RNDr. Lenka Skálová, Ph.D. Název diplomové práce: Degradace kyseliny hyaluronové v kůži a perkutánní absorpce exogenní kyseliny hyaluronové kůží V kůži se nachází až polovina celkového tělesného obsahu kyseliny hyaluronové (HA). HA funguje jako organizátor extracelulární matrix a podílí se na udržení hydratace a elasticity kůže. V poslední dekádě přibylo v oblasti degradace HA v kůži několik významných poznatků a jedním z cílů této diplomové práce bylo utvořit jejich ucelený přehled. Degradace HA v dermis byla objasněna objevením esenciální role, kterou hraje protein KIAA1199 při degradaci HA v dermálních fibroblastech. V epidermis ve stratum granulosum těsně pod stratum corneum (SC) byla prokázaná nadměrná exprese lyzosomálního enzymu hyaluronidázy 1. Není jisté, jakým způsobem je HA do keratinocytů internalizována nebo jestli je tímto způsobem degradována pouze intracelulární HA. Epidermální degradace HA tak zůstává téměř neobjasněna. HA je součástí řady topicky aplikovaných produktů na kůži zejména kvůli svým unikátním hydratačním vlastnostem, avšak není zcela objasněno, jestli je HA vůbec schopna pronikat do hlubších vrstev kůže. HA acts as an organizer of the extracellular matrix and is involved in maintaining skin hydration and elasticity. In the last decade, several important findings have been added to the field of HA degradation in the skin, and one of the aims of this thesis was to create a comprehensive overview. Degradation of HA in the dermis was elucidated by the discovery of the essential role in the degradation of HA in dermal fibroblasts played by the KIAA1199 protein. In the epidermis, the lysosomal enzyme hyaluronidase 1 is overexpressed in the stratum granulosum just below the stratum corneum (SC). It is uncertain how HA is internalized into the keratinocytes or whether only intracellular HA is degraded in this way. Epidermal degradation of HA remains elusive.
