Hemoproteiny představují vzhledem k jejich širokému výskytu a rozmanitým funkcím nepostradatelnou součást mnoha živých organismů. Mezi typické funkce hemoproteinů patří participace na transportu a uskladnění kyslíku v živých organismech, transport elektronů v rámci dýchacího řetězce či enzymová katalýza reakcí, kterých se účastní kyslík, případně peroxid vodíku. Poměrně nedávno byla objevena nová skupina hemoproteinů, jejíž zástupci disponují schopností detekovat buď změny koncentrace hemu (senzorové proteiny, které detekují hem), nebo změny koncentrace dvouatomových molekul plynů (hemové senzorové proteiny, které detekují plyny) v jejich okolním prostředí. Signály generované v rámci této detekce způsobují strukturní změny příslušných senzorových proteinů a ovlivňují tak jejich funkce. Předmětem této diplomové práce je příprava a následná charakterizace eukaryotického senzorového proteinu Bach1, který detekuje hem. Důraz je kladen především na interakční schopnosti proteinu Bach1 s heminem a komparaci vlastností apoformy a holoformy Bach1. Pozornost je věnována hlavně počtu molekul heminu, které jsou schopny specificky interagovat s Bach1, a to za různých podmínek (odlišný redoxní stav iontu železa v hemu, přítomnost interakčních partnerů Bach1 apod.). Součástí této práce je také analýza vlivu... Hemoproteins represent very important components of many living organisms. Participation in the processes of oxygen transport and storage, electron transport or enzymatic catalysis of reactions involving oxygen or hydrogen peroxide are commonly known functions of hemoproteins. Recently, there has been discovered a new group of hemoproteins. The main feature of this new group of proteins is their ability to detect changes in heme concentration (heme-responsive proteins) or changes in diatomic gas concentration (gas-responsive heme-containing sensor proteins) in their vicinity. Detection of these concentration changes generates signals that induce structural changes of the respective sensor proteins. Finally, the structural changes of the respective sensor proteins affect their functions or activities. The subject of this diploma thesis is the preparation and characterization of the eukaryotic heme sensor Bach1. We especially focused on the ability of Bach1 to bind heme molecules and on the comparison of various Bach1 properties in its apoform and holoform. Determination of the exact amount of heme molecules that specifically interact with heme sensor Bach1 represents very important part of this thesis.
