Transport studies in vitro on 2D and 3D cellular level

Trojrozměrné (3D) buněčné modely lépe napodobují buněčné podmínky in vivo než tradičně používané dvourozměrné (2D) buněčné modely. Transportéry pro organické anionty (OATs) a kationty (OCTs) hrají důležitou úlohu v ledvinné eliminaci léčiv a mají vliv na jejich farmakokinetické vlastnosti. Cílem práce bylo vytvořit 2D a 3D buněčný model z embryonálních ledvinných buněk HEK293, oba modely přechodně transfekovat transportérem hOAT1, transfekci optimalizovat a provést transportní inhibiční studie s vybranými NSAIDs (diklofenakem, ibuprofenem, indometacinem a naproxenem). 2D buněčný model jsme vytvořili ve 24jamkové destičce ve formě monovrstvy, jako 3D buněčný model jsme používali sféroidy, které jsme zhotovili na Petriho misce metodou visící kapky. Při transportních studiích jsme buňky inkubovali s radioaktivním substrátem OATs p-aminohippurovou kyselinou ([3 H]PAH) s nebo bez příslušného inhibitoru. Inhibiční vliv NSAIDs jsme porovnávali pomocí parametru IC50. 2D i 3D model z buněk HEK293 se nám povedlo vytvořit, ale následná transfekce byla úspěšná pouze v monovrstvě buněk.  Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Chemistry and Drug Analysis Sample preparation prior to an essay plays a key role in the drug analysis. Example of a widely used sample preparation technique is a Solid Phase Extraction. This method is characterised by high selectivity, which is given by the type of used sorbent. The aim of this thesis is to study the application of a new sorbent in Solid phase extraction which is zirconium oxide modified by graphene. Graphene is an allotropic modification of carbon possessing a specific adsorptive properties. Therefore it can be used besides other areas in chemical analysis. This thesis focuses on the testing on adsorptive capability of this sorbent using a mixture of model basic drugs. The attention was also paid to pH of solution used for conditioning of the sorbent and type of eluents. After the extraction, the samples were analysed by high- performance liquid chromatography, using a gradient elution. The most appropriate extraction environment was aqueous environment. The thesis also completes the knowledge already obtained from the diploma thesis of Lucie Bouzková (2016), which deals with finding suitable conditions for the separation of analytes. The work examines the influence of the concentration of the phosphoric acid, ammonium fluoride, sodium fluoride, triethylamine and phosphate buffers in the mobile phases on the elution of the analytes. The thesis further looks into the influence of the composition of the mobile phases with different ratios of the organic and aqueous components. The effect of increasing temperature on the retention of acyclovir and guanine was also observed. A higher content of organic component in the mobile phase leads to shortening of retention of acyclovir and guanine. Time-resolved spectroscopy of wide-bandgap semiconductors

In this thesis experimental samples of multiple quantum wells in the InGaN/GaN structures will be compared using methods of laser spectroscopy. In particular, the optical properties of the samples will be investigated. The samples were prepared under different conditions; therefore one of the aims is to compare them. The knowledge of the influence of preparation enables utilization not only for fundamental research, but also for the construction of radiation sources or scintillation detectors. Measurements of absorption and photoluminescence will be carried out and their dynamic properties will be measured as well. There will be examined the effect of different excitation power and different excitation wavelength on the intensity of photoluminescence. From dynamic properties there will be examined the effect of different excitation wavelength on the lifetime of the absorption and how does temperature influence the lifetime of the photoluminescence. Individual quantities will be compared amongst samples and their suitability for further applications will be discussed.
GALERIE VE VĚŽI A JEJÍ VÝZNAM V DĚJINÁCH KAŽDODENNÍ KULTURY NA MĚLNÍKU VE DRUHÉ POLOVINĚ 20. STOLETÍ

This rigorous thesis is a micro-historical probe into the history of everyday culture of the second half of the 20th century. It deals with the influence of the Communist regime on the cultural and social life in a central Bohemian town, at the background of the everyday life in a local fine art gallery. The Ve Věži gallery in Mělník was established at the end of the 60s of the 20th century at the premises of the medieval Prague Gate and fell under the administration of the city?s main cultural institution ? Cultural House of Captain Otakar Jaroš who set the official cultural direction of the then socialistic society in the Mělník region. The gallery was established by the academic painter Vladimír Veselý, a teacher at the local People?s School of Art Mělník, who built up this unusual fine art gallery that functioned throughout the period of the so-called normalization in Czechoslovakia, a period full of reluctance to accept people?s desire for free artistic expression. Despite this, the exhibition hall always offered a piece of freedom and liberty. The Ve Věži gallery showed works of young progressive artists and professionals achieving nation-wide and, often even, worldwide recognition and prevented by the then Communist regime from participating in the cultural life of the Czechoslovak society. However, they were always welcomed in the Mělník gallery. Nevertheless, the intention of this thesis is to show the importance and contribution of the gallery to social and cultural life in the Mělník region, where it became an island of freedom in an unfree period of totalitarian Czechoslovakia, rather than assessing the gallery in terms of history of fine arts.
The Diverged Trypanosome MICOS Complex as a Hub for Mitochondrial Cristae Shaping and Protein Import

This work deals with MICOS, which stands for mitochondrial contact site and cristae organization system. Until now this multiprotein complex has been analyzed experimentally only in yeast and mammals, who belong to the supergroup Opisthokonta. Our study was done on the parasitic protist T. brucei, a member of the another supergroup called Excavata, which is very diverged from opisthokonts.Thus, it is the very first study done outside of Opisthokonta. This could be very useful in the future for a comparative analysis approach. Our results show that the MICOS complex in T. brucei is composed of 9 subunits, most of which are essential for normal growth. It is required for the maintenance of discoidal cristae that typify excavates such as kinetoplastids and euglenids and mediating the mitochondrial outer and inner membranes contacts. In addition, we discovered that the mitochondrial contact site and cristae organization system may participate in the intermembrane space protein import and help in the oxydative phosphorylation complex formation. It seems that this interesting complex is involved in even more cellular processes.

The main aim of the thesis is to map out problematic pupil from the teacher's perspectvie, specifically the perspective of school prevention methodologhist. In the theoretical part, it mentions problematic behavior in general. The following chapters deal specifically with problematic behavior at school. There are mentioned its causes, the differences between the pupil with behavioral disorders and the pupil with problematic behavior as well as the characteristics of the problem child. The third chapter deals with the troubled pupil's family and the communication between school and parents. In the next part of the text there are mentioned some researches, which focused on the perception of the problematic pupils from the teachers point of view, their relationship and their approach, including the personality of pedagogue and burnout syndrome sub-chapters. Personality of the music theorist and pedagogue Ondřej Bednarčík (1929-1998)

The thesis presents the life story and professional growth of Ondřej Bednarčík, a musician theorist and pedagogue from Ostrava, who lived between 1929-1998. The profile features of this big name of music crystallized pursuant to the analysis of Bednarčík's pedagogical, scientific, research, educational and musical activities. As far as Bednarčík's main scientific interests are concerned, attention is paid to contemporary Polish music, listening to New Music, musical and didactic issues of 20th century music, whilst focusing on the teaching of musical and theoretical subjects and, finally, to two distinct personalities - composer Bogusław Schaeffer and his colleague Professor Josef Schreiber, who strongly aided in the shaping of Ondřej Bednarčík in his scientific research and human approach. The thesis is divided into the following eight chapters. The first one is dedicated to the current state of the research on the personality of Ondřej Bednarčík, with main and partial goals, methodological principles, used research methods and processing of Bednarčík's written inheritance. The second section introduces us to Ondřej Bednarčík's life story, musical interests and activities. Museum educational programmes for National Pedagogical Museum of J. A. Comenius in Prague

The main aim of the Diploma Theses is to prepare, verify in practice and evaluate museum educational programme for pupils from 12 to 18 (junior schools and secondary schools) which is devoted to the changes in schooling in 20th century. The aim is to introduce to pupils historical facts by the means of three educational programmes and interactive elements in the exhibition. Pupils work with topics covering World War II, 50s and so called normalization period. Throughout the educational process they will use period realia from the above mentioned periods, such as schoolbooks, educational boards and educational aids from the collection of Comenius National Pedagogical Museum and Library in Prague. Analysis of work performance management of teaching staff in the schools established under part 16 paragraph 9 of the Czech Education Act

The aim of the thesis is the analysis of the performance management of teachers in a school established in accordance with § 16 para. 9 of the Education Act. The theoretical part of the thesis specifies the concept of human resource management and defines the individual personnel activities as a tool for human resource management. It highlights from personal activities the function of performance management and describes it using the literature. It specifies in detail the phases of the cycle of the performance management process. The theoretical part contains the description of the process of performance management of teaching staff, defines the identification of the specific conditions in a school established by § 16 para. 9 of the Education Act, which may have an impact on the monitored personal activities. The analytical part of the thesis provides the results of research in primary schools established by § 16 para. 9 of the Education Act. The survey was conducted using explorative method in questionnaire and the type of the case study. The data collection in the case study was realized using individual semi-standardized interviews and the analysis of school documents and guidelines.
