Cíle: 1. Stanovení AGEs (Advanced Glycation End products) u pacientů s různými typy nefropatií. 2. Vztah AGEs s nutričními parametry a anémií. 3. Vliv renálních funkcí na hladiny sRAGE (soluble form of Receptor for Advanced Glycation End products). 4. Technika kultivace podocytů. 5. Stanovování podocytů v moči. Metodika: Fluoreskující AGEs jsme stanovovali spektrofluorimetricky, sRAGE nekompetitivní sendvičovou ELISA metodou. Podocyty jsme dále pasážovali a identifikovali imunocytochemicky. Podocyty v moči jsme stanovovali průtokovou cytometrií. Výsledky: 1. Nenalezli jsme signifikantní rozdíly v sérových hladinách AGEs mezi jednotlivými typy nefropatií, ač mají rozdílnou patogenezi. 2. Hladiny albuminu a prealbuminu korelují pozitivně a hladiny hemoglobinu negativně s hladinami AGEs u pacientů s CKD ve stádiu 1-5, bez bez nutnosti náhrady funkce ledvin. 3. Hladiny sRAGE se zvyšují u pacientů se sníženou renální funkcí nezávisle na příčině renálního onemocnění. 4. Osvojení postupů a technik kultivace podocytů. 5. Sledování citlivosti podocyturie ve vztahu k aktivitě onemocnění. Závěr: Potvrdili jsme, že hladiny AGEs závisí více na renální funkci než na typu nefropatie a jsou ovlivněny nutričními parametry a hemoglobinem. Jako první jsme prokázali, že koncentrace sRAGE úzce koreluje s renální... Aims: 1. Determination of AGEs (Advanced Glycation End products) in patients with various types of nephropathy. 2. Association AGEs with nutritional parameters and anemia. 3. Influence of renal parameters on sRAGE (soluble form of Receptor for Advanced Glycation End products) levels. 4. Technics and proceeding methods of the podocytes cultivation. 5. Determination of urine podocytes. Methods: We determined fluorescent AGEs by spectrofluorometry, sRAGE by Enzyme-Linked ImmunoSorbent Assay (ELISA). Podocytes were passaged and identified immunocytochemically. Podocytes in urine were specified by flow cytometry method. Results: 1. We did not find significant differences in AGEs serum levels among various types of nephropathy, even though the pathogenesis differs. 2. The albumin and prealbumin levels positively and haemoglobin levels negatively correlate with AGEs in patients with CKD grade 1-5, without necessity of dialysis. 3. Serum sRAGE levels are increased in patients with decreased renal function independently on the course of renal disease. 4. We implemented the methods and technics of podocyte cultivation. 5.
