Tato bakalářská práce se zabývá studiem reaktivity bakteriálních extracelulárních polysacharidů. V praktické části pojednává o extracelulárním polysacharidu FucoPol. Kvůli přítomnosti acylových skupin má polymer aniontový charakter a může podporovat interakce s jinými kladně nabitými látkami. V rámci bakalářské práce byly studovány interakce FucoPolu s kovy. Pro měření byl využíván zejména izotermický titrační kalorimetr a byly provedeny sorpční experimenty. Hlavním cílem byla optimalizace podmínek procesů měření a posouzení interakcí mezi kovy a FucoPolem. Pomocí titrační kalorimetrie a sorpčních experimentů byla potvrzena vazba kationtu kovu na matrici FucoPolu. Míra interakce se lišila v závislosti na použitém kovu, zejména Co2+, Pb2+ a Zn2+ ionty se vázaly velice slabě, a naopak ionty Cu2+ a Fe3+ měly velkou afinitu ke studovanému biopolymeru. FucoPol jakožto biodegradabilní biopolymer by se mohl v budoucnu stát jedním z důležitých průmyslově vyráběných polysacharidů, který by se dále mohl využívat v obalových materiálech, zejména pro jeho interakci s kovy, dále v zemědělství jako biohnojivo anebo také v lékařství s potenciálem pro použití v protinádorových, protizánětlivých a imunitně posilujících léčivech. This bachelor thesis deals with the reactivity of bacterial extracellular polysaccharides. The experimental part deals with extracellular polysaccharide FucoPol. Due to the presence of acyl groups, the polymer has an anionic character and can promote interactions with other positively charged substances. In this bachelor thesis interactions of FucoPol with metals were studied. For measuring, an isothermal titration calorimeter was used, and sorption experiments were performed. The main objective of the work was optimization of conditions for measuring interactions between metals and FucoPol. Connection of metal cations to FucoPol matrix were confirmed by titration calorimetry and sorption experiments. The interaction rate was different depending on the metal which was used. Co2+, Pb2+ a Zn2+ ions showed very weak binding, otherwise Cu2+ a Fe3+ ions had high affinity for the studied biopolymer. FucoPol, as a biodegradable biopolymer, can be in the future one of important industrially produced polysaccharides. It could be used for packaging, biofuel production and production of anti-inflammatory and immune-boosting drugs.
