Cytochromy P450 hrají klíčovou roli při metabolismu endogenních a exogenních sloučenin. Účastní se jak biotransformace, tak biosyntézy steroidních hormonů. Významným zástupcem těchto hemových enzymů je cytochrom P450 19, aromatasa, která katalyzuje finální krok biosyntézy estrogenních hormonů, aromatizaci androgenů na estrogeny. K fyziologickým funkcím estrogenů se řadí vývoj sekundárních pohlavních znaků, podpora tvorby kostní hmoty nebo regulace sekrece gonadotropinu. Narušení hormonální rovnováhy vlivem endokrinních disruptorů však může vést mimo jiné k rozvoji nádorových onemocnění nebo reprodukčním problémům. Mezi sloučeniny s těmito účinky patří kromě kontaminant životního prostředí také některá léčiva a kosmetická aditiva, kterým jsou lidé vystaveni každodenně. V této bakalářské práci byl zkoumán vliv vybraných parfémů a antiperspirantů na metabolickou konverzi testosteronu na estradiol, katalyzovanou enzymem aromatasa. Aktivita tohoto enzymu byla orientačně detekována s využitím TLC chromatografie. Experimentálně bylo zjištěno, že vybrané antiperspiranty nemají na aktivitu aromatasy vliv. U většiny testovaných parfémů byla zaznamenána jen velmi nízká míra inhibice aromatasy. Pouze jeden vzorek tento efekt vykazoval ve vyšší míře. These enzymes are involved both in biotransformation and steroid hormone biosynthesis. An important member of this family is cytochrome P450 19, aromatase, which catalyzes the final step of estrogen hormone biosynthesis, conversion of androgens into estrogens. The physiologic functions of estrogens include development of secondary sexual characteristicts, maintenance of bone mass or regulation of gonadotropin secretion. However, hormonal imbalance due to endocrine disruptors can result in development of certain cancers or impaired reproduction. Compounds with these effects include, in addition to environmetal pollutants, some drugs and cosmetic additives which people are exposed to on a daily basis. In the present study, the effect of selected perfumes and antiperspirants on the metabolic conversion of testosterone into estradiol, catalyzed by aromatase, was examined. The activity of this enzyme was detected using TLC chromatography. The experiments showed that selected antiperspirants do not affect aromatase activity.
