Language management in historic buildings with guided tours
Hanušová, Kateřina ; Sherman, Tamah (advisor) ; Nekvapil, Jiří (referee)
The presented thesis studies the language management of six historical objects currently maintained by the National Heritage Institute (NHI). With each, several different aspects were studied. Firstly, the aspect of organized language management, particularly the use of language when communicating with visitors, presenting the object publicly and organizing tours. The linguistic landscape analysis focused primarily on the visitor information system of each of the heritage objects. This provided crucial data for the study of the spatial use and distribution of languages. Furthermore, the matter of language ideologies and their influence on the language hierarchies was addressed. The last, aspect to be scrutinized, was the organizational structure of the NHI, particularly the levels responsible for language-related decisions. The thesis aims to describe the workings of such heritage sites from the aforementioned viewpoints, thus illustrating the language management process in the tourist industry.
Temporal characteristics in the speech of guides in repeated texts
Hanušová, Kateřina ; Palková, Zdena (advisor) ; Veroňková, Jitka (referee)
The guide's speech is a very specific type of discourse. It is a semi-planned monologue performed, with minimal deviations from the pattern, several times in succcession. These qualities therefore allow for a study of each speaker's articulatory rate. This paper analyzes the articulatory and speech rate, pauses, as well as the distinction of prosodic units, using four guides' speeches (utterances?). Each of the speeches was studied in two iterations. This paper presents a detailed description of the observed qualities for each speaker and a comprehensive summary comparison of the guides' speech patterns.
Opera Perdita of Josef Nešvera in context of its source material
Hanušová, Kateřina ; Gabrielová, Jarmila (advisor) ; Myslivcová, Eva (referee)
Perdita is an opera written by Jaroslav kvapil and composed by Josef Nešvera. Its' libretto is based on one of William Shakespeare's late romances The Winter's Tale. First part of this thesis focuses on characterization of the genre of romance as well as on The Winter's Tale and its place among other Shakespeare's plays. It also describes life and work of both Josef Nešvera and Jaroslav Kvapil, and Perdita's place among their work, which is not primarily operatic for neither of them. Perdita is described on basis of operatic style, derived from plays written and composed by these authors. In the second part of this thesis Perdita is compared to The Winter's Tale, in structure, work with themes and characters. At the end the thesis focuses on the opera's first performance and it`s reflections in press and contemporary sources.
Risk assessment of interspecific hybridization between endemic Campanula bohemica and widespread C. rotundifolia s.l.
Hanušová, Kateřina ; Suda, Jan (advisor) ; Krahulec, František (referee)
The thesis deals with phenotypic, ploidy and genetic variation of two Campanula species occurring in the Krkonoše Mts., namely the endemic C. bohemica and widespread C. rotundifolia subsp. rotundifolia. For comparative purposes subspecies sudetica of the latter species was also included. The main aim was to get insight into population structure and assess the threat of interspecific hybridization to the survival of the endemic bluebell. Flow cytometry, distance-based morphometrics and molecular analysis were used to address these questions. Three distinct groups of fluorescence intensities were revealed by flow cytometry, corresponding to DNA diploids, tetraploids and pentaploids. While diploids morphologically matched the nominate subspecies of C. rotundifolia, tetraploids corresponded either to C. rotundifolia subsp. sudetica or C. bohemica. Most populations from the Krkonoše Mts. were uniform although a sympatric growth of diploids and tetraploids was encountered in 12 populations. Only two pentaploids individuals, most likely of hybrid origin, were found, which indicated that interspecific hybridization is much less common than previously assumed. The two tetraploid taxa were distinguished by molecular markers. A combination of flow cytometry and molecular analyses thus allowed reliable...
Microevolutionary processes in the Czech endemic Campanula bohemica
Hanušová, Kateřina ; Suda, Jan (advisor) ; Vít, Petr (referee)
The genus Campanula L. - bellflower - is the largest group of the family Campanulaceae with a subcosmopolitan distribution and poorly resolved infrageneric classification. The evolutionary history of the genus has been shaped by a number of microevolutionary processes, including interspecific hybridization, genome duplication and geographical isolation, that resulted in the genesis of several endemic or geographically restricted species. The centre of endemism in the Czech Republic lies in subalpine altitudes of the Jeseníky and the Krkonoše (Giant) mountains, where three endemic taxa can be found: C. gelida Kovanda, C. rotundifolia L. subsp. sudetica (Hruby) Soó and C. bohemica Hruby. Despite their evolutionary and biogeographical value, there is a lack of information about their phenotypic variation, population structure, evolutionary history and processes acting in their populations. A critical assessment of these topics would require application of modern biosystematics tools. Campanula bohemica is an endangered neoendemics of higher altitudes in the Krkonoše Mts., closely related to C. scheuchzeri Vill., native to the Alps. The endemic species often grows in sympatry with related and morphologically similar C. rotundifolia. The incidence of intermediate morphotypes suggest that both species can...
Spirulina platensis effects on atherogenesis in mouse model of atherosclerosis.
Hanusová, Kateřina ; Mičuda, Stanislav (referee) ; Nachtigal, Petr (advisor)
Spirulina platensis is a single cell blue-green algae of Cyanobacteria strain. Spirulina belongs among foodstuffs with the highest protein content and contains all essential amino-acids. It is also a source of some non essential amino-acids, important nutrients as for example gamma linol acid, a lot of vitamins (B1, B2, B6, biotin, etc.) and trace elements (e.g. selenium, chrome, iron, calcium). Moreover it contains natural pigments carotenoids, chlorophyll and phycocyanin. The aim of this thesis was to test potential hypolipidemic and anti- inflammatory effects of Spirulina platensis on an experimental animal model of apoE/LDLr deficient mice. Therefore the parameters of lipid spectrum in blood and VCAM-1 expressions in arterial endothelium were monitored. Mice with deficit of apolipoprotein E (apoE-/- ) and LDL receptor were weaned of and for two weeks fed with a standard diet. At the age of eight weeks they started to be fed with atherogenic diet containing 0.15% of cholesterol. This continued for eight weeks (control group). In Spirulina platensis group the mice were fed with the same atherogenic diet with daily addition of 20 mg of Spirulina platensis. Biochemical analysis of lipid spectrum as well as histochemical and imunohistochemical analyses of atherosclerotic plates and VCAM-1...
T. G. Masaryk and The Czechoslovak Church
HANUŠOVÁ, Kateřina
This Master´s work deals with the relation of T. G. Masaryk and The Czechoslovak Church. The goal is to give reasons why Masaryk has never entered the Church despite the Church highly recognized his ideas and was based on many of them. The output of this work is supposed to be mostly the analysis of Masaryk´s texts besides the biographical texts and the written texts dedicated to the Czechoslovak Church. This work can be divided into three main parts. The first part follows Masaryk´s religious development. The second part comprises his opinions on particular religions issues. The third part introduces the basic information about the Czechoslovak Church and the last, fourth handles especially Masaryk´s attitude to the Church.
