Dolichol je membránový lipid, který se podílí na glykosylaci proteinů a biosyntéze glykosylfosfatidylinositolové kotvy. Jeho struktura je složená z isoprenových jednotek a je přítomen ve všech tkáních a většině membránových organel eukaryotických buněk. Nedávno byly některé typy dědičných poruch glykosylace popsány jako důsledek poruchy biosyntézy a metabolismu dolicholu, které však nejsou zachytitelné běžnými metodami. Cílem bylo zavést analýzu obsahu dolicholu v moči a tkáních pomocí tandemové hmotnostní spektrometrie, analyzovat obsah dolicholu v souboru pacientů s vybranými typy dědičných poruch glykosylace a studovat dopad poruchy biosyntézy dolicholu na energetický metabolismus. Biologický materiál pro analýzu obsahu dolicholu tvořil kontrolní soubor močí od 76 zdravých kontrol ve věku od 1 měsíce do 81 let, 6 pacientů s dědičnými poruchami glykosylace, 43 pacientů s podezřením na toto onemocnění; a vzorky tkání: frontálního kortexu, jater, kosterního svalu a srdce od dvou pacientů s poruchou v genu NUS1 a kontrol. Pro funkční studie energetického metabolismu byly použity linie kultivovaných kožních fibroblastů. Lipidová složka byla po extrakci separována na kapalinovém chromatografu a dolicholy byly analyzovány na tandemovém hmotnostním spektrometru. Byly detekovány dolicholy se 17-20... Dolichol is a membrane lipid, which carries monnosaccharides and glycans for N-linked protein glycosylation and glycosylphosphatidylinositol-anchor biosynthesis occuring in endoplasmic reticulum. Its structure is composed of isoprenoid units. Dolichol is present in all tissues and in most of the membrane organelles of eukaryotic cells. Recently some types of congenital disorders of glycosylation have been described as a consequence of dolichol biosynthesis and metabolism defects, which are not detectable by standard methods. The aim of this diploma thesis was to analyze dolichol content in urine and in different tissues from patients with deficiency in dolichol biosynthesis by mass spectrometry and to study the impact of these defects on energetic metabolism. Biological material for this study consisted of urine samples from 76 controls with age ranging from 1 months to 81 years, 6 patients with congenital disorders of glycosylation and 43 patients with suspicion of congenital disorder of glycosylation; samples of frontal cortex, liver, muscle and heart tissues from 2 patients with mutation in NUS1 gene and controls.
