6 ZÁVĚR 27 6 ZÁVĚR U nervových kmenových buněk byly pomocí dvojrozměrné gelové elektroforézy a následné hmotnostní spektrometrie identifikovány konstitutivně syntetizované proteiny v 66 proteinových skvrnách. Nejčastěji byly zastoupeny proteiny účastnící se metabolismu a úprav RNA a proteinů, následované cytoskeletárními proteiny a anexiny, metabolickými proteiny a enzymy, proteiny účastnící se tvorby energie, obrany buňky a buněčných signalizací. Bylo identifikováno 16 proteinů, jejichž hladina se statisticky významně měnila během diferenciace nervových kmenových buněk do nervových buněk. Zvýšení hladiny α- B crystallinu, hnRNP A1 a hnRNP A2/B1 a jejich lokalizace v nervových buňkách byly potvrzeny imunoblotem a imunocytochemií. Pomocí protilátkových mikročipů byly u nervových kmenových buněk detekovány fosforylace signálních proteinů související většinou s buněčnou proliferací (CDK1 a 2, PKC mu, PKCγ, Erk5 a α-B crystallinu) a zvýšená hladina proteinu GRK2. Naopak u diferencovaných buněk byl prokázán nárůst hladiny protein fosfatázy 4 (její katalytické podjednotky), hem-oxygenázy 2, kináz MEK3 a RafB a pro-kaspázy 1 a zvýšená fosforylace 40 kDa substrátu Akt kinázy bohatého na prolin. U nádorových buněk bylo po separaci proteinů systémem ProteomeLabTM PF 2D identifikováno 8 proteinů, jejichž hladina byla... CoNcLUsIoNs In protein profiling of neural stem cells using 2-dimensional gel electrophoresis and mass spectrometry, constitutively expressed proteins in 66 protein spots were identified. Most of the individual protein species were related to RNA and protein metabolism, processing and turnover, including some chaperones and stress response proteins. Proteins involved in cellular organization (e.g. cýoskeletal proteins and annexins), metabolic proteins (mostly enrymes),cellular energetics,cell defenseand signallingfollowed in lower numbers. Proteins in 16 spots significantly regulated during neural differentiation were identified. Induction of levels of o-B crystallin, hnRNP Al and hnRNP AZIBI during differentiation and protein localization within neural cells were studied by westernblottingand immunocýochemistry. Using antibody microarrays, in neural stem cells an increase in GRK2 level and phosphorylationsof signalling molecules(CDKI|Z, PKC mu, PKCy, Erk5 and o-B crystallin) involved mostly in cellular proliferation were detected.On the contrary, in differentiatedneural cells levels of protein-phosphatase4, heme-oxygenase2, MEK3, RafB, pro-caspase 1 and phosphorylation of 40 kDa proline-rich Akt substratewere induced.
