Cíl práce: Cílem práce bylo vytvořit přehled možných lékových interakcí u léčiv používaných v gynekologické a urologické praxi. Metodika: Z teoretického úvodu byla vybrána, pomocí webové databáze SÚKLu, léčiva registrovaná v ČR k 1. 1. 2015. Informace o lékových interakcích byly získávány a porovnávány z databází Vademecum infopharm a Micromedex. Z databáze Micromedex byly vybírány lékové interakce s kvalitou dokumentace "excellent" nebo "good". K vytvoření přehledu dostupných informací o vybraných lékových interakcí byla využita databáze PubMed (www.pubmed.gov) se zadáváním klíčových slov "základní léčivo", "drug interaction" a "interakční léčivo". Pro doplnění informací bylo také čerpáno z časopisů Praktické lékárenství, Klinická farmakologie a farmacie. V případě nenalezení relevantních informací jsem čerpal ze souhrnné informace (SPC) dílčích léčivých přípravků. Výsledky: Výsledky byly sepsány jako rešeršní...
The effect of hydrolysis on chemical and physical properties of PAN hydrogels

Presented diploma thesis describes the preparation of polyacrylonitrile hydrogels and characterization of their physical and chemical properties. Theoretical part sums up basic knowledge about hydrogels and polyacrylonitrile . It also deals with processing of polyacrylonitrile into reactive form and its alkaline hydrolysis which leads to multi-blocks copolymer able to create a three-dimensional network (gel). The experimental part presents a characterization of polyacrylonitrile hydrogels (HYPAN) prepared by alkaline catalyzed hydrolysis of a reactive form of polyacrylonitrile (aquagel). The aquagel was produced by dissolving followed by an extrusion. The final fiber was processed into pellets, which underwent alkaline hydrolysis by sodium hydroxide solution. Hydrolysis has carried out under various conditions (temperature, NaOH concentration and reaction time) to obtain products with a different degree of conversion of -CN groups. The hydrophobic blocks and hydrophilic blocks made by hydrolysis can form a 3D network of various properties depending on the ratio between blocks. The hydrogels were prepared from hydrolyzates in an alkaline environment. Some of the hydrolyzates have not created hydrogels but only suspensions (indecent degree of conversion of -CN groups). Therefore, rheological and optical properties were characterized only at samples forming hydrogels. By optimization of reaction conditions, multi-blocks copolymers able to create a gel with suitable physical properties (viscosity, transparency) were prepared with the potentional used in biomedicine e. g. as implants in ophthalmology.

Příprava a charakterizace kopolymerů na bázi poly-3-hydroxybutyrátu

Tato práce se zabývá přípravou kopolymerů na bázi poly-3-hydroxybutyrátu (PHB). Snahou je dosáhnout materiálu, kde by byl obsah polymeru PHB co nejvyšší, zároveň ale obsah krystalické fáze co nejmenší. Nízkomolekulární PHB s koncovými hydroxylovými skupinami, použitý při syntéze kopolymerů, byl připraven alkoholýzou PHB polymeru 1,4-butandiolem. Jako druhý reagující polymer v této práci byl vybrán polyetylenglykol (PEG) v různých molárních hmotnostech a poměrech. Další polymer pro kopolymeraci s PHB byl zvolen poly(butylen adipát-co-tereftalát) (PBAT) a jako netoxické spojovací činidlo byl použit hexametylendiisokyanát (HDI). Z těchto látek byly v roztoku připraveny blokové kopolymery, které jsou tvořeny z měkkých a tvrdých segmentů. Tvrdé segmenty zde představuje PHB a jeho krystalická struktura, měkké segmenty tvoří řetězce PEG, případně PBAT. U připravených kopolymerů byl zkoumán vliv přítomnosti měkkých segmentů různé délky a poměru na celkové vlastnosti. Získané materiály byly charakterizovány pomocí termogravimetrické analýzy (TGA), diferenční kompenzační kolorimetrie (DSC) a infračervené spektroskopie (FTIR). Molekulová hmotnost byla zjištěná pomocí gelové permeační chromatografie (GPC).
Bioresorbable polyurethanes with controlled mechanical properties

Presented diploma thesis deals with preparation of bioresorbable polyurethanes (PUR) and their characterization. The theoretical part describes the feedstocks used for the PUR preparation and summarizes the knowledge about PUR used in medical applications. Experimental part presents characterization of bioresorbable PUR films prepared by reactive casting in one step without the use of organic solvents. The absence of solvents represents a great advantage due to their toxicity and subsequent removal from the resulting product. The synthesis of PUR was conducted under an inert atmosphere by polyaddition reaction of hydrophobic poly(e-caprolactone) (PCL) and hydrophilic poly(ethylene glycol) (PEG) with hexamethylene diisocyanate (HDI). Synthesis under an inert atmosphere was necessary due to a side reaction of isocyanate with atmospheric humidity, which causes the formation of porous films instead of continuous ones. Prepared PUR films were characterized for different PCL/PEG ratios and different isocyanate index (ratio of NCO/OH reacted groups). PUR (isocyanate index 1.05) with PCL content 90 wt. % and higher demonstrated tought behavior in mechanical tests. Increase of isocyanate index and decrease of PCL content under 90 wt. % caused fragile sample behavior. The reason of such behavior was the different ability of PCL to crystallize in the structure of the polyurethane network. Increasing the PEG content has improved the ability of PUR films to absorb water and enhanced the rate of hydrolytic degradation. By adjusting the PCL/PEG ratio and the amount of isocyanate, solvent free bioresorbable PUR with suitable mechanical (flexibility, toughness) and physical properties (swelling, degradation) can be obtained. Prepared PUR films could be used in biomedicine e.g. as vascular grafts.
Přímá syntéza vysokomolekulárních polymerů kyseliny mléčné

Diplomová práce se zabývá problematikou přímé syntézy polymerů kyseliny mléčné. Teoretická část je zaměřena na výrobu kyseliny mléčné jako monomeru, a to jak přírodní, tak i syntetickou cestou, jejich výhodami a vlastnostmi. Dále se teoretická část zaměřuje na syntézy poly(mléčné kyseliny) (PLA) z laktidu a přímou polykondenzaci z kyseliny mléčné a pojednává o vlivu katalyzátorů, kokatalyzátorů a extendérů řetězce. Experimentální část práce je zaměřena na syntézu polymerů a kopolymerů PLA, kde zkoumá vhodné katalyzátory, reakční prostředí pro azeotropickou dehydrataci a vliv kokatalyzátoru pro přípravu požadovaného produktu. V závěru se experimentální část zabývá analýzou připravených polymerů PLA pomocí termických a analytických metod.
Modification of Biodegradable Polyurethanes by Biologically Active Substances

Presented dissertation thesis is focused on novel preparation of biodegradable polyurethanes (PUs) and their modification by biologically active cellulose nanocrystals. Literary review deals with current state of bioresorbable PUs used in tissue engineering. Examples of prepared PU elastomers, scaffolds and injectable PUs, together with biodegradation pathways to non-toxic products are summarized. The last part of the literary review is targeting on nanocellulose, which has gained much attention for the use as biomedical material due to its remarkable physical (high specific surface area, mechanical reinforcement) and biological (biocompatibility, biodegradability and low toxicity) properties. Experimental part presents characterization of biodegradable amphiphilic polyurethane films (bio-PUs) synthesized by solvent free polyaddition reaction of hydrophilic poly(ethylene glycol) (PEG) and hydrophobic poly(e-caprolactone) (PCL) as macrodiols with hexamethylene diisocyanate. Prepared bio-PUs were characterized on one hand by means of different PEG/PCL ratio and on the other hand by changing the isocyanate ratio between NCO/OH groups. Abrupt enhancement of mechanical properties was observed when PEG/PCL weight ratio was equal to or less than 20/80 and was ascribed to the PCL ability to form crystalline domains. The increasing amount of PEG promoted the ability of bio-PUs to absorb water and enhance the rate of hydrolytic degradation. Whereas, reducing the ability of bio-PUs to absorb water and prolonged time of hydrolytic degradation was achieved with increasing the crosslink density by enhancing the isocyanate ratio. The last part deals with novel solvent free preparation of nanocomposite utilizing bio-PU as a matrix and cellulose nanocrystals either neat or surface grafted by PEG. Structural analysis demonstrated that the presence of rod-like nanoparticles causes the immobilization of the PU chains in matrix resulting in increased stiffness and rigidity of bio-PU/cellulose nanocomposite. By adjusting the PEG/PCL ratio, the amount of isocyanate or the presence of nanofiller, the novel bio-PU material with desirable mechanical (toughness, flexibility) and physical (swelling, degradation) properties can be obtained. Prepared solvent free bio-PUs may advantageously be used in regenerative medicine for soft tissue regeneration (e.g. as vascular grafts).
Biodegradable polyurethanes based on poly(ethylene glycol)

Presented diploma thesis deals with synthesis of biodegradable polyurethanes (bio-PUs) based on poly(ethylene glycol) (PEG) and poly(e-caprolactone) (PCL) with potential use in medicine. The aim of the work was to develop the methodology for synthesis of elastomeric polyurethane that could be formed into scaffold applicable in tissue engineering for regenerative human medicine. The theoretical part summarizes the knowledge of various kinds of materials which are possible to use for intended application. Scaffold forming techniques, their biocompatibility and characterization of properties of resulting materials are involved. The experimental part is focused on the development of suitable methodology to prepare functional samples based on PEG, PCL, hexamethylene diisocyanate (HMDI) and stannous octoate as the catalyst. Effect of bio-PUs composition (mainly different amount and molecular weight of PEG) on the swelling and hydrolytic stability was investigated together with testing mechanical properties, monitoring the net formation and the degree of conversion. The morphology of prepared samples was analyzed by optical microscopy, chemical composition was confirmed by infrared spectroscopy and the thermal properties were measured by differential scanning calorimetry. As for methodology development, it was found that it is necessary to degas all feedstocks before synthesis, otherwise non-well polymerized samples with different pore size have been obtained. Synthesis of bio-PU was realized under the nitrogen atmosphere in two steps. First, both polyols (PEG and PCL) were homogenized on the vacuum line at 130 °C followed by the addition of HMDI in the second step at 65 °C in glove-bag to obtain bio-PU samples. Finally samples were cured at the mould for 48 hours at 65 °C in oven. Based on the physical conditions of samples preparations, flexible bio-PU from white films through cloudy (with visible phase separation) to transparent films (clear) were obtained. Phase separation was confirmed by optical microscopy showing typical spherulite structure of crystalline phase (PCL) in amorphous matrix of PEG. The monitoring of net formation confirmed maximal conversion of polymerization (96 %) already after 4 hours of curing in the oven. Mechanical properties testing showed that important influence to rupture strength has isocyanate index (NCO/OH ratio). Different molecular weight of PEG showed influence to tensile properties as the specimens were completely cross-linked. Characterization of swelling exhibited increase the water uptake of samples with growing molecular weight of PEG (from 28 up to 58 wt.%). As well as, the bio- PU samples having higher molecular weight of PEG degraded faster in water at 37 °C. In presented diploma thesis was confirmed that it is possible to control the hydrolytic stability of obtained biodegradable polyurethane elastomers by the amount and the molecular weight of PEG.
Modification of polyurethane by biodegradable poly(hydroxybutyrate)

The aim of the presented bachelor thesis was the synthesis of elastomeric polyurethane filled by biodegradable polyhydroxybutyrate. The morphology of samples was analyzed by scanning electron microscopy and mechanical properties were tested by tensile measurement. Moreover, the work deals with prediction of mechanical properties based on rubber elasticity theory. It was found that prepared specimens have a character of particle – reinforced composite materials. Corresponding to divination, the biodegradable filler content enhancement in polyurethane matrix leaded to an increase of the elastic modulus. Prediction of stress-strain curves, which was done by SEC.exe software, showed for real behavior of stress-strain curve having unreal values of network density of polymer chains. While real values of chains network density were used in simulation, stress-strain data were higher than the measured ones.
