To find out if the haematopoietic system is common feature of vertebrates, we decided to examine haematopoiesis in a sea lamprey (Petromyzon marinus). All blood cells arises from the haematopoietic stem cells in higher vertebrates. We assume that this is common for the higher vertebrates and a jawless vertebrates, but nobody was interested in the jawless haematopoiesis since 1970. Using a reverse genetic, we identify homologues of important hematopoietic of higher vertebrates in transcriptome of the sea lamprey with emphasis on important receptors or transcription factors, because they can be used as the specific markers of different blood cells and their progenitors. Then we use those sequences for cloning, expression measurements and other work. We picked up sea lamprey as model organism because its unique phylogenetic position, important foe evo-devo studies, but also because lack of elementary knowledge about sea lamprey haematopoiesis. Key words: Petromyzon marinus, haematopoiesis, HSC, evo-devo Abychom zjistili, zda je systém krvetvorby skutečně společným znakem obratlovců, rozhodli jsme se prozkoumat krvetvorbu u fylogenticky nejstarších žijících zástupců obratlovců, tedy kruhoústých, jmenovitě u mihule mořské (Petromyzon marinus). U vyšších obratlovců pochází všechny krevní buňky z HSC. Předpokládáme, že je tomu tak i u kruhoústých, nikdo se ale o krvetvorbu u těchto organismů od sedmdesátých let zvlášť nezajímal. Vydali jsme se po cestě reverzní genetiky a hledali homology genů důležitých pro krvetvorbu vyšších obratlovců v transkriptomu mihule mořské s důrazem především na významné receptory a transkripční faktory, jelikož se dají využít jako markery různých krevních buněk a jejich progenitorů. Pomocí bioinformatických metod jsme v datech z NGS nové generace nalezli homology klíčových genů krvetvorby, které jsme dále využili pro klonování těchto sekvencí, měření jejich exprese a další práci s nimi. V naší laboratoři jsme si tedy vybrali mihuli mořskou jako modelový organismus, nejen kvůli unikátnímu fylogenetickému postavení, obzvlášť pro evo-devo studie, ale také kvůli absenci základních poznatků z oblasti krvetvorby.
