Souhrn Glutamátové receptory NMDA a AMPA typu procházejí během časných stadií postnatálního vývoje intenzivními funkčními změnami danými modifikací jejich podjednotkového složení (Pachernegg et al., 2012: Paoletti et al., 2013). Velké zastoupení NMDA receptorů obsahujících NR2B podjednotku a AMPA receptorů neobsahujících GluA2 podjednotku (prostupných pro Ca 2+ ) v nezralém mozku se podílejí na vysoké excitabilitě, vzniku záchvatů, excitotoxicitě a zániku neuronů (Vizi et al., 2013). IEM1460 jako specifický antagonista AMPA receptorů prostupných pro Ca 2+ vykazuje antikonvulsivní působení v časných stadiích vývoje, přičemž účinek IEM1460 závisí na místu vzniku záchvatů, stadiu vývoje a expresi GluA2 podjednotky. Ro 25-6981 maleát (selektivní antagonista NMDA receptorů obsahujících podjednotku NR2B) vykazuje antikonvulsivní aktivitu v časném postnatálním období. Antikonvulsivní efekt u 12-denních potkanů trval minimálně 110 minut, to znamená, že tato látka by mohla být použitelná i klinicky. Vývojový profil exprese NR2B, NR2A a GluA2 podjednotek (tedy těch, které jsme ovlivňovali v elektrofyziologických pokusech) v různých částech mozku může přinést informace o potenciálních cílech věkově specifické antiepileptické a neuroprotektivni léčby. Stanovovali jsme změny v genech GRIN2A, GRIN2B a GRIA2A a proteinu... During early stages of postnatal development, glutamate receptors of NMDA and AMPA type, undergo intensive functional changes due to modifications of their subunit composition (Pachernegg et al., 2012: Paoletti et al., 2013). The NR2B-containing NMDARs (NR2B/NMDARs) and GluA2-lacking AMPARs (Ca 2+ -permeable) that are highly expressed in immature brain, are implicated in increased excitability, seizures generation, excitotoxicity, and neuronal death (Vizi et al., 2013). Pharmacological blockade of these types of receptors by their specific antagonists, can exhibit anticonvulsant effects at early stages of postnatal development. Therefore, we tested the influence of the IEM1460, a specific antagonist of Ca 2+ -permeable AMPARs and the Ro 25-6981 maleate, a highly selective and activity-dependent antagonist of NR2B/NMDARs on physiological excitability and epileptic phenomena induced in immature rats. Anticonvulsant action of IEM1460 was tested in two models of epileptic seizures: pentylenetetrazol (PTZ)- induced convulsions and cortical afterdischarges (ADs), induced in animals at P12, P18 and P25.
