Tato práce měla za cíl identifikovat proteiny tenké spojovací vrstvy dentin-enamel (DEJ) přítomné v lidských zubech moudrosti. Byly vyzkoušeny tři různé metody analýzy. V jednom případě byly zuby demineralizovány v roztoku EDTA pufru, přičemž došlo k uvolnění proteinové vrstvy DEJ. V dalších dvou případech byly připraveny tenké řezy zubní tkáně a příslušná DEJ vrstva byla vyříznuta pomocí laserové mikrodisekce. Několika metodami byla zkoumána extrakce proteinů ze zkoumaných vzorků DEJ (pro srovnání i ze vzorků dentinu, enamelu). Extrahované proteiny byly štěpeny trypsinem na peptidy. Následně byly peptidy purifikovány pomocí Stage tips a analyzovány metodou nLC MS/ MS. Optimální metodou bylo ve vzorku DEJ nalezeno přes 40 různých proteinů např. apolipoproteiny, vimentiny, vitronektiny, clusteriny, biglykany. Klíčová slova: proteomika, dentin-enamelová vrstva, laserová mikrodisekce, hmotnostní spektrometrie This thesis deals with the analysis of the dentin-enamel junction layer in human third molar teeth. Three different methods of analysis were tested. In the first one teeth were demineralized in EDTA buffer to release the protein layer DEJ. In the other two cases thin slils of dental tissue were preparedand from them the DEJ layer was cut using laser microdissection. Severals methods were tested for extraction of proteins from DEJ samples (for comparsio also from dentin and enamel samples). Extracted proteins were cleavaged into peptides with trypsin. Subsequently, peptides were purified by Stage tips and analyzed by nLC MS/ MS. Using the optimal method over 40 different proteins, for example: apolipoproteins, vimentin, vitronectin, clusterin, biglycan weres found in the sample DEJ.
