Simple flow-focusing microfluidic chip for droplet generation
Křivánková, Jana ; Basova, E. Y. ; Foret, František
There is an ascending requirement for analysis of chemical content of individual cells involving combination of various bioanalytical and optical sensors and arrangements. At the beginning of the cell analysis there is a need for creation of suitable transport vessels - droplets - that can be easily counted, sorted and serve for manipulation of cells with preserved biocompability as well. Among all of bioanalytical approaches droplet-based microfluidics proved its potential for producing droplets with precisely controlled size, shape as well as for encapsulation of reactants and delivering drugs. This study presents a simple microfluidic chip with flow-focusing geometry for generation of water droplets in fluorocarbon oil.

Development of sensitive, miniaturized and fast methods aims to reliable analyze even small amounts of samples. Micromanipulation systems such as laser capture microdissection enable to collect just single cells from cell suspensions or tissue samples. One option for further analysis is the bioluminescence reaction based on luciferin/luciferase conversion. This approach was used for detection of active caspases, caspase-3/7. Droplets based microfluidic systems provide benefits in biological, pharmaceutical and chemical research.
