Mesenchymal stromal cells and biological scaffolds for neural tissue regeneration

Despite tremendous progress in medicine, injuries of the adult central neural system remain without satisfactory solution. Regenerative medicine employs tissue engineering, cellular therapies, medical devices, gene therapy, or growth factors with the aim to bridge the lesion, re-establish lost connections and enhance endogenous repair in order to restore neural function. The aim of my thesis was to evaluate therapeutic potential of two approaches, transplantation of human mesenchymal stromal cells (hMSCs) and biological scaffolds derived from extracellular matrix (ECM) for neural regeneration, particularly in models of spinal cord injury (SCI). First, hMSCs from various sources - bone marrow (BM), adipose tissue (AT) and Wharton's jelly (WJ) - were isolated and characterized in vitro. All cell types met the minimal criteria for MSC phenotype and displayed similar properties in terms of their surface marker expression, differentiation potential, migratory capacity, and secretion of cytokines and growth factors. On the other hand, the cell yield from WJ and AT was significantly higher, and MSCs isolated from these tissues proliferated better than from BM. Clinical applications of stem cells for the treatment of neurodegenerative diseases of CNS

Stem cells have a huge therapeutic potential due to their ability to differentiate in multiple tissues. They could be used for neurodegenerative diseases treatment, which are typical for loss of specific groups of neurons, progressive course and lack of effective treatment due to their complicated pathophysiology, only therapies for elongation and simplification of patients' life are available. This thesis summarizes results of completed clinical studies and informs about ongoing studies, in which stem cell treatments are used for selected neurodegenerative diseases. Stem cell therapy for multiple sclerosis and amyotrophic lateral sclerosis have a long history, some of the studies has proven therapeutic efficiency of stem cells. We don't know much about effect of stem cell treatment for patients with Alzheimer's and Parkinson's diseases, because first clinical studies were finished recently. But all clinical trials have proven safety of stem cell treatment. Replacement of damaged neurons haven't been reached yet, just protection of remaining neurons by neurotrophic and immunomodulatory factors secreted by stem cells. Methodology of using low-temperature plasma in veterinary treatment

The aim of the methodology is application of low-temperature plasma in veterinary treatment. Low-temperature plasma (NTP) has non-specific antimicrobial effects and it can be safely applied to living tissues without damaging them. The methodology presents the solution of dermatological problems with potential bacterial, fungal or yeast contamination. The method can also be used for antimicrobial decontamination and for the promotion of healing of chronic and non-healing wounds.

The article discusses itemized dissertation on “Open space of prefab housing estates in Brno”. The aim of the thesis is to analyse and describe the unused premises of panel house estates in Brno and to create and valorise usable system of data and information (regarding chronological sequence). The obtained data and the entire dissertation can be used in planning and designing interventions in prefab housing estates and in other structures of residential areas, respectively, as part of a multi-criteria analysis of particular housing estates.

Immunogenicity of stem cells and their derivatives Nikola Doležalová Abstract Stem cells (SCs) have the potential to be used in regenerative medicine on the basis of their differentiation capacity and promising immunological properties, including low expression of histocompatibility antigens and costimulatory molecules, or secretion of suppressive cytokines. Their immunogenicity has often been ignored in the past but it is becoming clear that rejection of genetically incompatible SCs represents a very common issue. At present, SCs are extensively studied from the immunological point of view, since it represents a critical aspect of the safety of SC therapy. This thesis presents an overview of current knowledge about immunogenicity of SCs and their derivatives, including both pluripotent SCs (embryonic and induced pluripotent SCs) and adult SCs (mesenchymal, limbal, neural, haematopoietic and umbilical cord blood SCs). The expression of immunologically relevant molecules on their surface and interaction with the immune cells in vitro and in vivo will be discussed, together with suggestions for overcoming the immunological barriers for transplantation. Detailed analysis of these aspects necessarily has to precede the safe clinical translation of SC therapies.
Treatment of spinal cord injury by transplantation different types of stem cells

Spinal cord injury (SCI) is complicated injury with serious socioeconomic consequences for the patient and his whole family. Big difficulty cause also extremely high living expenses for the patient with this type of injury. That's why there is a need for therapeutic methods which would help patients after SCI to recover the lost functions and be able at least partially to return to their normal life. Different therapeutic methods are being used for SCI treatment. In this study we used four various types of stem cells: human bone marrow stem cells (hBM-MSCs), human umbilical cord mesenchymal stem cells (hUC-MSCs), neural precursors derived from induced pluripotent stem cells (iPS-NPs) and neural stem cell line derived from human fetal spinal cord tissue (SPC-01). These cells have been transplanted intrathecally or intraspinally 7 days after induction of the experimental model of SCI in the rat. We studied expressions of genes related to neurogenesis, growth factors and inflammation 10 and 28 days after SCI. Our analysis showed significant changes in gene expression 10 days after SCI. Variscan igneous activity at the Bohemicum/moldanubicum boundary

Variscan dike swarms associated with the Central Bohemian Plutonic Complex (CBPC) at the boundary between the Teplá-Barrandian and Moldanubian Units of the Bohemian Massif represent one of the most interesting geological phenomena. Frequency of dykes and their chemical variability do not have any comparable analogy in the whole European Variscides. This work is focused on the study of dyke rocks in the NE periphery of CBPC in geologically very complicated area with intrusions of predominantly deformed granitoids, contact metamorphosed sediments and magmatic rocks of "Islet Zone" with different protolith ages (forming remnants of the original roof of CBPC), deformed basic rocks of uncertain origin and age. The area extends up to the western boundary of the northernmost part of the Moldanubian high-grade metamorphic complex, the boundary itself being also tectonically problematic. Several localities with dyke rocks under study are situated in the area east of Senohraby (SE of Prague), on the northern side (right coast) of the Sázava river, and extend up to the area of Stříbrná Skalice. This area is rich in dykes of gabbro to diorite porphyry accompanied in some places with tonalite (rarely quartz diorite) porphyry and more rarely with amphibole lamprophyres (spessartite). Mesenchymal stem cells for treatment of spinal cord injury

Poranenie miechy a jeho terapia pomocou mezenchymálnych kmeňových buniek Abstract Patients with serious spinal cord injury (SCI) have a little chance of a full return to their original life. This kind of injuries leaves lifelong consequences as on the patients themselves, as well as their surroundings and family. Considerable financial resources are, worldwide, granted for reintegratation of these patients into society. This work focuses on the description of spinal cord injury, its complexity and the possibility of treatment by new therapeutic methods of mesenchymal cells. The first section discusses the severity of spinal cord injury and its pathophysiology with the performance of subsequent clinical phases. The second part is dedicated to new treatment methods and approaches, with an emphasis on cell therapy. The third part presents treatment method using mesenchymal stem cells (MSCs), which immunomodulatory and anti-inflammatory properties, such as their ability to migration to the injured area and the possibility of genetic modification, made them a great candidates for the treatment of serious injuries. Targeted differentiation of mesenchymal stem cells and their clinical application

The goal of this work is to point out possibilities of neuronal differentiation of mesenchymal stem cells and their application for clinical purposes, primary for repairing and regeneration of central nervous system tissues. Because this system works as a control center for functional features of the whole body, and treating this injuries and degenerations often bring many problems and obstacles, the possibility of using autologous cells for a transplantation or inducer of the natural regenerative properties of tissues is worth deeper research. This work progressively focuses on basic characteristics of stem cells and their differentiation potential, characterizing further mesenchymal stem cells together with possibilities of their isolation and cultivation. The main part of the text is formed by studies and methods used for targeted differentiation of mesenchymal stem cells and attempts of their transdifferentiation into neural cell line, together with present and possible future application of these cells in central nervous system therapies.
Completion of the area Údolní 53 for the Faculty of Architecture and Faculty of Fine Arts

It is necessary to ask the question of the relationship and the corresponding forms of buildings of the faculty of architecture and faculty of fine arts with the public. Due to the efforts of the faculties to open up to the public, I find it undesirable to place new objects in the courtyard and hide behind the walls of historic buildings. Much more meaningful is the use of existing buildings at the boundary of the plot. It is important to protect our history, but the protection of existing buildings should not get to the point where it will defend the presence, be on the same level, and thus ensure its quality and together to shape the future. The newly allocated creative use of the original barracks has greater demands on space, whether capacity or its generosity. Therefore, it is necessary to customize the objects for the current function. The superstructure will help keep faculties in one building. An advantage is also the connection to existing networks or vertical communications. The proposed minimalist intervention is in contrast to the historic building, making it stand out, while the verticalism of the superstructure raises the object of the original barracks to the desired institution of the Faculty of Architecture and the Faculty of Fine Arts.
