Study on the relation between G-quadruplexes and p53-driven regulation

This thesis deals with the role of G-quadruplex structures, their stabilisation using ligands and the role of p53 in transcription regulation. G-quadruplex is a type of nucleic acid secondary structure composed of 2 – 4 tetrads. Each tetrad is formed by four guanines linked by Hoogsten base pairing. This work consisted of theoretical and experimental parts. The theoretical part described the nature of G-quadruplex structures and their potential for cancer treatment, the role of p53 in the cell cycle and its regulation, and the ligands curcumin and TMPyP4 used for G4 stabilization. In the experimental part, the interactions of the curcumin and TMPyP4 with nucleic acid structures were studied. Reporter yeast strains containing PUMA, KSHV, and their combinations were transformed with plasmids encoding only selection markers and plasmids encoding the wild type form of the gene for protein p53. In the viability assay, the optimal ligand (curcumin and TMPyP4) concentration was determined for reporter yeast strain PUMA, which was further used in Luciferase assay. A fluorescence indicator displacement assay with Thioflavin T as a fluorescence dye proved the interaction between G4-forming oligonucleotides, and curcumin and TMPyP4. Luciferase assay was used to evaluate the interaction between transformed reporter yeast strains and both ligands. The results after 24 hours showed statistical significance for each strain in the same environment, where the PUMA strain showed the highest transactivation activity: the PUMA insert is a target gene for p53. Sequences with both PUMA and KSHV showed significantly lower transactivation activity, as ligands may have stabilized the G4-structure present in KSHV and thus down- regulate transcription. The presence of G4 in the KSHV sequence was also confirmed using the G4Hunter, with a G4Score of 3.182. Transactivation was significantly lower in the PUMA strains present in curcumin and TMPyP4 environment, although this sequence was not predicted to form G4, which was also confirmed in the G4Hunter analysis. This suggests that studied ligands curcumin and TMPyP4 may regulate the cell metabolism in a different than previously predicted way.
Suggestions for the Optimisation of the Remuneration Policies in Particular Company

This bachelor thesis is concerned with a search for optimalisation of the current employee remuneration system in chosen company. The first part of the thesis focuses on describing a theoretical introduction of the topic remuneration system and evaluation of employees, and is also used as a basis for consequent parts of the thesis. Second part concentrates on the analysis of the current situation in the company. Based on the analysis the last part deals with suggestions for optimal changes, which are meant to positively affect the future of remuneration in the chosen company.
