Trichomonas vaginalis je lidský parazit, který ročně ohrožuje přibližně 258 milionů lidí. U tohoto parazita byla popsána organela mitochondriálního původu nazvaná hydrogenosom, která je schopná vytvářet energii v podobě ATP za anaerobních podmínek. Zkoumání a popisování nových proteinů, vyskytujících se v této organele, může přispět k objevení nových cílů pro antiparazitární léčbu. Zároveň nám tento výzkum pomůže porozumět fungování a evolučnímu původu této odvozené organely. Dostupnost již popsaných protokolů pro izolaci hydrogenosomů společně s kompletně zmapovaným genomem T. vaginalis umožňuje studovat tyto organely pomocí moderních metod hmotnostní spektrometrie a bioinformatických analýz. V rámci našeho výzkumu jsme použili různé přístupy umožňující popsat proteiny v těchto organelách. Jednalo se o transkriptomické a proteomické analýzy zjišťující buněčnou odpověď parazita T. vaginalis na změny koncentrace železa v prostředí. A dále změny v obsahu proteinů hydrogenosomů u T. vaginalis rezistentních na metronidazol. Organely byly v obou případech izolovány pomocí diferenciální centrifugace gradientu OptiPrepu. Takto připravené hydrogenosomy byly analyzovány hmotnostním spektrometrem pomocí nano-RP- HPLC/MALDI-TOF/TOF. Zároveň jsme pomocí Tritonu X-114 oddělili membránové a matrixové proteiny a... Trichomonas vaginalis is a human pathogen that affects annually approximately 258 million people worldwide. This parasite possesses organelles of mitochondrial origin called hydrogenosomes, which generate ATP under anaerobic conditions. The identification of the protein content at the subcellular level may provide new targets for antiparasitic drugs developments as well as it contributes for our understanding of the organelles function and evolution. The availability of protocols for organelles purification and the complete genome sequence allow the study of the organellar proteomes using mass spectrometry and bioinformatics, providing a powerful strategy that combine cell biology and proteomics. In our research, we used several approaches to identify the protein composition in hydrogenosomes and mitosomes. We performed transcriptomic and proteomic analysis to investigate the molecular responses of Trichomonas vaginalis upon iron availability. Furthermore, the changes in the proteome during the development of metronidazole resistance were also studied. The organelles separated by differential and Optiprep-sucrose gradient centrifugation were analyzed with nano- RP-HPLC/MALDI-TOF/TOF.
