Úvod: Epoxyeikosatrienové kyseliny (EETs) jsou přeměněny enzymem solubilní epoxid hydrolázy (sEH) na biologicky neaktivní dihydroxyeikosatrienové kyseliny (DHETs). EETs se významně podílí na kontrole krevního tlaku, ovlivňují vaskulární tonus a renální transportní mechanismy. Inhibitor sEH snižuje krevní tlak zvýšením biologické dostupností EETs u mnoha modelů hypertenze. Cíl: Zjistit, zda inhibitor sEH snižuje krevní tlak a zlepšuje renální funkce během rozvoje maligní hypertenze u transgenních potkanů po indukci myšího reninového genu. Metodika: Hypertenze u Cyp1a1-Ren-2 transgenních potkanů byla navozena podáním diety s obsahem přírodního xenobiotika indol-3-karbinolu (I3C, 0.3%) po dobu 3 a 11 dní. I3C aktivuje reninový gen. U 3-denní indukce hypertenze byl současně podáván inhibitor NO syntházy (L-NAME, 600 mg/l) v pitné vodě. Inhibitor sEH c-AUCB byl podáván v pitné vodě v dávce 13 nebo 26 mg/l 48 hod před podáním diety obsahující I3C a L-NAME. U bdělých zvířat bylo provedeno radiotelemetrické měření krevního tlaku a sledování exkrečních parametrů ledvin. U anestezovaných zvířat byly zjištěny účinky na hemodynamiku a exkreční funkce ledvin akutními renálními studiemi. Výsledky: Podávání I3C u Cyp1a1-Ren-2 transgenních potkanů vedlo k rozvoji maligní hypertenze se zvýšením systolického... Introduction: Epoxyeicosatrienoic acids (EETs) are converted by the enzyme soluble epoxid hydrolase (sEH) to the biologically inactive dihydroxyeicosatrienoic acids (DHETs). EETs are significantly involved in the control of blood pressure, they influence vascular tone and renal transport mechanism. sEH inhibitor reduce blood pressure by increasing the bioavailability of EETs in many models of hypertension. Aim of the study: To determine that sEH inhibitor decreases blood pressure and improves the renal function during the development of malignant hypertension in transgenic rats after the induction of the mouse renin gene. Methods: Hypertension in Cyp1a1-Ren-2 transgenic rats was induced through a dietary administration of the natural xenobiotic indole-3-carbinol (I3C, 0.3 %) for 3 and 11 days. I3C activates the renin gene. At the same time, during a three-day induction of hypertension, the inhibitor of nitric oxide synthase L-NAME (600 mg/l) was administered in drinking water. The sEH inhibitor c-AUCB was given in drinking water at a dose of 13 or 26 mg/l, starting 48 hours before the initiation of I3C and L-NAME administration.
