The role of Erv1 in the mitochondrial import machinery and iron sulphur cluster export machinery in Trypanosoma brucei

Sulfhydryl oxidase Erv1 is a ubiquitous conserved protein of the mitochondrial intermembrane space, which is implicated in mitochondrial protein import as well as in the biogenesis of iron sulphur cluster proteins. While the role of Erv1 is quite well established in higher eukaryotes, Erv1 orthologues of parasitic protists exhibit some structural peculiarities. The study presented in this thesis therefore attempted to elucidate the function of Erv1 in Trypanosoma brucei.
The role of Q tRNA modification in the virulence of \kur{Leishmania mexicana}

The aim of the thesis was to indentify the orthologues of TGT1 and TGT2 genes in Leishmania mexicana and to verify the involvement of the respective gene products in the formation of Q-tRNAs in this species. Additionally, infection experiments in vitro as well as in vivo were carried out in order to test the plausible effect of Q-tRNA modification presence and/or absence on the virulence of the parasite.

The aim of this thesis was to determine the subcellular localization of Erv1 and to verify its involvement in the cytosolic iron sulphur cluster assembly. Additionally a search for potential interaction partners was conducted, which could possibly assist Erv1 in the processes where it was suggested to take a part, so either in the biogenesis of iron-sulphur cluster proteins or in the mitochondrial protein import, in procyclic T. brucei.
