Legal position and activities of the Czech National Bank
Král, Vladimír ; Vondráčková, Pavlína (advisor) ; Vybíral, Roman (referee)
Central banks world-widely are institutions of crucial importance for economics of their countries, especially during the years of financial instability, in some cases having also an influence to other economies. The thesis deals with the topic of Czech National Bank (as the central bank of the Czech Republic), its legal position within the Czech legal system and its activities with regards to dynamic development in this area in recent years. The thesis is divided into chapters accordingly to its name and the subject matter. Introduction aims to introduce into topic, briefly describes the structure of the thesis. Introduction is followed by two main chapters - "Legal position of the CNB" and "Activities of the CNB", respectively. These chapters are composed of fractional subchapters and more fractional parts. Thesis is ended by conclusion. Second chapter analyses constitutional provisions relating to Czech National Bank and statutory provision included in Act No. 6/1993 Coll., on the Czech National Bank, with references to respective provisions of law of the European Union. It concerns also two significant rulings of Constitutional court related to the CNB and involves thoughts of some rules currently applied. Special attention is dedicated to the legal conception of CNB's independent position in relation...

Bioartificial liver in the treatment of experimental acute hepatic failure in a large laboratory animal Aim: To evaluate bioartificial liver (BAL) in the treatment of acute liver failure in the experiment. Methods: 10 minipigs weight 25 - 30 kilograms with experimental acute liver failure (ALF) by surgical devascularization with portocaval by-pass were treated by BAL. The biochemical and hemodynamical parameters of ALF including intracranial pressure (ICP) were measured. The results were compared to control group without BAL treatment. Results: To compare plasma bilirubin there was statistically significant difference between experimental and control groups in 6th and 9th hours to favourite BAL group of minipigs (20,86 vs. 10,60, 22.29 vs. 11,75) and in the 12th hour (37,4 vs. 12,88). The value of ICP in both groups was not significant. Conclusion: The functionality of BAL on our own surgical model of ALF was proven. Initial 85 % viability of fresh isolated porcine hepatocytes and their survival in the time of BAL treatment was reached. Bilirubin plasma concentration has significantly changed in comparison in both groups. ICP values were not changed significantly. Key words: acute liver failure - minipig - bioartificial liver - BAL - Bioreactor - hepatocytes - intracranial pressure

77.. SSUUMMMMAARRYY During the dissertation I have managed to prepare various compounds of phthalocyanine (Pc) structures and measure their production of singlet oxygen. The synthesis of Pc is not overly problematic, however complications arise during purification. Phthalocyanines are known for their instability in visible light spectrum and their aggregation in more concentrated solutions. The singlet oxygen quantum yield is usually low at the aggregates, thus further work is limited. The aim of my work was to create new Pcs with both increased solubility and functional groups suitable for conjugation with biomolecules. First experiments were performed with silicon complex of Pc (SiPc). However, synthesis of axialy substituted SiPc was not successful. Similar compounds with peripheral alkylsulphanyl substitution were also not obtained. These compounds disintegrated during synthesis. Another method that I have tried to insert a central aluminium atom into a Pc platform that was soluble into common organic solvents. Not all peripheral substituents were suitable (5, 6). Even by this method I haven't achieved satisfactory reactions yields. We have then turned our attention to zinc and magnesium complexes of Pc (ZnPc, MgPc). These synthesis methods are already known. Their reaction conditions are less...
Eukaryotic and prokaryotic nitric oxide synthase - structure-function studies

Nitric oxide (NO) is an important signaling molecule in organisms. It plays a role in wide spectrum of physiological and pathophysiological processes, including vasodilatation, neurotransmission and host defense. The gaseous molecule of NO is produced by oxidative reaction catalyzed by proteins from the family of nitric oxide synthases (NOSs). Three NOS isoforms were identified in mammals, endothelial (eNOS), neuronal (nNOS) and inducible or immunologic (iNOS). Some bacteria harbor genes coding for proteins homologous to the mammalian NOS oxygenase domain and showing NO-producing activity in vitro. NO generated by pathologic organisms such as B. anthracis and S. aureus is supposed to play a critical role in the pathophysiological processes during the infection. Comparative study of bacterial NOS-like proteins and mammalian NOSs confirmed their principal similarity, but also revealed differences in the interactions of distinct bacterial proteins and mammalian NOS isoforms with different analogs of substrate L-arginine and various ligands. On the basis of the kinetics measurement of NO-rebinding a second NO-binding site in the active center of NOS was predicted. Further, the regulation of NO dynamic and release from the protein by the active site Hbonding network connecting the heme, the substrate and BH4...

Induction therapy in lung transplantation: initial single-center experience comparing daclizumab and anti-thymocyte globulin BACKGROUND: Acute and chronic rejection remain unresolved problems after lung transplantation, despite heavy multidrug immunosuppression. In turn, the strong immunosuppression has been responsible for mortality and pervasive morbidity. Because acute rejection is associated with inferior outcomes in lung transplantation, we have routinely employed anti-thymocyte globulin (ATG), or daclizumab as adjuncts to reduce rejection. Daclizumab is a human monoclonal antibody that binds to the interleukin-2 receptor. METHOD: We performed a controlled clinical trial of these 2 therapies to determine differences in post-operative rejection, infection, bronchiolitis obliterans syndrome (BOS) and survival. 25 consecutive lung transplant patients received ATG (n = 12)(Group 1) and daclizumab (n = 13)(Group2) as induction agents. The groups had similar demographics and immunosuppression protocols differing only in induction agents used. RESULTS: No differences were observed in immediate post-operative outcomes such as length of hospitalization, ICU stay, or time on ventilators. No patient receiving daclizumab developed drug specific side-effects. There were no significant differences in the...
Upgrading of Trutnov hl.n. Railway Station
Král, Vladimír ; Wrana, Radim (referee) ; Svoboda, Richard (advisor)
The aim of the diploma thesis is a design of the reconstruction railway station Trutnov main station for compliance with current operations and especially with regard to persons with reduced mobility. So it was necessary to design the platform edge with a height of 550 mm above rail level. The design also deals with the drainage of reconstructed tracks and sidings connection.
