Tato bakalářská práce je zaměřena na stanovení chlorofylu v listech metodou extrakční spektrofotometrie. Teoretická část přináší informace o fotosyntéze, stresových faktorech či adaptaci rostlin, dále o fotosyntetických pigmentech (zejména chlorofylu-a) a aplikaci spektrofotometrie a dalších metod stanovení chlorofylu. Experimentální část se zaměřuje na nasazení ekotoxikologického testu a především jeho vyhodnocení dvěma nezávislými metodami. Konkrétně se jednalo o stanovení inhibice růstu biomasy a nově o spektrofotometrické stanovení množství chlorofylu. Výsledky mezi sebou navzájem korelují, a proto se stanovení chlorofylu jeví jako vhodné rozšíření obvyklých vyhodnocovacích metod ekotoxikologických testů. This bachelor thesis is focused on determination of chlorophyll in leaves by the method of extraction spectrophotometry. The theoretical part provides information about photosynthesis, stress factors or plant adaptation, photosynthetic pigments (especially chlorophyll-a) and application of spectrophotometry and other methods of chlorophyll determination. The experimental part is focused on the application of the ecotoxicological test and, above all, it is evaluation by two independent methods. Specifically, it was determination of biomass growth inhibition and newly spectrophotometrical determination of chlorophyll mass. The results correlate with each other, therefore, the determination of chlorophyll seems to be an appropriate extension of usual evaluation methods of ecological tests.
