Music Therapy and use for disabled children

The thesis contains a theoretical and a practical part. The theoretical part deals with a concept and a definition of music therapy, history, aims, methods of music therapy and also organisations where music therapy is done. The thesis deals with connection of music therapy and children with disabilities. The topic of disability is described in several categories. It lists the types of disabilities, connection between disabled children and music therapy. It also deals with a funcion of a music therapist and it inroduces different therapists who work with us. The practical part contains my personal project, which consists of 3 lesson in length of 45 minutes each. I took advantage of the fact that in my surrounding there is a disabled child and I organized lessons of music therapy with him.
Physico-Chemical Properties of Drugs

Thesis title: Physico-Chemical properties of drugs This thesis deals with measuring dissociation constants (the pKa value) and the lipophilicity of nitrogenous heterocycle derivatives produced at the Faculty of Pharmacy in Hradec Králové. Two methods were used to determine the pK value: potentiometric titration and spectrophotometry. Potentiometric titration almost invariably produced relatively exact results, however the results of pK value obtained by spectrophotometry were sometimes difficult to replicate. Dissociation constants were compared on the basis of the method applied, and also with the theoretically determined dissociation constant calculated by means of the ChemDraw Ultra 12.0 software. The results obtained by the two experimental methods turned out to be very similar, corresponding to the theoretical pK value. Lipophilicity is another essential property of drugs tested in the preclinical stage. It specifies the aqueous or lipophilic phase affinity of a drug, substantially affecting the drug's ADMET in organism. PharmDr. Veronika Nováková, Ph.D. Student: Kateřina Pekařová Title of Thesis: Cationic surface active compounds and evaluation of its properties Quaternary ammonium salts are used as surfactants in many branches of the food, pharmaceutical and chemical industries. The basic properties of these substances were studied in the thesis. The critical micellar concentration of the homologous series of five substances was determined using the conductometric method. The theory of declining critical micellar concentration with the increasing length of the side alkyl chain has been verified. The hydrolytic activity of two commonly used cationic surfactants benzalconium chloride and benzoxonium chloride for the decomposition of the organophosphorus pesticide fenitrothion was tested. For these two substances, the effect of pH and temperature changes was evaluated. The influence of tenside concentration was also monitored. Benzoxonium chloride at 50 řC and pH 11was found as the most effective hydrolytic catalyst for the decomposition of the organophosphorus pesticide fenitrothione.
Determination of the fatty acids spectrum in patients undergoing treatment with taxanes

Title of Diploma Thesis: Determination of the fatty acids spectrum in patients undergoing treatment with taxanes The diploma thesis deals with the determination of fatty acids spectrum in erythrocytes in patients treated with taxanes. The theoretical part describes general features of fatty acids, their synthesis and degradation. Then it is focused on the topic of breast cancer, taxane treatment and negative side effects associated with taxane treatment, especially polyneuropathy. The gas chromatography, which is commonly used in practice, is also described in this part. The experimental part is divided into three parts. The first part describes working process, chromatographic analysis and evaluation of this analysis. In the second part there is comparing of spectrum of fatty acids of patients without and with polyneroupathy. And in the last part there is a comparing of spectrum of fatty acids of patients before taxane treatment, closely after treatment and about month after the treatment. MicroRNA expression in glucocorticoid-treated patients with systemic autoimmune

Rheumatoid arthritis is the most common joint disease of autoimmune origin. It is accompanied by inflammatory conditions that lead to irreversible changes in the joints, their deformities ending with permanent disability. Treatment of the disease involves routine regimens, surgical, as well as pharmacological treatment, which is necessary for advanced forms. Glucocorticoids play an important role in the therapeutic intervention in the course and progression of the disease. In spite of their anti-inflammatory effect, which is a key to improving the condition of the patient, they have a number of side effects in the long term- use. In this study, we have focused on the impact of these drugs on microRNA expression changes in arthritic patients treated with pulsed doses of glucocorticoids. MicroRNAs are nowadays widely studied due to their possible use as biomarkers in monitoring disease progression and the effect of treatment. MiRNA expression analysis was performed by quantitative real-time PCR array of 754 miRNAs with reverse transcription using stem-loop primers that allow amplification of short sequences that microRNAs are. The influence of anesthesia on the degree of DNA oxidative damage

Background: Oxidative damage is one of the most frequent types of cell components damage leading to oxidation of lipids, proteins and the molecule of DNA. As a consequence, there is a higher occurrence of several pathologies such as atherosclerosis, neurodegenerative diseases, cancer; or diabetes. In our study, influence of whole body anesthesia during minor surgery on the level of DNA damage was examined using comet assay technique. Methods: The basic principle of this method is fixing the cells (lymphocytes) in agarose, their lysis for the removal of membranes, incubation with the specific enzymes and electrophoresis of the released cell nuclei. During the electrophoresis, free low-molecular weight and negatively charged fragments of DNA move towards anode which causes the formation of the typical comet cell shape. Finally, the gels are stained by ethidium bromide (DNA intercalating dye) and visualized. Results: We have observed single strand breakages (SSBs) and, with the use of modified assay using specific enzymes for detection of specific lesions, also oxidized purines and pyrimidines. Compound adjectives in baroque period and their development in czech national revival

Topic of this dissertation are compound adjective, their development and usage during two historical periods. Compounding is on the edge of czech word-formation methods. This method of creating new lexical units has spread the most during Czech National Revival as one way of expanding czech lexicon. Authors used mostly semantic fullness, economy of expression and uncommonness of compounds in both specialized and artistical texts and in both prosaical and poetical texts. Compounding already started to evolve more in Baroque period, compounds were used for their communicative value and unusualness. Basic classification is adopted from monograph Česká kompozita diachronně by Dušan Šlosar (1999), secondarily from monograph Tvoření slov skládáním by Ivana Bozděchová (1994). The aim of the work is to capture development of structures used in adjective compounds formation during these two periods, also to examine their occurences, functions and types, how they developed and changed during the periods of Baroque and Czech National Revival. The work contributes to analytical examination of the historical development of word-formation level of czech language, specifically of the method of compounding.
Determination of fatty acids in human tissues

Title of Diploma Thesis: Determination of fatty acids in human tissues The fatty acids are non-negligible component of lipids as one of the basic nutrients. This thesis in its theoretical part presents above all the group of polyunsaturated fatty acids (PUFA), which are important structural units of the cell membranes, they are also the precursors to several significant biologically active substances. In the human organism PUFA participate in many physiological and pathological processes this way, where they cause the large spectrum of actions. Onwards the thesis describes the gas chromatography method that is used in chemical analysis of lipids for the fatty acid determination as the gold standard. Experimentally, the fatty acid representation was determined in the blood, muscle, heart, liver and kidney among 26 cadavers divided into two groups according to the presence/absence of inflammatory process at the time of death. Salt intake by 5 years-old and influence on their blood pressure

Thesis deals with problems of excessive salt intake in children. The goal of thesis was to monitor recieved amount of salt in five years old children. The second goal was to find out what is the effect of salt on their blood pressure. Theoretical part deals about physiology of sodium in human body, its balance and metabolism. Further are described negative impacts on health of individual, especially in children with excessive salt intake in the diet. Thesis points out what foods to avoid, or at least take a limited quantity. It also focus on the possibilities of reducing salt in food and how is salt used in food industry. In the next section I deal with children food and especially what kind of factors influence an individual's lifestyle and habits. In the practical part I deal with research of fifteen respondents. Research group is divided into three groups. The first group consists of respondents diet in kindergarten, the second group are children in orphanages and the last group are children, which eat at home. I used qualitative strategy for processing research, where I evaluated seven day diets of children in orphanages and in home environment. At the last set I evaluated their five-day meals and only snack, lunch and afternoon snack that children received in kindergarten. All forms for recording diet I evaluated in program "Nutriservis Professional". I also measured blood pressure to all respondents with digital barometer. I transferred measured blood pressure to the percentile values, which are intended for children. I compared the final amounts of received salt with the reference values determined by age. The results showed, that not a single respondent comply the recommended daily amount of salt. Correlation of blood pressure with salt intake is among respondents different.
