Management plan of small water supply reservoirs of Landštejn type

The aim of the methodology is to provide managers and users of small water reservoirs Landštejn type with methodological tools for optimization of the water quality monitoring based on the current level of knowledge and research. Furthermore, provides a design of a methodology for fish community monitoring and optimization of treatments to reduce unwanted fish species and strengthen the status of suitable fish species with the aim of achieving the topdown effect and maintaining or improving water quality in these water reservoirs. The presented methodology summarizes the findings obtained during the monitoring and evaluation of selected parameters of water quality, planktonic and fish communities in water supply reservoirs Landštejn in recent years. It also contains an evaluation of applied treatments, focused mainly on fish stock. Unwanted fish species were caught and the predatory fish were stocked with the aim of optimizing the fish community composition and positively influencing the quality of the water in the reservoirs. The methodology contains recommendations for monitoring of the water quality in the reservoir, including monitoring of planktonic communities, monitoring of fish and recommended manipulation with fish stock. The methodology provides guidance and information for optimization of monitoring and treatments for small water supply reservoirs of the Landštejn type.

Morfologie chovaných skupin lososovitých ryb

Cílem studie bylo sledování vybraných morfologických znaků sledovaných druhů či skupin lososovitých ryb : pstruh duhový (Oncorhynchus mykiss) původem z Dánska, Itálie, Jihoafrické republiky, siven americký (Salvelinus fontinalis), siven arktický (S. alpinus), siven obrovský (S. namaycush), a jejich hybridi (S. fontinalis x S. alpinus). Sledovány byly vybrané morfologické parametry - stavba těla, hmotnost vnitřních orgánů, struktura pokožky a žáber.
DNA servis a konkrétní genetické produkty pro chovatele nejen lososovitých ryb

Mnohaleté zkušenosti s poskytováním genetického servisu a s řešením projektů spojených s chovem akvakulturních ryb a ochranářským managementem ryb ve volných vodách vedl vědecko-výzkumný tým k přípravě genetického toolboxu pro chovatele, rybáře a ochranáře. Tři připravené sady - "Standard", "Salmonid" a "Rezistence" poskytují nástroje a postupy řešící tematické oblasti: identifikační genetiky, reprofish genetiky, geneticky kontrolovaného chovného managementu a imunogenetiky včetně NGS technik. Nabízené postupy a řešení vycházejí ze vzájemné spolupráce univerzit, výzkumných institucí, chovatelských firem, rybářských sdružení a ochranářských organizací. Konkrétněji především z národního a evropského patentu a certifikovaných metodik připravených v Ústavu biologie obratlovců (ÚBO), Ústavu zoologie, rybářství, hydrobiologie a včelařství AF MENDELU a Ústavu ekologie a chorob zvěře, ryb a včel FVHE VFU.
Možnosti značení vysazovaných ryb pomocí ARS - aplikace a detekce

Cílem studie bylo vyzkoušet využití Alizarinové červeně při značení vybraných druhů ryb, současně ho prakticky ověřit při vysazovaní a dále ověřit jednoduchý způsob detekce značení při následném odlovu. V průběhu testování byl ověřena toxicita použitého barviva, délka barvení a minimální velikost vhodná pro aplikaci barviva. Součástí vyhodnocení bylo stanovení detekovatelnosti značení v průběhu růstu ryb.

Preparation and storage of fish samples for flow cytometry

Flow cytometry enabling rapid measurement of cellular DNA content has been widely used in ichthyology and fishery practice. The aim of the methodology is to contribute to optimization of work on the flow cytometer by helping with the selection, sampling and possibilities of fixation of analyzed tissue and interpretation of obtained data. The target group are people from fishery companies and ichthyologists dealing with the problem of the fish population structure in free water. During the sampling and analyzing of samples problem can be limited availability of fixative and techniques as well as analyzes of material not originally retained for the purpose of ploidy determination. Typical examples are temporary usable or archive preparations, museum material, etc. For the verification were preferred representatives of polyploid complexes or economically exploited species with a known existence of polyploid individuals.
