Sprouty proteiny se řadí mezi negativní regulátory RTK (receptor tyrozin kináz), včetně FGF signální dráhy. Signály FGF signálních drah se podílejí na vývoji patra či zubních základů, mimo jiné řídí proliferaci a diferenciaci osteoblastů a podílí se na formování končetin. Především jeden ze čtyř FGF receptorů, FGFR3, významně ovlivňuje například chondrocyty v růstové ploténce a jeho mutace se projevuje chondrodysplastickými syndromy. Sprouty2 (Spry2) gen se v embryonálním vývoji účastní vývoje končetin, ledvin, větvení plic a také zubní morfogeneze. Spry2 deficientní jedinci vykazují nadpočetné zuby v diastemě mandibuly, abnormality ve tvaru lebky či rozštěpy patra. V kraniofaciálním vývoji a také u končetin hraje významnou úlohu i gen Sonic hedgehog (Shh). Cílem předložené diplomové práce bylo popsat expresi Spry2 v kraniofaciální oblasti a u končetinových základů s důrazem na raná stádia vývoje. Dále jsme se s využitím Spry2 deficientních myší zaměřili na vzájemný vztah mezi geny Spry2 a Sonic hedgehog (Shh) v kraniofaciální oblasti a v končetinovém pupenu. Exprese Spry2 a Shh genů byla detekována pomocí in situ hybridizace. Aktivita těchto genů byla hodnocena na základě vizualizace exprese příslušných proteinů s využitím immunohistochemie. Sprouty proteins are known as negative regulators of the pathways downstream of receptor tyrosine kinases (RTKs), including fibroblast growth factor (FGF) pathways. FGFs are involved in patalal and tooth formation, osteoblasts proliferation, differentiation and bone formation. Especially FGFR3, one of the fibroblast receptors, affects chondrocytes in the growth plate and its gain-of-function mutation is responsible for chondrodysplastic syndromes. Sprouty2 (Spry) gene plays an important role in embryonic development, as in the development of limb, kidney, pulmonary branching and morfogenesis of teeth. Spry2 knock-out results in supernumerary tooth formation in diastema of mandible, abnormalities of skull shape or cleft palate. Sonic hedgehod gene (Shh) also plays an important role in the craniofacial development and in the development of limb bud. The aim of this master thesis was to describe the expression of Spry2 in the craniofacial area and in the developing limbs with focus on the early stages of embryogenesis. We focused also on the relationships between Spry2 and Sonic hedgehog (Shh) gene expressions in the craniofacial area and in the limb buds.
