Actuoecology of testate amoebae in fresh water and soil environment in enteraction with fungi and their analysis with new microscopic techniques

4 Abstract The present thesis focuses on testate amoebae (TA) and their relationship to their natural environment, as well as on relevant microscopic imaging methods. The bulk of the data has been published in original scientific papers and is compiled into three separate chapters (Pt I, Pt II and Pt III), each annotated by a brief introduction. (Pt I) The methods section is devoted to specialized microscopic techniques employed to broaden the scope of the ecological analyses. In particular, precise discrimination between live and dead individuals, biomass determination inside individual tests and a multi-modal visualization of the cytoplasm and organelles enhance the data. Laser scanning confocal microscopy and two-photon microscopy are the main imaging modalities employed to study TA morphology in detail. The data have implications for taxonomy and ecophysiology, including the use of TA as bioindicators of pollution. (Pt II) An actuoecological analysis focuses on the seasonal variability of TA species composition in a freshwater ecosystem, namely the Komo any ponds in Prague, during the course of the year. The theoretical part of this thesis is divided into three main chapters that deal with the issue storm deposits and their presence in the sedimentary record of borehole HR-1. One of the chapters dedicated to foraminifera tests as taphonomic indicators, which are stratigraphically and paleoecologically significant. Within the frame of this thesis it was thought to foraminifera more sedimentological than palaeontological or rather the tests of foraminifera were considered as specific particles, which on the seabed or in the water column were moving such as a grain of sand. The principal parameters for were selected size of test, mechanical wearing and life strategy (benthic forms inhabiting at the bottom and planktonic forms inhabiting within the whole or only in some parts of the water column). The second theoretical chapter is dealing with the tempestites, especially the condition of origin, identification and interchangeability for deposits of turbidite currents, Hummocky cross-stratification (HCS) and relation between tempestites and climate changes or atmospheric CO2. / SUMMARY The doctoral thesis discusses thecamoebians assemblages coming from five glacial lakes in the National Park Šumavaand from the Lipno Reservoir in the Southern Bohemia (the Czech Rep.). The research analyzes parameters of various assemblages commensurate with environmental characteristics. The aim of this research is to find relations useful in paleoecology. Thecamoebians are freshwater microorganisms with amoeboid cell and simple organic or anorganic test. Species with anorganic tests have been examined because of their better possibilities to fossilize. This taxon may prove to be a key taxon for monitoring environmental change during 21't century So it is expected that it could bear useful information about freshwater benthic paleoenvironment as well. Forty-six surface samples from the bottom of five glacial lakes in the ŠumavaMts., Czech Republic. collected in2002 were investigated and twelve species of thecamoebians with anorganic wall were identified in total. 96 oÁ samples contain the species Difflugia globulus. Other common species are Difflugia oblonga and Centropyxis orbicularis. Dominance oť DiÍÍlugiaglobulus and representation of Centropyxis orbiculari.r and Trigonopyxis arcula are the considerable differences comparing testate fauna from other world lakes.
