Biosystematic study of the Carlina vulgaris complex in Central Europe based on molecular and morphometric approaches

The Carlina vulgaris complex in central Europe includes several lineages defined by their ecology, morphology and distribution. This diploma thesis is focused on relationships between the taxa recognized in the Czech Republic, namely Carlina vulgaris subsp. vulgaris, C. biebersteinii subsp. biebersteinii, C. biebersteinii subsp. brevibracteata and C. biebersteinii subsp. sudetica. Molecular analysis revealed two genetically defined groups. One includes samples from relict populations in western Bohemia and from high mountains classified as C. biebersteinii subsp. biebersteinii and C. biebersteinii subsp. sudetica. The other is represented by plants classified as C. vulgaris and C. biebersteinii subsp. brevibracteata. This genetic differentiation was also confirmed by morphometric analysis. However, relationships within these two groups remain unclear. The Czech populations of Carlina biebersteinii subsp. biebersteinii as well as of C. biebersteinii subsp. sudetica are closely related to the mountain populations in the Alps and Carpathians. Their occurrence in the Czech Republic is relict and they should be in focus of nature conservation. However, the separate taxonomic position of the claimed endemic C. biebersteinii subsp. sudetica is probably unjustified.
Application of Hyb-Seq method for reconstruction of reticulate infrageneric phylogeny: example from polyploid genus Curcuma L. (Zingiberaceae)

This master thesis focuses on the phylogeny of hybridogenous and polyploid genus Curcuma from family Zingiberaceae using Next-Generation Sequencing data from hundreds to thousands nuclear loci. This approach seems to be better than widely used cpDNA and ITS sequencing especially in the case of hybridogenous and polyploid groups. Data for phylogeny reconstruction were generated using Hybridization-based sequencing (Hyb-Seq) method which combines target enrichment and genome skimming strategies. Data analysis was performed primarily using pipeline HybPhyloMaker especially created for Hyb-Seq data analysis. Twenty-seven species from the genus Curcuma and three outgroup species were sequenced in this work. Phylogenetic trees based on all 1 154 and 811 selected nuclear low- copy genes show high support values of all nodes which is in contrast to plastome and rDNA phylogeny with lower support values in some nodes and incongruences in topology compared to low-copy genes phylogeny. Phylogenetic networks inferred from low-copy genes, lineage movement analysis and monophyly tests agree with published hypotheses of interlineage hybrid origin of three species - C. vamana, C. myanmarensis and C. roscoeana. Systematics and phylogeography of diploid lineages of Arabidopsis arenosa complex

The Arabidopsis arenosa species complex is closely related to the model species Arabidopsis thaliana. Distribution range of the diploid lineage covers most parts of Central and South-eastern Europe with a remarkable diversity of ecological niches from sea level to alpine peaks. There is still little known about genetic relationships among the diploid populations of A. arenosa. The evolutionary history of the diploid cytotype across its entire range was explored by using 14 nuclear microsatellite loci. Five lineages occupying biogeographically distinct regions were identified in Arabidopsis arenosa complex. These lineages were labelled as Western Carpathians, Eastern Carpathians, Pannonian Lowland, Dinaric Alps and Baltic Shore (according to geographical regions where these lineages occur). Molecular phylogeny and evolutionary trends in Hieracium (Asteraceae, Lactuceae)

The hawkweed subgenus Hieracium s. str. is notoriously known for its extreme morphological variability and variation in ploidy levels that is associated with differences in modes of reproduction. Extensive past hybridization is supposed for the subgenus, but recent hybridization was evidenced only in few cases. The subgenus attracts the attention of botanists already for more than a century. Therefore the species diversity is largely examined and the taxonomy of the subgenus is well elaborated, although several contradictory taxonomic concepts exist. However the relationships among the species are unknown and haven't been studied yet. The investigation of these relationships from a phylogenetic perspective using molecular approaches was the main aim of the presented thesis. Basic species (both diploid and polyploid), representing morphologically unique taxa, that are supposed to be the basic evolutionary units of the subgenus were studied. The sequences of two intergenic spacers of the cpDNA (trnT-trnL and trnV-ndhC) and the external transcribed spacer of the nuclear ribosomal DNA (nrDNA ETS) were analyzed.
