Epigenetika je vědní obor zabývající se studiem dědičných změn fenotypu, které nejsou způsobeny změnou v sekvenci DNA. Mezi hlavní epigenetické mechanismy spadá kovalentní modifikace DNA, zejména methylace, modifikace histonů a s tím spojená modifikace chromatinu, a RNA interference. Tyto mechanismy se mohou uplatnit během řady procesů, od transkripce až po translaci. LEDGF/p75, z angl. Lens epithelium-derived growth factor, je všudypřítomný protein lidského těla považovaný za transkripční koaktivátor upregulovaný za stresových podmínek. Skládá se z několika domén, z nichž na svém N-konci obsahuje PWWP doménu, která je schopna vázat di- a trimethylovaný lysin 36 na histonu 3 - tedy epigenetickou značku transkripčně aktivního chromatinu. Mezi jeho interakční partnery patří například HIV integráza, MLL1-MENIN komplex nebo protein MeCP2. Kratší izoformou tohoto proteinu je LEDGF/p52 sdílící s LEDGF/p75 jeho N-koncovou část důležitou pro interakci s DNA a chromatinem. MeCP2, z angl. Methyl-CpG-binding protein 2, je rovněž všudypřítomným proteinem lidského těla, ovšem s nejvyšší koncentrací v mozku. Jedná se o transkripční modulátor remodelující chromatin, jehož funkcí je proto aktivace nebo represe genu v závislosti na molekulárním a buněčném kontextu. Major epigenetic mechanisms include covalent DNA modifications (particularly methylation), histone and chromatin modifications and RNA interference. These mechanisms are involved in number of processes from transcription to translation. Lens epithelium-derived growth factor (LEDGF/p75) is ubiquitously expressed in human body and it is considered to be a transcriptional coactivator upregulated upon stress conditions. LEDGF/p75 consists of several domains. The N-terminal PWWP domain plays very important role from epigenetic point of view as it is able to bind di- and trimethylated lysine 36 of histone 3, which is considered as an epigenetic marker of transcriptionally active chromatin. LEDGF/p75 interaction partners include e.g. HIV integrase, MLL1-MENIN complex or MeCP2. A shorter isoform of LEDGF/p75 called LEDGF/p52 shares with LEDGF/p75 its N- terminal part that is responsible for interaction with DNA and chromatin. Methyl-CpG-binding protein 2 (MeCP2) is present everywhere in human body with the highest abundance in brain.
