other organs. Last two decades have improved our knowledge of pathophysiology in patients with primary and secondary brain injury including traumatic contusions. In a moment of injury, primary brain injury which is traumatic contusion can not be therapeutically prevented it can only have a preventive effect. Secondary brain injury can be therapeutically affected although both types of injuries have common pathophysiological signs of disturbed metabolism on cells and subcellular level. Research methods based on molecular biology opens a new zone of research reaction of central neurological system for trauma or ischemia with effort to get involved immediately at the beginning of developing adverse pathophysiological cascade leading to a death of neurotic cell.
